The formation of heteroduplexes from polymerase chain reaction (PCR) products has recently become a diagnostic tool that is routinely used for the prenatal detection of small deletions or insertions in a number of disease-causing alleles. We present evidence illustrating that heterozygous PCR products can manifest ‘invisible’ heteroduplexes that can ultimately lead to genotyping errors. Organic compounds derived from coal-tar wastes in a contaminated aquifer in St. Louis Park, Minnesota, were identified, and their partition coefficients between the tar phase and aqueous phase were determined and compared with the corresponding n-octanol/water partition coefficients. Coal tar contains numerous polycyclic aromatic compounds, many of which are suspected carcinogens or mutagens. Groundwater contamination by these toxic compounds may pose an environmental health hazard in nearby public water-supply wells. Fluid samples from this aquifer developed two phases upon settling: an upper aqueous phase, and a lower oily-tar phase. After separating the phases, polycyclic aromatic compounds in each phase were isolated using complexation with N-methyl-2-pyrrolidone and identified by fused-silica capillary gas chromatography/mass spectrometry. Thirty-one of the polycyclic aromatic compounds were chosen for further study from four different classes: 12 polycyclic aromatic hydrocarbons, 10 nitrogen heterocycles, 5 sulfur heterocycles, and 4 oxygen heterocycles. Within each compound class, the tar/water partition coefficients of these compounds were reasonably comparable with the respective n-octanol/water partition coefficient. 相似文献

Microbial populations were studied by plate counts and flow cytometry along the polishing sequence of a wastewater plant. The comparison between plate count and flow cytometry showed comparable trends, but plate counts detected less than 10% of the total intact bacteria counted by flow cytometry. Six months monitoring showed that, in spite of the high ozone doses, the disinfection effect of ozonation was insignificant. This is in agreement with the variability of ozone demand, not only related to chemical oxygen demand or total suspended solids. Membrane-intact bacterial population decreased after ozonation, slightly regrew between ozone and biological activated carbon (BAC), and sharply increased after BAC, probably for damaged bacteria regrowth and saprophytes release; BAC effluent had the highest bacterial counts. Preliminary investigations on the effluent microbial composition showed that the beta Proteobacteria subclass is the most represented in the BAC effluent, whereas the alpha subclass is the most sensitive to ozone effect. 相似文献

The invasion of Salvinia molesta in the Lower Senegal River Delta in Mauritania and Senegal in 1999 posed a serious threat to the socioeconomic conditions of the local people as well as to wetland biodiversity. Eventually, an effective biological control of S. molesta was obtained by means of the weevil Cyrtobagous salviniae, which was introduced in the river in Senegal and Mauritania in May 2000 and in Senegal in April 2001. In October 2001, it became apparent that the weevils were doing a magnificent job. The color of the plants was turning from green to dark-brown or black, and subsequently the plants started to sink to the bottom. Detailed monitoring of the dispersal of C. salviniae in November-December 2001 confirmed the visual observations of the outcome of biological control. In April 2002, it could be concluded that S. molesta was no longer a problem in the Senegal River. 相似文献

Determining human exposure to suspended particulate concentrations requires measurements that quantify different particle properties in microenvironments where people live, work, and play. Particle mass, size, and chemical composition are important exposure variables, and these are typically measured with time-integrated samples on filters that are later submitted to laboratory analyses. This requires substantial sample handling, quality assurance, and data reduction. Newer technologies are being developed that allow in-situ, time-resolved measurements for mass, carbon, sulfate, nitrate, particle size, and other variables. These are large measurement systems that are more suitable for fixed monitoring sites than for personal applications. Human exposure studies need to be designed to accomplish specific objectives rather than to serve too many purposes. Resources need to be divided among study design, field sampling, laboratory analysis, quality assurance, data management, and data analysis phases. Biogenic emissions of volatile organic compounds (VOCs) play a fundamental role in atmospheric chemistry. Vegetation is the most abundant natural source of VOCs, while terpenoids, as limonene, α and β pinene and mircene, top the plants emission list. Much interest has been demonstrated in oxidation and photooxidation reactions of VOCs, particularly of monoterpenoids, owing to their diversity and to uncertainties regarding their mechanism of reaction. Quantification of primary carbonylic compounds, as well as of biradical reaction components, is highly relevant to the understanding of the major reactions. In this context, taking into account both structural factors and the fact that these compounds are found in the essential oils of plants typically found in Brazil and that they may be present in the atmosphere from emission by the plants, the monoterpenoids (S)-(+)-carvone, (R)-(−)-carvone, (−)-carveol, geraniol and citral (a mixture of the isomers geranial and neral) were selected for this study.The ozonolysis reactions of the monoterpenoids were carried out under dark conditions for all experiments, due to their photochemical reactivity. The analysis of the results lets us propose a mechanism by which these reactions occur. The observed results of the ozonolysis of S and R carvone suggest that the stereochemistry of asymmetric carbon does not affect either in the yields of both formaldehyde and of OH radicals produced in the reaction, or in the reactivity of these compounds, for which the rate constants were in the scale of 10−6 s−1.We found that, in the (−)-carveol's cis and trans mixture, even though the hydroxyl in the axial position—in the case of trans-(C) and cis-(D′) isomers—favors the attack by the ozone molecule on the external double bond, thus increasing the mixture's reactivity , it affects the average production of formaldehyde. The presence of geraniol and citral led to the production of formaldehyde, propanone, glyoxal, methyl–glyoxal and cyclohexanone (OH radicals) as reaction products. The influence of an electron attractor group bonded to the carbon of the double bond, on the reactivity of the double bond, could not be observed in the case of citral, due to strong interference occurring in the instrument in all experiments with this monoterpenoid. Here, we show a fast and sensitive method for the determination of inorganic arsenic in natural waters using differential
pulse cathodic stripping voltammetry. All the arsenite determinations were done in 2.0 mol L−1 HCl + 3.15 × 10−4 mol L−1 Cu(II) supporting electrolyte. 1 × 10−3 mol L−1 sodium thiosulphate was used as As(V) reducing agent. The detection limit was 0.5 μg L−1 for both species. The method has been applied to water samples collected in an arsenic-contaminated region of Brazil, in

ABSTRACT: Protection of ground water quality is of considerable importance to local, state, and federal governments. This study uses a 15-year mathematical programming model to evaluate the effectiveness of low-input agriculture, under alternative policy scenarios, as a strategy to protect ground water quality in Richmond County, Virginia. The analysis considers eight policy alternatives: cost-sharing for green manures, two restrictions on atrazine applications levels, chemical taxation, a restriction on potential chemical and nitrogen levels in ground water only and in surface and ground water, and two types of land retirement programs. The CREAMS and GLEAMS models were used to estimate nitrate and chemical leaching from the crop root zone. The economic model evaluates production practices, policy constraints, and water quality given a long-term profit maximizing objective. The results indicate that low-input agriculture alone may not be an effective ground water protection strategy. The policy impacts include partial adoption of low-input practices, land retirement, and the substitution of chemicals. Only mandatory land retirement policies reduced all chemical and nutrient loadings of ground water; however, they did not promote the use of low-input agricultural practices. 相似文献

New decision support methods are needed to address complex multiple land use conflicts. Leaf-cutting ants of the generaAcromyrmex andAtta are considered the principal polyphagous pests of the Neotropics Although some members of these genera are of economic importance, have a broad geographic distribution, and are extremely good colonizers, others are endemic and closely interact with native ecosystems. Control is generally practiced against any colony, irrespective of its taxonomic status. Indiscriminate control coupled with habitat destruction threatens endemic species with extinction, and, through habitat simplification, favors other pest species.
