A simple method is presented to evaluate microbial activity in aquatic sediments. The method is based on resazurin reduction by microbial electron transport chains and reduced chemical compounds present in the sample. The addition of m-cresol, which inhibits enzyme activity, allows one to measure microbial metabolism by difference. Small aliquots of sediment (about 1 g FW) are incubated at 20°C with resazurin solution, with and without m-cresol. The sample is then filtered and the unreduced resazurin is measured at 600 nm.

Testing the method with a bacterial suspension gave a resazurin reduction of 89 μg/h/109 cells. In a few marine coastal sediment samples, the resazurin reduction in aerobic conditions was in the range of 0.31 to 219.3 μg/h/g DW, which is equivalent to an oxygen consumption of 0.02 to 15.32 μg/h/g DW.  相似文献

Denitrification walls are constructed by mixing a carbon source such as sawdust into soils through which ground water passes. These systems can reduce nitrate inputs to receiving waters by enhancing denitrification. Maximum rates of nitrate removal by denitrification need to be determined for design purposes. To determine maximum rates of nitrate removal we added excess nitrate (50 mg N L(-1)) to a trench up-gradient of a denitrification wall during a 9-d trial. Bromide (100 g L(-1)) was also added as a conservative tracer. Movement of nitrate and bromide was measured from shallow wells and soil samples were removed for measurements of denitrification, carbon availability, nitrate, and other microbial parameters. Rates of nitrate removal, determined from the ratio of NO3-N to Br and ground water flow, averaged 1.4 g N m(-3) of wall d(-1) and were markedly greater than denitrification rates determined using the acetylene block technique (average: 0.11 g N m(-3) of wall d(-1)). These nitrate removal rates were generally lower than reported in other denitrification walls. Denitrification rates increased when nitrate was added to the laboratory incubations, indicating that despite large nitrate inputs in the field, denitrification remained limited by nitrate. This limitation was partially attributed to nitrate predominantly moving through zones of greater hydraulic conductivity or in the mobile fraction of the ground water and slow diffusion to the immobile fraction where denitrifiers were active.  相似文献

The nature of life on Earth provides a singular example of carbon-based, water-borne, photosynthesis-driven biology. Within our understanding of chemistry and the physical laws governing the universe, however, lies the possibility that alien life could be based on different chemistries, solvents, and energy sources from the one example provided by Terran biology. In this paper, we review some of these possibilities. Silanes may be used as functional analogs to carbon molecules in environments very different from Earth; solvents other than water may be compatible for life-supporting processes, especially in cold environments, and a variety of energy sources may be utilized, some of which have no Terran analog. We provide a detailed discussion of two possible habitats for alien life which are generally not considered as such: the lower cloud level of the Venusian atmosphere and Titan’s surface environment.  相似文献

Cadmium accumulation in crop plants, such as tobacco (Nicotiana tabacum L.), can lead to human exposure to this carcinogenic metal. To better define actual Cd distribution in cured or processed tobacco leaves from chief tobacco-producing regions, we analyzed 755 leaf samples of three major types (Flue-cured, Burley, and Oriental) obtained from 13 countries during 2001-2003. This survey may help identify regions with low- or high-Cd concentrations in tobacco to obtain insight into the cause of the concentration extremes and to assist in defining strategies to reduce Cd in tobacco. Cadmium concentrations in the samples ranged from 0 to 6.78 microg g(-1), as determined by ICP-MS. Significant differences were found among types and among countries, but significant interactions between type and country were found. Variations in Cd concentrations were also found in all countries. Our results suggest an important contribution of the field (e.g., bioavailable Cd in soil, other soil characteristics) to the Cd concentration in tobacco. Finally, the correlation between the concentration of Cd and that of other elements differed among the types, which could be effectively discriminated based on the concentrations of 20 elements.  相似文献

The availability of genomic data for an increasing number of species makes it possible to incorporate evolutionary processes into conservation plans. Recent studies show how genetic data can inform spatial conservation prioritization (SCP), but they focus on metrics of diversity and distinctness derived primarily from neutral genetic data sets. Identifying adaptive genetic markers can provide important information regarding the capacity for populations to adapt to environmental change. Yet, the effect of including metrics based on adaptive genomic data into SCP in comparison to more widely used neutral genetic metrics has not been explored. We used existing genomic data on a commercially exploited species, the giant California sea cucumber (Parastichopus californicus), to perform SCP for the coastal region of British Columbia (BC), Canada. Using a RAD-seq data set for 717 P. californicus individuals across 24 sampling locations, we identified putatively adaptive (i.e., candidate) single nucleotide polymorphisms (SNPs) based on genotype–environment associations with seafloor temperature. We calculated various metrics for both neutral and candidate SNPs and compared SCP outcomes with independent metrics and combinations of metrics. Priority areas varied depending on whether neutral or candidate SNPs were used and on the specific metric used. For example, targeting sites with a high frequency of warm-temperature-associated alleles to support persistence under future warming prioritized areas in the southern coastal region. In contrast, targeting sites with high expected heterozygosity at candidate loci to support persistence under future environmental uncertainty prioritized areas in the north. When combining metrics, all scenarios generated intermediate solutions, protecting sites that span latitudinal and thermal gradients. Our results demonstrate that distinguishing between neutral and adaptive markers can affect conservation solutions and emphasize the importance of defining objectives when choosing among various genomic metrics for SCP.  相似文献

