Procedurally defined periphyton frequently includes substantial quantities of hydrous iron (Fe) and manganese (Mn) oxides. As these oxides are strong sorbers of heavy metals, their presence may complicate estimation of metal bioaccumulation by periphyton. We examined the relationship between nickel (Ni) sorption and the development time, biomass, and Fe and Mn oxide content of stream periphyton. Development time, the time during which periphyton accrued on submerged tile substrata, was used to provide variation in biomass, Fe and Mn levels. Stream periphyton from four development times was exposed to Ni for 2 h in the laboratory, and then ashed. Development time was significantly associated with ash-free dry mass (AFDM), Fe and Mn levels (ANOVA, P < or = 0.003). Ni extracted by a mild reductant (hydroxylamine hydrochloride) was significantly associated with development time, and with AFDM, Fe and Mn levels (linear models, P < or = 0.0002). A subsequent acid digestion yielded similar associations with the same variables (linear models, P < or = 0.0001). For both extractions, AFDM was significantly and positively correlated with Fe (r = 0.68 and 0.89) and with Mn (r = 0.77 and 0.93) (Spearman rank, P < or = 0.005). These data demonstrate the importance of periphyton development time in influencing both metal sorption and levels of biomass and ferromanganese oxides. The data also suggest that metal contaminant levels in periphyton should not be attributed automatically to biotic sorption. Periphyton metal-accumulation studies conducted where ferromanganese oxide concentrations are elevated should address the potential metal-sorbing roles of Fe and Mn oxides within the periphyton matrix.  相似文献

Detection of genetically modified (GM) traits in corn (Zea mays L.) is urgently needed for preservation of genetic identity and marketing GM products. A laboratory study was conducted to evaluate the efficiency, accuracy, and reliability of different analytical methods to detect GM traits in corn. Samples with known fractions of GM concentrations (Bacillus thuringiensis [Bt], Liberty Link [LL] and stacked [Bt/LL] genes) from commercial seed companies and those derived from yellow kernels in a white corn ear, outcrossed by pollen of neighboring Bt hybrid, were tested by lateral flow quick test kits and by enzyme-linked immunosorbent assay (ELISA)-based test strips purchased from different companies. Liberty Link hybrids are resistant to glufosinate (Liberty or Basta) herbicides, whereas Bt corn is developed for the control of European corn borer (Ostrinia nubilalis). Selected samples of GM concentrations were also tested in a commercial laboratory using DNA-based polymerase chain reaction (PCR) test. The results demonstrated that qualitative lateral flow quick tests could detect samples containing 1% or higher Bt and 2% or higher LL concentrations within the recommended time limit of the test. Faint test lines for samples containing 0.5 to 1% Bt or 1 to 2% LL concentrations appeared if samples remained in the test cup overnight. ELISA test strips detected the Bt content semiquantitatively in the range of 0.5 to 2.0%. Grain samples derived from non-Bt corn outcrossed by neighboring Bt pollen had usually lower GM concentrations than commercial GM seed samples. Both ELISA- and DNA-based PCR tests distinguished samples with GM concentrations between 0.1 to 0.5%, but the precision of quantification at this range was very low and results were highly inconsistent.  相似文献

The euryalinid snake star Astrobrachion constrictum (Farquhar), a normally deep-water species, exists within SCUBA diving range in New Zealand flords, and thus presents a unique opportunity for study. A population of A. constrictum in Doubtful Sound was investigated from February 1990 to July 1994 to determine the reproductive cycle. Unlike most ophiuroids, the gonads of A. constrictum are positioned along the aboral coelom in the proximal portion of the arms. The proximal 2 to 3 cm of the arms of individuals were thus removed on an approximately monthly basis for histological examination of the gonads. Image analysis was used to determine oocyte size-frequency distributions. A gonad index was calculated for each individual based on the ratio of cross-sectional area of the gonads to cross-sectional area of the arm. Maturity indices were calculated based on stage of development of the gonads of each individual. A. constrictum has a period of enhanced breeding activity in April–June, after which both the gonad and maturity indices decline. The increase in gametogenic activity during these months was correlated with rising sea temperature within the fiord. Although the females showed an annual pattern of gonad development, oogenesis was continuous throughout the year, with oocytes at different stages of development present in the gonads. Males also showed continuous spermatogenesis and, although the same annual pattern of gonad development was evident as seen in the females, it was less marked. The presence of females with large oocytes and ripe males throughout the study indicates that breeding in the Doubtful Sound population is prolonged, and possibly continuous, as found in many deep-sea ophiuroids.  相似文献

