Environmental scanning electron microscopy (ESEM) with an energy-dispersive X-ray spectrometer (EDS) and time-of-flight secondary ion mass spectrometry (ToF-SIMS) were demonstrated to be useful analytical tools for investigating surface and bulk components of individual floc particles from both full- and bench-scale activated sludge systems. Detailed surface imaging of various hydrated biological floc particles by ESEM revealed substantial differences in surface features between treatment systems, while EDS identified spatial differences in the iron and the aluminum distributions. The ToF-SIMS spectra had signature fragments of protein and polysaccharide material from the floc surface, suggesting that this technique is capable of surface profiling extracellular polymeric substances. Principal-component analysis of the positive ion ToF-SIMS spectra from the mixed-liquor-suspended solid (MLSS) samples and reference aquatic organic materials found slight differences between the full- and bench-scale MLSS surface properties but substantial differences among MLSS and treated effluent from the same facility. 相似文献

Service quality in a construction technology laboratory can result in an improvement in construction companies’ competitiveness. This research aims to evaluate the service quality provided by a construction technology laboratory, specifically in the segment of performing testing through the rupturing of concrete samples. For this research, a survey was conducted using a structured questionnaire that was created based on a literature review regarding the surveying of laboratory customers at a university in the south of Brazil. Our research was both exploratory and quantitative, and the data were analyzed by applying multivariate data analysis. The conclusions of this study show that the dimensions of quality services are: communication, reliability, responsiveness, credibility, and tangibility. These five dimensions together provide a high explained variance standard of 79.75%. Having a suitable number of attendants available to serve customers and presenting agility in the construction laboratory is one of the managerial research contributions. Moreover, through information availability, this research generates greater traceability that can be used both by customers and by institutional management. Constantly and thoroughly monitoring the customer service cycle is another relevant factor that can improve the quality of the services provided. 相似文献

We present and test a conceptual and methodological approach for interdisciplinary sustainability assessments of water governance systems based on what we call the sustainability wheel. The approach combines transparent identification of sustainability principles, their regional contextualization through sub-principles (indicators), and the scoring of these indicators through deliberative dialogue within an interdisciplinary team of researchers, taking into account their various qualitative and quantitative research results. The approach was applied to a sustainability assessment of a complex water governance system in the Swiss Alps. We conclude that the applied approach is advantageous for structuring complex and heterogeneous knowledge, gaining a holistic and comprehensive perspective on water sustainability, and communicating this perspective to stakeholders. 相似文献

A series of investigations were conducted using sequencing batch biofilm reactor(SBBR) to explore the influence of C:N:P ratio on biological dephosphatation including the denitrifying dephosphatation and the denitrification process.Biomass in the reactor occurred mainly in the form of a biofilm attached to completely submerged disks.Acetic acid was used as the source of organic carbon.C:N:P ratios have had a significant effect on the profiles of phosphate release and phosphate uptake and nitrogen removal.The highest rates of phosphate release and phosphate uptake were recorded at the C:N:P ratio of 140:70:7.The C:N ratio of 2.5:1 ensured complete denitrification.The highest rate of denitrification was achieved at the C:N:P ratio of 140:35:7.The increase of nitrogen load caused an increase in phosphates removal until a ratio C:N:P of 140:140:7.Bacteria of the biofilm exposed to alternate conditions of mixing and aeration exhibited enhanced intracellular accumulation of polyphosphates.Also,the structure of the biofilm encouraged anaerobic–aerobic as well as anoxic–anaerobic and absolutely anaerobic conditions in a SBBR.These heterogeneous conditions in the presence of nitrates may be a significant factor determining the promotion of denitrifying polyphosphate accumulating organism(DNPAO) development. 相似文献

