Prenatal diagnosis of the hyperornithinaemia, hyperammonaemia, and homocitrullinuria syndrome is described by the analysis of ornithine incorporation in second-trimester cultured amniotic fluid cells. An unaffected fetus was predicted and confirmed in the newborn child. This is the third reported prenatal diagnosis for this disorder and the second predicting an unaffected fetus.  相似文献

A general model of resource scarcity is presented from which previous models can be derived as special cases. Previous models of scarcity are compared and contrasted, refining the concept of scarcity and leading to a resolution of current disputes over the appropriate measure of scarcity. An update of the original work done by Barnett and Morse is summarized, leading to reconsideration of their original conclusions. A pilot study was conducted at Davies Reef on the central Great Barrier Reef between 20 November 1991 and 20 January 1992 to assess the suitability of settlement collectors for measuring larval settlement rates of echinoderms on coral reefs. The collectors were deployed for two months during summer when many echinoderm species are known to spawn. A total of 657 newly settled echinoderms were recovered from just 47 collectors, each having a volume of just 0.005 m3 but with a utilisable surface area of about 1.44 m2, indicating that competent-to-settle larvae were very abundant during the sampling period. Echinoids (7.8 sampler-1) and ophiuroids (5.6 sampler-1) were the most abundant groups on the collectors. Asteroids (0.2 sampler-1) and crinoids (0.3 sampler-1 were less common. The asteroids were all identified to species and included the crown-of-thorns starfish Acanthaster planci, Choriaster granulatus and Culcita novaeguineae. The newly settled echinoids could not be distinguished from each other but included Echinometra mathaei and Mespilia globulus. The abundance of each of the five different classes of echinoderms on the samplers was correlated with their abundance on the natural substratum. Significant spatial variability was found in settlement rates of echinoids over 1000s of metres and ophiuroids over 100s of metres, but not over smaller spatial scales. It is concluded that the collectors can provide a useful tool for monitoring spatial and temporal variability in settlement rates of echinoderms on coral reefs and for testing hypotheses about patterns of larval dispersal.  相似文献

Toxin content (fmol cell–1) and a suite of elemental and macromolecular variables were measured in batch cultures of the dinoflagellatesAlexandrium fundyense, A. tamarense andAlexandrium sp. from the southern New England region, USA. A different perspective was provided by semicontinuous cultures which revealed sustained, steady-state physiological adaptations by cells to N and P limitation. Two types of variability were investigated. In batch culture, changes in nutrient availability with time caused growth stage variability in toxin content, which often peaked in mid-exponential growth. A second type of variability that could be superimposed on growth stage differences is best exemplified by the high toxin content of cells grown at suboptimal temperatures. Calculations of the net rate of toxin production (R tox ; fmol cell–1 d–1) for these different culture treatments and modes made it possible to separate the dynamics of toxin production from cell division. Over a wide range of growth rates, cells produced toxin at rates approximating those needed to replace losses to daughter cells during division. The exception to this direct proportionality was with P limitation, which was associated with a dramatic increase in the rate of toxin production as cells stopped dividing due to nutrient limitation in batch culture. Growth stage variability in batch culture thus reflects small imbalances (generally within a factor of two) between the specific rates of toxin production and cell division. N limitation and CO2 depletion both affect pathways involved in toxin synthesis before those needed for cell division; P limitation does the opposite. The patterns of toxin accumulation were the same as for major cellular metabolites or elemental pools. The highest rates of toxin production appear to result from an increased availability of arginine (Arg) within the cell, due to either a lack of competition for this amino acid from pathways involved in cell division or to increased de novo synthesis. There were no significant changes in toxin content with either acclimated growth at elevated salinity, or with short term increases or decreases of salinity. These results demonstrate that toxin production is a complex process which, under some conditions, is closely coupled to growth rate; under other conditions, these processes are completely uncoupled. Explanations for the observed variability probably relate to pool sizes of important metabolites and to the differential response of key biochemical reactions to these pool sizes and to environmental conditions.  相似文献

Lysimeters located outdoors have been used to evaluate the decomposition of buried oily beach sand waste (OBS) prepared using Forties light crude oil and sand from different locations around the British coast. The OBS (5% oil by weight) was buried as a 12-cm layer over dune pasture sub-sand and overlain by 20 cm of dune pasture topsoil. Decomposition rates of oil residues averaged 2300 kg ha(-1) in the first year and the pattern of oil decomposition may be represented by a power curve. Oil decomposition was strongly related to the temperature in the OBS layer, but was also significantly affected by rainfall in the previous 12 h. The CO(2) flux at the surface of the treatment lysimeters followed the relationship [log(10) CO(2) (mg C m(-2) h(-1))=0.93+0.058x OBS temp. (degrees C)-0.042x12 h rain (mm)]. There was considerable variation in the rate of oil decomposition in sands collected from different sites. Sand from Askernish supported most microbial activity whilst sand from Tain was relatively inactive. The decomposition process appeared to cease when the sand became saturated with water, i.e. temporarily anaerobic. However, decomposition recommenced when the soil dried out. The fastest rate of decomposition occurred in sand from one of the two sites predicted to have high populations of hydrocarbon-degrading bacteria. Larger particle size and higher Ca content may also be significant factors governing the rate of decomposition.  相似文献