The effects of salinity stress on biomass yield, photosynthetic O2 evolution and nitrogenase activity were investigated using axenic cultures of Nodularia harveyana (Thwaites) Thuret originally isolated from a salt marsh at Gibraltar Point, Lincolnshire, UK in 1971 and studied in this laboratory in 1983. Biomass yields, as chlorophyll a per culture, were highest in the 0 to 100% seawater (0 to 35 sea salt) range with negligible growth in 200% seawater; growth on NH 4 + was greater than on N2 and NO 3 - , which did not differ significantly from each other. In short-term experiments, photosynthetic O2 evolution remained high at salinities up to 150% seawater (52.5 sea salt); nitrogenase activity remained high at salinities up to 100% seawater (35 sea salt). The major internal low molecular weight carbohydrate which accumulated in response to increased salinity was sucrose, the levels of which fluctuated markedly and rapidly in response to salinity change.  相似文献

Periodic wildfire maintains the integrity and species composition of many ecosystems, including the mediterranean-climate shrublands of California. However, human activities alter natural fire regimes, which can lead to cascading ecological effects. Increased human ignitions at the wildland-urban interface (WUI) have recently gained attention, but fire activity and risk are typically estimated using only biophysical variables. Our goal was to determine how humans influence fire in California and to examine whether this influence was linear, by relating contemporary (2000) and historic (1960-2000) fire data to both human and biophysical variables. Data for the human variables included fine-resolution maps of the WUI produced using housing density and land cover data. Interface WUI, where development abuts wildland vegetation, was differentiated from intermix WUI, where development intermingles with wildland vegetation. Additional explanatory variables included distance to WUI, population density, road density, vegetation type, and ecoregion. All data were summarized at the county level and analyzed using bivariate and multiple regression methods. We found highly significant relationships between humans and fire on the contemporary landscape, and our models explained fire frequency (R2 = 0.72) better than area burned (R2 = 0.50). Population density, intermix WUI, and distance to WUI explained the most variability in fire frequency, suggesting that the spatial pattern of development may be an important variable to consider when estimating fire risk. We found nonlinear effects such that fire frequency and area burned were highest at intermediate levels of human activity, but declined beyond certain thresholds. Human activities also explained change in fire frequency and area burned (1960-2000), but our models had greater explanatory power during the years 1960-1980, when there was more dramatic change in fire frequency. Understanding wildfire as a function of the spatial arrangement of ignitions and fuels on the landscape, in addition to nonlinear relationships, will be important to fire managers and conservation planners because fire risk may be related to specific levels of housing density that can be accounted for in land use planning. With more fires occurring in close proximity to human infrastructure, there may also be devastating ecological impacts if development continues to grow farther into wildland vegetation.  相似文献

The fate of the protease trypsin in intestines of individual herring larvae Clupea harengus L. was studied following digestion of the copepod Acartia tonsa. Trypsin was retained in the intestine during two consecutive pulses of feeding and defaecation of copepods. Quantification of herring trypsin in digested, defaecated copepods showed that ca. 1% of larval intestinal enzyme was defaecated along with 1 to 3 copepods. Following ingestion of a single meal, the level of intestinal trypsin post-ingestion declined to pre-ingestion levels within 1 to 2 d of starvation. All enzyme data thus indicated that trypsin, released in response to ingestion of a meal, was retained. In addition, analysis of fed subgroups of starved larvae clearly indicated that release of trypsin from the pancreas stopped after 6 to 8 d of starvation. As the fish still contained substantial amounts of trypsinogen, the underlying cause might be defective release mechanisms. Daily secretion of trypsin and processes responsible for enzyme retention in the gut are discussed. Assimilation efficiency in herring larvae was estimated for copepodite prey. The concentrations of lead, zinc, copper, manganese, cadmium and calcium have been measured in three peat bogs. The distribution of Pb, Zn, Mn and Ca are dependent on the position and fluctuations of the water table. They all become depleted in the waterlogged, anaerobic zone. Copper and cadmium are uniformly distributed and appear to be immobilised, probably by the formation of metal/organic complexes, and independent of the acid and redox conditions existing in the bogs. The use of the concentration profiles of the metal ions as a means of historical monitoring of trace metal contamination is complex for Pb, Zn, Mn and Ca but may be more straightforward for Cu and Cd.  相似文献