Temporal variations in protein, carbohydrate, and lipid levels were studied in a passive suspension feeder, the gorgonian Paramuricea clavata. The samples were collected every month for mature and immature colonies over a three-year period (1997–2000). The relationship between biochemical composition and reproductive output was examined on the basis of the 1998 and 1999 data. In female and male P. clavata colonies, the tissue displayed differences in lipid concentrations only in winter–spring, due to the high lipid levels attained by female colonies in this period. Immature colonies showed significant differences in lipid concentration only with respect to mature females in spring. There were clear seasonal trends in the lipid and carbohydrate levels in P. clavata, with maximum values in winter–spring [male lipid 212±75 SD μg mg−1 of organic matter (OM) and female lipid 274±103 SD μg mg−1 of OM; male and female carbohydrate 68±14 SD μg mg−1 of OM], coinciding with maximum food concentration/quality, and minimum values in summer–autumn (male and female lipid 155±57 SD μg mg−1 of OM; male and female carbohydrate 56±14 SD μg mg−1 of OM), coinciding with low food concentration/quality. The relationship between reproductive output and tissue concentrations of lipids, carbohydrates, and proteins was not straightforward, although there was an evident overlap of the lipid accumulation and the gonadal development. The results of this study show that protein, carbohydrate, and lipid levels may provide a record of episodes in the ecological cycle bearing on the trophic aspects of the target species. The results indicate that information on seasonal biochemical levels may explain benthopelagic coupling processes, provided factors such as natural diet, feeding rates, reproduction, and growth are well understood.  相似文献

Marine reserves are an increasingly important tool for the management of marine ecosystems around the world. However, the effects of proposed marine reserve configurations on sustainability and yield of populations are typically not estimated because of the computational intensity of direct simulation and uncertainty in larval dispersal and density-dependent recruitment. Here we develop a method for efficiently assessing a marine reserve configuration for persistence and yield of a population with sedentary adults and dispersing larvae. The method extends the familiar sustainability criteria of individual replacement for single populations based on eggs-per-recruit (EPR) to spatially distributed populations with sedentary adults, a dispersing larval phase, and limited carrying capacity in the settlement-recruit relationship. We refer to this approach as dispersal-per-recruit (DPR). In some cases, a single DPR calculation, based on the assumption that post-settlement habitat is saturated (i.e., at maximum recruitment), is sufficient to determine population persistence, while in other cases further iterative calculations are required. These additional calculations reach an equilibrium more rapidly than a full simulation of age- or size-structured populations. From the DPR result, fishery yield can be computed from yield-per-recruit (YPR) at each point. We assess the utility of DPR calculations by applying them to single reserves, uniformly distributed systems of reserves, and randomly sized and spaced systems of reserves on a linear coastline. We find that for low levels of EPR in fished areas (e.g., 10% or less of the natural, unfished EPR when post-settlement habitats are saturated by 35% of natural settlement), a single DPR calculation is sufficient to determine persistence of the population. We also show that, in uniform systems of reserves with finite reserve size, maximal fisheries yield occurs when the density of reserves is such that all post-settlement habitat is nearly saturated with settlers. Finally, we demonstrate the application of this approach to a realistic proposed marine reserve configuration.  相似文献

Summary In 1981–1985, we studied territory dynamics in a southeastern Idaho sage sparrow population to evaluate the hypothesis that territory shifts represent adaptive adjustments in site use. We predicted that shifts should (1) result in changes in territory characteristics, (2) be influenced by previous reproductive success and result in greater success, and (3) decline in magnitude for individual males over time. Habitat features of territories changed little as a result of shifts, but territory size increased. Correspondingly, habitat features of territories were unrelated to reproductive success, whereas territory size was positively related. The magnitude of territory shifts was negatively correlated with preshift fledging success, and after shifts, males experienced greater average reproductive success than before. Successive territory shifts by individual males became progressively smaller. Thus, territory shifting by sage sparrows seems to be adaptive behavior aimed primarily at increasing territory size.  相似文献