Blubber samples from grey seal (Halichoerus grypus) pups were collected in 1998 through to 2000. Twenty four newly weaned pups were sampled in November 1998, 13 of which were re-sampled during their first year of life, between March and December 1999, and an additional 48 and 25 samples (new cohorts) were obtained in 1999 (November-December) and 2000 (May-September) respectively. SimgaPBDE concentrations (sum of Di-HxBDEs) ranged from 45 to 1500 ng/g lipid, with a geometric mean of 290 ng/g lipid. BDE 47 dominated the congener profile, followed by BDEs 100, 99, 153 and 154. SimgaPCB concentrations (sum of 41 congeners) ranged from 100 to 93000 ng/g lipid, with a geometric mean of 4600 ng/g lipid, with CB congeners 153, 138 and 180 making up 84% of the total PCB concentrations. The predominant OC pesticides were p,p'-DDE (geometric mean=860 ng/g lipid, range=160-3800 ng/g lipid) and p,p'-DDT (geometric mean=120 ng/g lipid, range=30-670 ng/g lipid). No significant differences were found between males and females, probably due to the young age of the animals. Significant decreases (P<0.01 for PBDEs and P<0.05 for PCBs/OCs) were observed between newly weaned seals in 1998 and 1999, but not for the majority of juveniles in 1999 and 2000. Significant differences (P<0.01) were also found between the different seasons in 1999 newly weaned and juvenile seals, reflecting changes in blubber mass.  相似文献

This study was designed to: determine dibutyltin (DBT), tributyltin (TBT) and tetrabutyltin (TTBT) bi-weekly for a four month period (June-September 1986) in the Port Annapolis Marina, Mears Marina, Back Creek and Severn River area of Northern Chesapeake Bay; measure DBT, TBT and TTBT for five successive days (Thursday-Monday) to determine possible daily effects (weekday versus weekend) and determine DBT, TBT and TTBT every two hours for one full tidal cycle in the study area. Maximum concentrations of TBT were reported at both Port Annapolis Marina (1801 ng L–1) and Mears Marina (1171 ng L–1) during early June followed by significant reductions in TBT during late summer and early fall. All 4 Back Creek Stations also had highest concentrations of TBT in early June; significant reductions occurred during the next three months. The highest concentration of TBT reported in the Severn River (48 ng L–1) occurred in September. The lowest TBT value (5 ng L-1) at this station occurred in June. TTBT was not detected in any of the samples. The day of week sampled (Thursday-Monday) during the daily experiments was not found to significantly affect TBT concentrations. TBT evaluations every two hours during the tidal cycle demonstrated that values peaked at 1400 and 1600 hr time intervals. Peak concentrations of TBT occurred during a rising tide. The possible consequence of the measured TBT concentrations for aquatic biota are discussed.  相似文献

The effects of carbonate on uranium (VI) solubility under aerobic and cementitious conditions have been investigated. The information is of relevance to low level nuclear waste disposal. Aqueous NaOH, KOH, Ca(OH)2 and a cement leachate solution were added to uranyl nitrate solution. Afterwards, increasing amounts of ammonium carbonate were added to re-dissolve the precipitates. The precipitates were characterised by means of X-ray powder diffraction (XRPD) measurements and modelling studies. The model calculations were performed using the MINTEQA2 speciation code, with an expanded database incorporating uranium stability constants taken from the HATCHES database. The measured and predicted amounts of CO3(2-) needed to dissolve the precipitates were compared. The knowledge gained from the 'pure' systems was used to rationalise the precipitation and re-dissolution behaviour observed in the leachate system. The lack of uranium solubility at low carbonate levels brought into question literature formation constants for UO2(OH)3- and UO2(OH)4(2-). An approximate log K value of 26.8 for K2U2O7 formation was estimated from the KOH results at pH 12. Generally, uranium solubilities are expected to be insignificant at low level nuclear waste sites because anaerobic conditions should persist. However, this study has demonstrated that solubility in leachate could rise at high pH and high carbonate levels, if aerobic conditions were to develop. Soluble U(VI) species would be formed. The novel part of the study has been to reproduce the effects and to correlate enhanced solubilities with model predictions. Consequently, the study has re-emphasised the need for appropriate environmental monitoring of such sites.  相似文献
