The effect of hydrostatic pressure on embryonic development of the calanoid copepod Calanus sinicus was studied. Differences of pressure effect among blastomere stages, 1-cell, 2-cell, 4-cell, 8-cell, 16-cell, blastula and limb-bud stage, were examined under two pressurizing conditions, abruptly (10 atm/min) and gradually (0.1 atm/min) increasing. Egg hatching success, deformity frequency and apoptotic cell degradation of hatched nauplii were examined. Egg hatching success rate was not significantly different between blastomere stages, and also between pressurizing conditions. Deformity frequencies of hatched nauplii were low in the early 1-cell and 2-cell stages, and high in the later blastula and limb-bud stages, in both abrupt and gradual pressurizing conditions. On the other hand, in regard to difference in pressurizing conditions, deformity frequency in gradual pressurizing was significantly higher than that in abrupt pressurizing rate. Gradual pressure increase seems to be more harmful to C. sinicus eggs than abrupt pressure change. Apoptosis induced in nauplii by hydrostatic pressure was detected for the first time in marine zooplankton. The embryos of C. sinicus are sensitive to pressure variations, that is, these embryos are supposed to sink to deeper waters, incurring greater risk of having deformities. In the field, C. sinicus ascend to the surface and spawn at night. By looking from this upward behavior, eggs are spawned at lower pressure and warmer temperature. Probably, the harmless low pressure and warm temperature lead eggs to hatch early and to recruit successfully.  相似文献

 The ability to introduce and express foreign genes can greatly facilitate in vitro studies of the molecular biology of an organism. We report here the first use of modified retroviral vectors to achieve foreign gene expression in primary cultured cells from the Oka organ and ovary of the blue shrimp Penaeus stylirostris (Litopenaeus) stylirostris (Holthuis 1980). Of the heterologous promoters tested, the Moloney murine leukemia virus and Rous sarcoma virus promoters mediated the highest levels of reporter gene expression. Firefly luciferase and Escherichia coliβ-galactosidase were successfully used as reporter genes in cultured cells from both Oka organ and ovary. The movement for global standardization is developing throughout all the economic areas of the EU. The aim of the Machine Directive that comes into effect in stages is to provide an international formula for a type of qualification in machine safety. EN292 is a nucleus for related standards, which are like a group of planets in a universe. It is a European standard as well as a guideline for implementation, showing practical safety requirements for machines. DFT calculations in gas and aqueous solution phases have been performed to study the mechanism of carbamate formation by the absorption of CO2 in 2-amino-2-methyl-1-propanol (AMP). The results reveal the importance of considering the effect of water as solvent for the reaction to proceed. Furthermore water molecules play an important role as a basic reactant leading to stable intermediates formation. These results point at a single-step, third order reaction as the most probable mechanism for the formation of carbamate by the absorption process.  相似文献

A serious forest decline of Betula ermanii Cham. has been observed at Mt. Mae-Shirane, Oku-Nikko, Japan, where high ozone (O3) concentration and severe water deficiency have been measured. In order to consider the possibility whether O3 and/or water stresses could have been the causes of the forest decline of B. ermanii, plant growth experiments were conducted in environment-controlled growth cabinets. Two-year-old seedlings of B. ermanii were exposed to either charcoal-filtered air (O3 concentration <5 ppb) or 50 ppb O3 (daily average, ranging between 20–100 ppb) for 123 days at 20.0/12.5 ± 1.0°C (day/night) and 70/80 ± 7% relative humidity (day/night). Simultaneously, seedlings were treated with three watering regime: 1.0 < pF < 1.8 (no water stress), 1.8 < pF < 2.5 (mild water stress) or 2.5 < pF < 3.0 (severe water stress). O3 exposure significantly reduced the dry weights of leaf, root and the whole plant, while water stress significantly reduced the dry weights of each organ and the whole plant. Significant reductions of net photosynthesis, transpiration and stomatal conductance were also observed under O3 and/or water deficiency treatments, while contents of RuBP carboxylase/oxygenase (Rubisco), chlorophyll a+b and some essential nutrient elements (N, P, K, Mg and Ca) were not markedly changed. It was suggested that the decrease in net photosynthetic rate induced mainly by stomatal closure was the major cause of the growth reduction under O3 and/or water stresses. No significant interactions between O3 and water stresses were observed in terms of the depression of dry matter production, which suggested that simultaneous stress treatments of O3 exposure and water deficiency could affect the tree growth of B. ermanii additively.  相似文献

