A composite index, based on function analysis and including thirteen sub-indices, was developed to assess the overall quality of urban and urbanized beaches in the Mediterranean area. The aggregation of components and sub-indices was based on two questionnaires completed by beach users and experts. Applying the new Beach Quality Index (BQI) demonstrated that the quality of beaches could be improved. In general, the strongest aspects of the beaches assessed were those related to short-term user demand, and the weakest were those related to the consequences of human pressure on the area, in particular, erosion problems. The composite index is intended to be used together with Environmental Management Beach Systems (EMBs) as a hierarchical management scorecard and in monitoring programs. This new tool could also make planning more proactive by synthesizing the state of the most important beach processes.  相似文献

Foodborne illnesses associated with contaminated fresh produce are a common public health problem and there is an upward trend of outbreaks caused by enteric viruses, especially human noroviruses (HNoVs) and hepatitis A virus (HAV). This study aimed to assess the use of DNase and RNase coupled to qPCR and RT-qPCR, respectively, to detect intact particles of human adenoviruses (HAdVs), HNoV GI and GII and HAV in fresh produce. Different concentrations of DNase and RNase were tested to optimize the degradation of free DNA and RNA from inactivated HAdV and murine norovirus (MNV), respectively. Results indicated that 10 µg/ml of RNase was able to degrade more than 4 log10 (99.99%) of free RNA, and 1 U of DNase degraded the range of 0.84–2.5 log10 of free DNA depending on the fresh produce analysed. The treatment with nucleases coupled to (RT)-qPCR was applied to detect potential infectious virus in organic lettuce, green onions and strawberries collected in different seasons. As a result, no intact particles of HNoV GI and GII were detected in the 36 samples analysed, HAdV was found in one sample and HAV was present in 33.3% of the samples, without any reasonable distribution pattern among seasons. In conclusion, RT-qPCR preceded by RNase treatment of eluted samples from fresh produce is a good alternative to detect undamaged RNA viruses and therefore, potential infectious viruses. Moreover, this study provides data about the prevalence of enteric viruses in organic fresh produce from Brazil.  相似文献

A mid-lake sample is the conventional sampling strategy used in lake chemistry monitoring programs. Hence, addressing the question of how representative a mid-lake sample is of in-lake conditions is important for interpreting changes in long-term monitoring programs. Our hypotheses were that; (i) the location of a sampling site within a lake will affect its representativity and (ii) a mid-lake sample could be considered representative of average surface water physico-chemistry. We focused on the surface water chemistry in 34 Swedish lakes. To obtain a wide diversity of lake types, the lakes were classified according to mean depth, water residence time, and three main ecoregions. Two sampling seasons were selected, representative of high and low biological activity and stratified and non-stratified conditions, respectively. Ten samples were collected in each lake. The mid-lake samples differed significantly from the nine remaining samples in less than 4% of the total number of two-sample t-tests performed. Samples collected close to lake inlets differed from the other samples more often than samples collected in the central parts of the lakes. We concluded that the location of a sampling site within a lake may affect the sample's representativity of lake water chemistry, and that a mid-lake surface water sample could be considered representative of surface water chemistry.  相似文献

To abate the severe acidification of Swedish lakes are about 7% directly limed (AL-lakes). These are important to include when assessing acidification. Consequently, we need to estimate their chemical status as if they were not lime-affected. We hypothesize that there is a spatial dependence for calcium (Ca) and magnesium (Mg) concentrations. Spatial variation is determined using variograms. We study lakes within two 150 ∗ 150 km quadrates (EMEP150-grid) in the middle and southern parts of Sweden. We model Ca in AL-lakes using the Ca and Mg concentrations in nearby, unaffected lakes (UL-lakes). The mean Ca/Mg for the three or seven closest UL-lakes is used. We assume that Ca/Mg is constant for UL-lakes and that Mg unaltered by liming. For UL-lakes, Ca and Mg are spatially dependent. For AL-lakes, there is a spatial dependence for Mg, but, as anticipated, not for Ca. The modeled Ca for UL-lakes show a good correspondence with measured Ca; r 2 > 0.6; slopes close to 1 and intercepts almost 0. Using three or seven nearby lakes when modeling Ca give similar results. Modeled Ca was higher than measured Ca for 36% (three closest lakes) and 38% (seven closest) of the modeled AL-lakes in the middle part of Sweden. The corresponding results for the southern part were 10% and 9%, respectively. This model is an acceptable estimator of Ca in limed lakes and for estimating critical loads and acidification status on an EMEP-scale.  相似文献

