This study was designed to evaluate the ability of an in-situ bioassay to assess the impact of complex effluents on freshwaters and to identify toxic components. Reductions in the feeding rate of Gammarus pulex proved to be a sensitive indicator of the impact of metalliferous effluents on receiving water quality. The effluents contained a mixture of five potentially toxic metals. By combining information on feeding rates with bioaccumulation data, two metals, iron and manganese, were identified as the probable toxic agents. Laboratory experiments validated the conclusions reached from the field study and confirmed that iron was a major toxicant. The sensitivities of Gammarus pulex from a metal-contaminated site and a clean site were compared during both the field and laboratory studies. Interpopulation differences in the response of G. pulex to toxicants were detected in the field study but not in the laboratory experiments. Possible reasons for this are discussed.  相似文献

We designed a new gas exchange system that concurrently measures foliar H2O, O3, and CO2 flux (HOC flux system) while delivering known O3 concentrations. Stomatal responses of three species were tested: snapbean, and seedlings of California black oak (deciduous broadleaf) and blue oak (evergreen broadleaf). Acute O3 exposure (120-250 ppb over an hour) was applied under moderate light and low vapor pressure deficits during near steady state conditions. The rate of stomatal closure was measured when the whole plant was placed in the dark. An adjacent leaf on each plant was also concurrently measured in an O3-free cuvette. Under some conditions, direct measurements and calculated foliar O3 flux were within the same order of magnitude; however, endogenously low gs or O3 exposure-induced depression of gs resulted in an overestimation of calculated O3 fluxes compared with measured O3 fluxes. Sluggish stomata in response to light extinction with concurrent O3 exposure, and incomplete stomatal closure likewise underestimated measured O3 flux.  相似文献

Here, we identify and characterise cholinesterase (ChE) and carboxylesterase (CbE) activities in the body tissues of the sediment dwelling worm Arenicola marina. Exposure to the organophosphorus pesticide azamethiphos yielded an in vitro IC50 of 5 microg l(-1) for propionylcholinesterase (PChE). PChE was significantly inhibited in vivo after a 10 day exposure to 100 microg l(-1) azamethiphos, equivalent to the recommended aquatic application rate (ANOVA; F=2.75, P=0.033). To determine sensitivity to environmental conditions, A. marina were exposed for 10 days to field collected sediments. PChE activity was significantly lower in worms exposed to sediments from an estuary classified to be at high risk from point source pollution by the UK Environment Agency (ANOVA; F=15.33, P<0.001). Whilst causality cannot be directly attributed from these latter exposures, they provide an important illustration of the potential utility of esterase activity as a biomarker of environmental quality in this ecologically relevant sentinel species.  相似文献

The experiment has been designed to test the effect of geometric design characteristics and road environment on the subjective safety of rural road curves. The method used here allows to investigate the effect of a real driving environment on safety in a complex way. This paper examined the effects of work and family stressors and conflicts on Air Force women's mental health and functioning. We analyzed data from a 1993 survey of representative stratified samples of 525 Air Force women from the active duty reserve and guard forces. The analyses of the data are guided by the comprehensive model of work–family conflict that has been tested by Frone, Russell, and Cooper (1992) using a large representative community sample. Structural equation modeling analyses provided support for the work–family conflict model. The analyses also provided support for an extension of the model, which included the separate effects of marital and parental roles on mental health. The extended model demonstrated that job and parental stresses had direct effects on work–family conflicts and that job and marital distress and family–work conflict had an independent adverse effect on mental health. Whereas job and parental involvement had a beneficial effect on distress, they had an adverse effect on work–family conflicts. Copyright © 1999 John Wiley & Sons, Ltd.  相似文献

The current U.S. Environmental Protection Agency (U.S. EPA) protocols for mercury determinations in aqueous and solid waste samples (SW-846 Methods 7470 and 7471) using recirculating cold-vapor atomic absorption spectrometry (CV-AAS) have been evaluated. The U.S. EPA methods are not sufficiently flexible to permit special quality control (QC) measures, have limited detectability for low-level mercury concentrations, and are plagued by spectral interferences caused by the nonspecific absorption of primary mercury radiation by volatile organic vapors. The U.S. EPA protocols have been modified in a single-laboratory study to facilitate additional QC measures, to enhance detectability for low-level mercury concentrations, and to eliminate nonspecific vapor absorption interferences. Volumetric manipulations for additional QC measures, if required, are facilitated by performing the sample digestions in Erlenmeyer flasks rather than in the current Biochemical Oxygen Demand (BOD) reduction-aeration bottles. Typical manipulations for additional QC measures that are now feasible include dilution of concentrated samples and multiple aliquot sampling for post-digestion spike and replicate analyses. Instrument detectability is improved 10-fold by using a gas sparging bottle as a dedicated reduction-aeration vessel and a silver wool-amalgamation CV-AAS system operated in an open configuration. The on-line amalgamation/thermal desorption process of the modified CV-AAS system eliminates interfering water and organic matrix vapors prior to the mercury absorption measurement. Good accuracy and precision have been obtained with the amalgamation CV-AAS system for the analyses of four reference sediment materials. The amalgamation CV-AAS measurements on the reference sediment digests have been successfully performed at absolute mercury concentration levels that are only 1 to 4 times above the instrumental detection limit of the U.S. EPA recirculating CV-AAS method.  相似文献

The combined XAD-8 and XAD-4 resin procedure for the isolation of dissolved organic solutes from water was found to isolate 85% or more of the organic solutes from Lake Skjervatjern in Norway. Approximately 65% of the dissolved organic carbon (DOC) was first removed on XAD-8 resin, and then an additional 20% of the DOC was removed on XAD-4 resin. Approximately 15% of the DOC solutes (primarily hydrophilic neutrals) were not sorbed or concentrated by the procedure. Of the 65% of the solutes removed on XAD-8 resin, 40% were fulvic acids, 16% were humic acids, and 9% were hydrophobic neutrals. Approximately 20% of the hydrophilic solutes that pass through the XAD-8 resin were sorbed solutes on the second resin, XAD-4 (i.e., they were hydrophobic relative to the XAD-4 resin). The fraction sorbed on XAD-4 resin was called XAD-4 acids because it represented approximately 85–90% of the hydrophilic XAD-8 acid fraction according to the original XAD-8 fractionation procedure. The recovery of hydrophobic acids (fulvic acids and humic acids) and the hydrophobic neutral fraction from XAD-8 resin was essentially quantitative at 96%, 98%, and 86%, respectively. The recovery of XAD-4 acids from the XAD-4 resin was only about 50%. The exact reason for this moderately low recovery is unknown, but could result from π-π bonding between these organic solutes and the aromatic matrix of XAD-4. The hydrophobic/hydrophilic solute separation on XAD-8 resin for water from background Side A and Side B of the lake was almost identical at 65 and 67%, respectively. This result suggested that both sides of the lake are similar in organic chemical composition even though the DOC variation from side to side is 20%.  相似文献