In this study, we confirmed the utility of airborne and portable on-ground scanning light detection and ranging (LIDARs) for three-dimensional visualization of an urban park and quantification of biophysical variables of trees in the park. The digital canopy height model (DCHM) and digital terrain model generated from airborne scanning LIDAR data provided precise images of the ground surface and individual tree canopies. The heights of 166 coniferous and broadleaf trees of 11 species in the park were estimated from the DCHM images with slight underestimation (mean error = −0.14 m, RMSE = 0.30 m). Portable on-ground scanning LIDAR provided images of individual trees with detailed features. Tree height and trunk diameter were estimated to be within 0.31 m and 1 cm, respectively, from the on-ground LIDAR images. We combined airborne and on-ground LIDAR images to overcome blind regions and created a complete three-dimensional model of three standing trees. The model allowed not only visual assessment from all viewpoints but also quantitative estimation of canopy volume, trunk volume, and canopy cross-sectional area.  相似文献

Bivalve molluscan shellfish like clams and oysters, etc., are capable to bioaccumulate surrounding contaminants from waters into their digestive systems and posing serious threats of food poisoning. Detection of rotaviruses (RVs) in shellfish is of particular importance because RVs are prone to genome reassortment resulting in the emergence of new RV variants that may compromise vaccine safety. Herein, we have detected the wild-type RVs and Rotarix/RotaTeq vaccine strains in freshwater clams collected on the riverside, Kawasaki city, from July 2019 to January 2020 and correlated the detected genotypes with that of gastroenteritis cases of nearby clinics to understand the transmission of RVs in the environment. The wild-type RVs were detected in 62 (64.6%) out of 96 freshwater clams in every study month: July, September, November, and January that are considered as off-season for RV infections. The most frequent genotypes were G2 (42.9%), G8 (28.6%), G3 (14.3%), G1 (7.1%), and G10 (7.1%), which remained comparable with genotypic distribution found in the clinical samples over the last few years indicating that these RVs may accumulate in clams since a long time. However, G10 genotype was detected in clam but not in clinical samples suggesting the presence of asymptomatic infection or RVs could be carried out from a long distance. Importantly, vaccine strains, RotaTeq (1%) but not Rotarix (0%), were also detected in a clam. Attention must be paid to monitoring the potential transmission of wild-type and vaccine RV strains in the environment to prevent the emergence of new variants generated from genome reassortment with vaccine strains.

Airborne particulates have a major influence on the quality of indoor environments, which in turn affects human health. Both allergens and proteases are major etiological factors in allergic disease. Thus, the monitoring of environmental protease particulates is very important for the management of allergic disease. Protease-monitoring systems have been introduced in industry, but have not been applied to domestic settings, probably because of the high cost and many complex steps involved in these systems. In this study, we developed an indoor protease-monitoring system which can easily detect protease particulates without need for pre-treatment of dust samples or complex measuring instruments such as fluorescent plate reader. As a protease substrate, gelatin thin film (GTF) was adopted to evaluate small amounts of house dust particulates. The principle of this assay is based on the hydrolysis of GTF. Protease molecules spread from a particulate to GTF can hydrolyze the gelatin, thereby producing a spot on the GTF. This hydrolyzed spot visualized by staining was much larger than the particulate size, making visible to naked eye. The GTF method facilitated the observation of a subtle increase in protease particulates fallen on the GTF after the turnover of a used bed-quilt. The newly developed GTF method seems to be a suitable tool for protease monitoring at home.  相似文献