Urban tinkering     
Cities are currently experiencing serious, multifaceted impacts from global environmental change, especially climate change, and the degree to which they will need to cope with and adapt to such challenges will continue to increase. A complex systems approach inspired by evolutionary theory can inform strategies for policies and interventions to deal with growing urban vulnerabilities. Such an approach would guide the design of new (and redesign of existing) urban structures, while promoting innovative integration of grey, green and blue infrastructure in service of environmental and health objectives. Moreover, it would contribute to more flexible, effective policies for urban management and the use of urban space. Four decades ago, in a seminal paper in Science, the French evolutionary biologist and philosopher Francois Jacob noted that evolution differs significantly in its characteristic modes of action from processes that are designed and engineered de novo (Jacob in Science 196(4295):1161–1166, 1977). He labeled the evolutionary process “tinkering”, recognizing its foundation in the modification and molding of existing traits and forms, with occasional dramatic shifts in function in the context of changing conditions. This contrasts greatly with conventional engineering and design approaches that apply tailor-made materials and tools to achieve well-defined functions that are specified a priori. We here propose that urban tinkering is the application of evolutionary thinking to urban design, engineering, ecological restoration, management and governance. We define urban tinkering as:
A mode of operation, encompassing policy, planning and management processes, that seeks to transform the use of existing and design of new urban systems in ways that diversify their functions, anticipate new uses and enhance adaptability, to better meet the social, economic and ecological needs of cities under conditions of deep uncertainty about the future.
This approach has the potential to substantially complement and augment conventional urban development, replacing predictability, linearity and monofunctional design with anticipation of uncertainty and non-linearity and design for multiple, potentially shifting functions. Urban tinkering can function by promoting a diversity of small-scale urban experiments that, in aggregate, lead to large-scale often playful innovative solutions to the problems of sustainable development. Moreover, the tinkering approach is naturally suited to exploring multi-functional uses and approaches (e.g., bricolage) for new and existing urban structures and policies through collaborative engagement and analysis. It is thus well worth exploring as a means of delivering co-benefits for environment and human health and wellbeing. Indeed, urban tinkering has close ties to systems approaches, which often are recognized as critical to sustainable development. We believe this concept can help forge much-closer, much-needed ties among engineers, architects, evolutionary ecologists, health specialists, and numerous other urban stakeholders in developing innovative, widely beneficial solutions for society and contribute to successful implementation of SDG11 and the New Urban Agenda.

Both Streptomyces species and mold species have previously been isolated from moisture-damaged building materials; however, an association between these two groups of microorganisms in indoor environments is not clear. In this study, we used a culture-independent method, PCR-denaturing gradient gel electrophoresis (PCR-DGGE), to investigate the composition of the Streptomyces community in house dust. Twenty-three dust samples each from two sets of homes categorized as high-mold and low-mold based on mold-specific quantitative PCR analysis were used in the study. Taxonomic identification of prominent bands was performed by cloning and sequencing. Associations between DGGE amplicon band intensities and home mold status were assessed using univariate analyses as well as multivariate recursive partitioning (decision trees) to test the predictive value of combinations of bands intensities. In the final classification tree, a combination of two bands was significantly associated with mold status of the home (p?=?0.001). The sequence corresponding to one of the bands in the final decision tree matched a group of Streptomyces species that included Streptomyces coelicolor and Streptomyces sampsonii, both of which have been isolated from moisture-damaged buildings previously. The closest match for the majority of sequences corresponding to a second band consisted of a group of Streptomyces species that included Streptomyces hygroscopicus, an important producer of antibiotics and immunosuppressors. Taken together, the study showed that DGGE can be a useful tool for identifying bacterial species that may be more prevalent in mold-damaged buildings.  相似文献

The aim of this study was to develop a method based on ultra high performance liquid chromatography coupled with mass spectrometry (UHPLC-MS) for lipid profiling in human placental choriocarcinoma (JEG-3) cells. Lipids were solid–liquid extracted from JEG-3 cells using a solution of chloroform/methanol (2:1, v/v) in a simple procedure requiring minimal sample alteration. Simultaneous separation of complex lipid mixtures in their major classes was achieved with a reversed-phase (C8) UHPLC column and a mobile phase containing methanol with 1 mM ammonium formate and 0.2 % formic acid (A)/water with 2 mM ammonium formate and 0.2 % formic acid (B). Lipids were characterized using time-of-flight (TOF) and Orbitrap under full scan and positive electrospray ionization mode with both analyzers. A total of 178 species of lipids, including 37 phosphatidylcholines (PC), 32 plasmalogen PC, 9 lyso PC, 4 lyso plasmalogen PC, 30 triacylglycerols, 22 diacylglycerols, 7 cholesterol esters, 25 phosphatidylethanolamines, and 12 sphingomyelins, were identified using TOF and Orbitrap. The identification of all lipid classes was based on exact mass characterization with an error < 5 ppm. The developed methodology was applied to study lipid alterations in human placental cells against the exposure to perfluorinated chemicals (PFCs) and tributyltin (TBT).  相似文献

In some fish species with paternal care, females prefer to spawn with males whose nests already contain eggs. Several hypotheses have been put forward to explain this behaviour, such as reduced risk of predation or cannibalism (the dilution effect), increased parental investment, and mate copying. This experimental study focuses on female mate choice in the sand goby, Pomatoschistus minutus. Females were found to choose males with eggs in their nests. In addition, hatching success increased with clutch size, mainly because males with larger clutches showed less filial cannibalism. Increased egg survival in large clutches may thus be explained by a combination of the dilution effect and higher parental investment. In another experiment, females did not seem to copy the observed mate choice of other females. In conclusion, female preference for males with eggs in their nests is adaptive, and can be explained by direct benefits, as more surviving offspring are produced.
