Adaptive sampling designs are recommended where, as is typical with freshwater mussels, the outcome of interest is rare and clustered. However, the performance of adaptive designs has not been investigated when outcomes are not only rare and clustered but also imperfectly detected. We address this combination of challenges using data simulated to mimic properties of freshwater mussels from a reach of the upper Mississippi River. Simulations were conducted under a range of sample sizes and detection probabilities. Under perfect detection, efficiency of the adaptive sampling design increased relative to the conventional design as sample size increased and as density decreased. Also, the probability of sampling occupied habitat was four times higher for adaptive than conventional sampling of the lowest density population examined. However, imperfect detection resulted in substantial biases in sample means and variances under both adaptive sampling and conventional designs. The efficiency of adaptive sampling declined with decreasing detectability. Also, the probability of encountering an occupied unit during adaptive sampling, relative to conventional sampling declined with decreasing detectability. Thus, the potential gains in the application of adaptive sampling to rare and clustered populations relative to conventional sampling are reduced when detection is imperfect. To assess the microbiological changes that occur during the maturation of overburden that has been disturbed by surface mining of coal, a surface mining-disturbed overburden unit in southeastern Ohio, USA was characterized. Overburden from the same unit that had been disturbed for 37 and 16 years were compared to undisturbed soil from the same region. Overburden and soil samples were collected as shallow subsurface cores from each subregion of the mined area (i.e., land 16 years and 37 years post-mining, and unmined land). Chemical and mineralogical characteristics of overburden samples were determined, as were microbial respiration rates. The composition of microbial communities associated with overburden and soil were determined using culture-independent, nucleic acid-based approaches. Chemical and mineralogical evaluation of overburden suggested that weathering of disturbed overburden gave rise to a setting with lower pH and more oxidized chemical constituents. Overburden-associated microbial biomass and respiration rates increased with time after overburden disturbance. Evaluation of 16S rRNA gene libraries that were produced by “next-generation” sequencing technology revealed that recently disturbed overburden contained an abundance of phylotypes attributable to sulfur-oxidizing Limnobacter spp., but with increasing time post-disturbance, overburden-associated microbial communities developed a structure similar to that of undisturbed soil, but retained characteristics of more recently disturbed overburden. Our results indicate that over time, the biogeochemical weathering of disturbed overburden leads to the development of geochemical conditions and microbial communities that approximate those of undisturbed soil, but that this transition is incomplete after 37 years of overburden maturation. 相似文献

This study investigated the effects of alkalinity on the anaerobic treatment of the organic solid wastes collected from the kitchen of Engineering Faculty in Dokuz Eylul University, Izmir, Turkey and the leachate characteristics treated in three simulated landfill anaerobic bioreactors. All of the reactors were operated with leachate recirculation. One reactor was operated without alkalinity addition. The second reactor was operated by the addition of 3 g l-1 d-1 of NaHCO3 alkalinity to the leachate and the third reactor was operated by the addition of 6 g l-1 d-1 NaHCO3 alkalinity to the leachate. After 65 d of anaerobic incubation, it was observed that the chemical oxygen demand (COD), volatile fatty acids (VFA) concentrations, and biochemical oxygen demand to chemical oxygen demand (BOD5/COD) ratios in the leachate samples produced from the alkalinity added reactors were lower than the control reactor while the pH values were higher than the control reactor. The COD values were measured as 18900, 3800 and 2900 mg l-1 while the VFA concentrations were 6900, 1400 and 1290 mg l-1, respectively, in the leachate samples of the control, and reactors containing 3 g l-1 NaHCO3 and 6 g l-1 NaHCO3 after 65 d of anaerobic incubation. The total nitrogen (TN), total phosphorus (TP) and ammonium nitrogen (NH4-N) concentrations in organic solid waste (OSW) significantly reduced in the reactor containing 6 g l-1 NaHCO3 by d 65. The values of pH were 6.54, 7.19 and 7.31, after 65 d of anaerobic incubation, respectively, in the aforementioned reactors results in neutral environmental conditions in alkalinity added reactors. Methane percentage of the control, reactors containing 3 g l-1 NaHCO3 and 6 g l-1 NaHCO3 were 37%, 64% and 65%, respectively, after 65 d of incubation. BOD5/COD ratios of 0.27 and 0.25 were achieved in the 3 and 6 g l-1 NaHCO3 containing reactors, indicating a better OSW stabilization. The consumption and production of food products in the municipality of Lisbon in the 1890–1900 decade is assessed with the support of historical cartography and statistical resources. For the first time, food production in a municipality in the turn to the twentieth century is accounted and simultaneously subject of a visual analysis of the land used for agriculture and of the water infrastructures that supported such uses. Agriculture occupied at least 40 % of the territory of the city, while the built environment occupied no more than 16 % of the territory. However, local production of food was far from supplying most of the citizens’ needs, and substantial food imports were needed. In this context, the municipality behaved like a heterotrophic system, highly dependent on the external supply of resources. Moreover, comparing to other European cities at the time Lisbon was facing in the end of the nineteenth century a late and slow transition from an agrarian social metabolism to an industrial one, suggesting that Lisbon was still relatively high-solar-powered as compared to other European cities at the time that were already highly fossil-fuel-powered. 相似文献

A nationwide programme for the prevention of sickle cell (SS and SC) disorders was initiated in Cuba in 1983. Couples at risk were identified by screening pregnant women and the partners of those who carry an abnormal haemoglobin, followed by genetic counselling and the offer of prenatal diagnosis. Prenatal diagnosis was performed in one laboratory, which had carried out 1068 prenatal tests for Hb SS and SC disorders by the end of 1992. We studied 101 couples to determine how far their reproductive behaviour was affected by the diagnosis of cystic fibrosis (CF) on its first occurrence in the couple's progeny and by the availability of prenatal diagnosis (PD). The couples were all resident in the Veneto and Trentino regions and attending the Verona CF Centre. CF had been diagnosed in the first affected child, during the period 1 January 1980-1 July 1990, before the age of 1 year. Couples received a questionnaire regarding socio-demographic status, reproduction data, and awareness of PD. Reproductive history was divided into three phases: prior to diagnosis of CF in the first affected child; from this time until PD was made available; and after the couples had learned of PD. In phase 2 (awareness of the genetic risk but not of PD), 54 couples showed a marked decrease in reproduction, none of the few pregnancies that occurred being taken to term. When couples became aware of PD, some resumption of reproductive activity occurred and 11 per cent of the 101 couples had another child; PD was used in 65 per cent of pregnancies and the abortion rate decreased to 35 per cent. This paper reports the results of first trimester prenatal diagnosis in a twin pregnancy at risk for homozygous β°−thalassaemia (β°−39 mutant). Trophoblast samples from both twins were obtained at 10 weeks gestation with a forceps guided by ultrasound. Trophoblast DNA analysis, carried out with the oligonucleotide technique, revealed that one fetus was homozygous and the other heterozygous for the β−39 mutant. This diagnosis was confirmed at 17 weeks gestation by amniocyte DNA analysis. DNA polymorphism analysis within the α-globin gene provided useful genetic markers for twin differentiation. 相似文献