Fish faunas were sampled seasonally using a large and a small beam trawl in three seagrass habitats comprising predominantly Amphibolis griffithii or Posidonia sinuosa or Posidonia coriacea, which differ in seagrass and meadow structure. Amphibolis griffithii and P. sinuosa both produce a relatively dense leaf canopy, but the former exhibits a distinct architecture, with the leaf canopy overlying relatively open spaces surrounding woody stems, compared to the uniformly dense blade-like leaves of P. sinuosa which emerge directly from the sediment. In comparison, P. coriacea provides a landscape of patchy seagrass amongst areas of bare sand. Since the latter seagrass habitat contains large areas of sand, fish were also sampled in adjacent unvegetated areas. The number of species and density of fish were greater (P<0.05) in P. sinuosa than in either A. griffithii or P. coriacea. The mean number of species caught using the large trawl ranged from 16 to 24 in the first of these habitats compared to 14–21 and 9–15 in the last two habitats, respectively, and the mean densities ranged between 78 and 291 fish 1000 m?2 in P. sinuosa compared to 31–59 fish 1000 m?2 in A. griffithii and 31–59 fish 1000 m?2 in P. coriacea. The biomass of fish was greater (P<0.05) in both P. sinuosa and A. griffithii than in P. coriacea (4.2–5.3 kg and 3.3–6.2 kg versus 0.7–1.9 kg 1000 m?2, respectively). Furthermore, the size-structure of fish differed among these habitats, where the median weight of fish was 72.1 g in A. griffithii, compared to 7.5 g and 19.8 g in P. sinuosa and P. coriacea, respectively. Ordination and ANOSIM demonstrated that the species-composition differed markedly among the three seagrass habitats (P<0.001), suggesting that fish species display a distinct preference for particular seagrasses characterised by different architecture. Differences in species-composition among the seagrass habitats partly reflected the size-composition of fish in each habitat. The open space below the canopy of A. griffthii is likely to allow larger fish to occupy this habitat, whereas only small fish would be able to penetrate the dense foliage of P. sinuosa. Differences in species- and size-composition of fish among these habitats may be the result of settlement-sized larvae discriminating between particular seagrass and meadow structures, or fish being subject to different levels of predation and/or accessibility to food or space. The species-composition in P. coriacea was highly dispersed and did not differ from that of unvegetated areas. While several species were associated with both P. coriacea and bare-sand habitats, some species did display a high affinity with the seagrass P. coriacea. This may reflect an association with the sparse and narrower leaves of this seagrass or with the patchy occurrence of the seagrass Heterozostera tasmanica, which commonly occurs as an understorey in this habitat.  相似文献

Walrus (Odobenus rosmarus) blubber samples from Inukjuak and Akulivik (East Hudson Bay), Foxe Basin (Igloolik and Hall Beach) and Loks Land (East Baffin Island) were analysed for PCB congeners (ortho and non-ortho substituted) and other persistent organochlorines (DDT, toxaphene, chlordanes, dieldrin, mirex), as well as chlorinated dioxins/furans, to document spatial trends in contaminants in Canadian Arctic marine biota. Samples from 19 of 53 individuals had concentrations of SigmaPCBs greater than 1000 ng g(-1) (wet wt); the remaining individuals had much lower concentrations (50-600 ng g(-1)). Highest concentrations were found in samples from Inukjuak where average concentrations in blubber of females (N = 9) were 1450 +/- 954 ng g(-1) toxaphene, 2750 +/- 1780 ng g(-1) SigmaCHLOR, 2160 +/- 925 ng g(-1) SigmaDDT and 4790 +/- 2380 ng g(-1) SigmaPCB. SigmaPCB and SigmaDDT concentrations greater than 1000 ng g(-1) were unexpected based on previous studies of walrus from Greenland and Alaska. Local contamination was ruled out because levels of all organochlorines were elevated in each animal from Inukjuak, and elevated levels were also found in animals from Akulivik and Loks Land. Walrus from Inukjuak had sigma13C and sigma15N values in muscle intermediate between those of ringed seals (Phoca hispida) and those of walrus from Akulivik with low organochlorine levels. There was a weak but significant correlation between and sigma15N and (log)SigmaPCB. The Inukjuak walrus also had higher proportions of highly chlorinated PCB congeners, and higher DDE/SigmaDDT ratios than walrus from Igloolik or Akulivik. The results suggest that the walrus with elevated organochlorines are feeding at a higher trophic level than those with low levels and are probably utilizing ringed seals for a portion of their diet.  相似文献
