Nuclear bud (NB) formation was investigated in blood erythrocytes of 1892 flounder Platichthys flesus, herring Clupea harengus, and eelpout Zoarces viviparus specimens that were collected at 82 sites representing different regions of the Baltic Sea in 2009–2011. This is the first attempt to evaluate the baseline levels of NB and rank the genotoxicity risk for native fish species. NB levels were compared to the previously published micronuclei (MN) data from the same individual fish specimens in order to compare the two methods of genotoxicity assessment and investigate the relationship between MN as the cytogenetic measure of genotoxicity and the DNA damage reflecting NB. In 2009–2011, elevated NB levels in 89.4 % of flounder sampling groups indicated high and extremely high genotoxicity risk levels. Herring and eelpout sampling groups showed elevated levels of NB, 74.6 and 45.7 %, respectively. In general, herring and eelpout NB measure was more sensitive as the genotoxicity biomarker than MN. 相似文献

Bioassessment tools should distinguish between the effects of anthropogenic degradation in communities and natural temporal changes. The present study tests the influence of natural seasonal variability on macroinvertebrate stream communities assessed by a predictive model (PORTRIV) and a multimetric index (IPtI) calibrated for spring. The scores of PORTRIV decreased significantly between spring and autumn, and between spring and winter (ca. 37 to 53 %, respectively), while those of IPtI did not change significantly between seasons. For non-reference samples, the results of the predictive model also indicate no significant differences. A correction factor (CF) was calculated to adjust the existing differences in the model assessments between seasons, based on the percentage of variation of reference site scores from spring to autumn and winter. After the application of the CF to the OE50 scores of spring reference samples, the differences were no longer significant. Independent reference validation sites confirmed this tendency. This method has the advantage of avoiding large efforts required for the construction of databases from other seasons and the development of new models to allow the assessment of streams in seasons other than spring. Predation is often considered as one of the most important biotic factor determining the success of exotic species. The freshwater amphipod Gammarus roeseli has widely colonized Western Europe, where it is frequently found in sympatry with the native species (Gammarus pulex). Previous laboratory experiments revealed that G. roeseli may have an advantage over G. pulex through differential predation by native fish (brown trout). Morphological anti-predator defences (spines) were found responsible for lower rates of predation on the invasive G. roeseli. Here, using both field surveys and laboratory experiments, we tested if a differential of predation exists with other fish predators naturally encountered by gammarids. The main predators present in our field site were nocturnal benthic feeders (mainly bullheads, Cottus gobio). Fish diet analysis showed that, compared to its global availability in the river, G. roeseli was less consumed than G. pulex. In the field, however, G. roeseli was found mainly in the aquatic vegetation whereas G. pulex was found in all habitat types. Laboratory experiments in microcosms revealed that G. roeseli was less prone to predation by C. gobio only when vegetation was present. Depending on the type of predator, the differential of predation could therefore be mediated by antipredator behaviour, and a better usage of refuges, rather than by morphological defences. 相似文献

The most common mutation in alpha-1-antitrypsin deficiency, conversion of a G to an A at base 9989 (PI-Z), was detected with the chemical cleavage of mismatch method, demonstrating the power of the method for prenatal diagnosis. Exon V of the gene was amplified using the polymerase chain reaction and heteroduplexes were formed to test for the presence of the mutation. One hundred and fifty-one women of advanced maternal age who underwent genetic termination of pregnancy (TOP) were studied for their reproductive behaviour and the type of procedure for prenatal diagnosis in a subsequent pregnancy. A total of 59 women (39 per cent) had a further pregnancy. In all continuing pregnancies prenatal diagnosis was performed, of which 75 per cent consisted of chorionic villus sampling (CVS). Reproductive behaviour following a genetic termination was negatively correlated with maternal age and parity. Both reproductive behaviour and the choice to undergo a diagnostic procedure in the next pregnancy were independent of the type of diagnostic procedure in the previous affected pregnancy. 相似文献

Over 14 million hectares of erosion prone cropland in the United States has been converted into grasslands through the Conservation Reserve Program (CRP) administered by the United States Department of Agriculture, however, studies of the effects of CRP enrollment on plant communities and subsequent plant succession are largely lacking. In Delta Junction, Alaska plant communities in CRP fields are transitioning from grasslands to shrub dominated plant communities, which are resulting in compliance problems with program regulations that state “fields must be maintained in a condition that permits easy conversion to cropland”. To determine plant succession and how previous land management and soils might influence the transition, we measured plant populations in 20 CRP fields throughout Delta Junction using modified-Whittaker plots. These data were combined with data on current management practices, previous farming history, soils, soil properties, diversity indices, and time since land was cleared and analyzed with nonmetric multidimensional scaling ordination to determine factors that influence plant succession. Time in the CRP was the only factor consistently influencing plant succession. As time in the CRP increased, the planted introduced grasses brome grass (Bromus inermis) and red fescue (Festuca rubra) and the native pteridophyte (Equisetum arvense) decreased, whereas a native grass (Calamigrostis canadensis), five native forb, two native shrub, and three native tree species increased. Plant diversity increased at a rate of more than 2 species per 1000 m2 per year. Regression analyses of plant species and plant groups using time in the CRP as the dependent variable resulted in the identification of outlier CRP fields with significantly more or less than expected covers of vegetation. All fields with these outliers had reasonable explanations for the differences in cover that were unrelated to the overall rate of plant succession. Hepatitis A virus (HAV) is currently recognized as an important human food borne pathogen, and it is one of the most resistant
enteric RNA viruses, is highly infectious, and may lead to widespread outbreaks. The aim of this study was to optimize the
methods to detect HAV from artificially contaminated food. To this end, strawberry and lettuce were experimentally contaminated
with HAV suspension containing 6 × 106 copies/ml. After contamination, HAV persistence and washing procedure were evaluated at 0, 1, 3, 7, and 9 days of storage.
Five elution buffers (PBS (pH 7.4)/0.1% Tween80; 50 mM glycine/3% (wt/vol) beef extract (pH 9.5); PBS (pH 7, 4); 25 mM glycine/0.1
Tween80; and 1 M sodium bicarbonate) were used to elute the virus, and qualitative and quantitative PCR were used for HAV
detection. HAV was detected by qualitative and quantitative PCR using any of the five elution buffers, but PBS was the most
effective. Even after washing, HAV was detected up to 9 days after contamination by quantitative PCR. Quantitative PCR was
more sensitive than qualitative PCR since samples containing viral load lower than 1.4 × 103 copies/ml could not be detected by qualitative PCR. Quantitative PCR can be used for rapid detection of food borne viruses