The honeybee has been the most important insect species for study of social behavior. The recently released draft genomic
sequence for the bee will accelerate honeybee behavioral genetics. Although we lack sufficient tools to manipulate this genome
easily, quantitative trait loci (QTLs) that influence natural variation in behavior have been identified and tested for their
effects on correlated behavioral traits. We review what is known about the genetics and physiology of two behavioral traits
in honeybees, foraging specialization (pollen versus nectar), and defensive behavior, and present evidence that map-based
cloning of genes is more feasible in the bee than in other metazoans. We also present bioinformatic analyses of candidate
genes within QTL confidence intervals (CIs). The high recombination rate of the bee made it possible to narrow the search
to regions containing only 17–61 predicted peptides for each QTL, although CIs covered large genetic distances. Knowledge
of correlated behavioral traits, comparative bioinformatics, and expression assays facilitated evaluation of candidate genes.
An overrepresentation of genes involved in ovarian development and insulin-like signaling components within pollen foraging
QTL regions suggests that an ancestral reproductive gene network was co-opted during the evolution of foraging specialization.
The major QTL influencing defensive/aggressive behavior contains orthologs of genes involved in central nervous system activity
and neurogenesis. Candidates at the other two defensive-behavior QTLs include modulators of sensory signaling (Am5HT

serotonin receptor, AmArr4 arrestin, and GABA-B-R1 receptor). These studies are the first step in linking natural variation in honeybee social behavior

Twenty-eight different tea samples sold in the United States were evaluated using high-performance liquid chromatography (HPLC) with fluorescence detection (FLD) for their contamination with polycyclic aromatic hydrocarbons (PAHs). Many PAHs exhibit carcinogenic, mutagenic, and teratogenic properties and have been related to several kinds of cancer in man and experimental animals. The presence of PAHs in environmental samples such as water, sediments, and particulate air has been extensively studied, but food samples have received little attention. Eighteen PAHs congeners were analyzed, with percentage recovery higher than 85%. Contamination expressed as the sum of the 18 analyzed PAHs was between 101 and 1337 μg/kg on dry mass and the average contents in all of the 28 examined samples was 300 μg/kg on dry mass. Seven of the congeners were found in all samples with wide ranges of concentrations as follows: fluorene (7–48 μg/kg), anthracene (1–31 μg/kg), pyrene (1–970 μg/kg), benzo(a)anthracene (1–18 μg/kg) chrysene (17–365 μg/kg), benzo(a)pyrene (1–29 μg/kg), and indeno(1,2,3-cd)pyrene (4–119 μg/kg). The two most toxic congeners benzo(a)pyrene and dibenzo(a,h)anthracene were found at high concentrations only in Earl Grey Twinnings, Earl Grey Harney& Sons Fine Teas, and Chai Ultra Spice Black Tea Twinnings. Six PAH congeners are considered as suspected carcinogens (U.S.EPA), formed the basis of the estimation of the toxic equivalent (TEQ), Chai Ultra-Spice Black Tea Twinnings had the highest TEQ (110.9) followed by two grey tea samples, Earl Grey Harney & Sons Fine Tea (57.7) and Earl Grey Twinnings (54.5). Individuals who are likely to realize the largest benefits from improvements in air quality often depend on other members of their households to make time or monetary contributions to their care. The presence of these dependency relationships among household members poses challenges for benefit estimation since it is unlikely that the conditions necessary for recovering the underlying individual preferences from household choices are satisfied in this setting. We propose a conceptual framework that highlights the role of these dependencies in the choice models used to estimate the willingness to pay for environmental quality improvements. We design a complementary stated preference survey that describes hypothetical dependency relationships for household members of different ages to test the implications of our conceptual model. Multivariate statistical techniques were applied to evaluate spatial/temporal variations, and to interpret water quality data set obtained at Alqueva reservoir (south of Portugal). The water quality was monitored at nine different sites, along the water line, over a period of 18 months (from January 2006 to May 2007) using 26 water quality parameters. The cluster analysis allowed the formation of five different similarity groups between sampling sites, reflecting differences on the water quality at different locations of the Alqueva reservoir system. The PCA/FA identified six varifactors, which were responsible for 64% of total variance in water quality data set. The principal parameters, which explained the variability of quality water, were total phosphorus, oxidability, iron, parameters that at high concentrations indicate pollution from anthropogenic sources, and herbicides indicative of an intensive agricultural activity. The spatial analysis showed that the water quality was worse in the north of the reservoir. 相似文献
