The nutrient distribution of the Cochin Backwater was studied at 6 stations throughout the year. The depth profiles of phosphorus (inorganic and organic), nitrogen (nitrate and nitrite) and silicon, showed a marked seasonal rhythm, induced by the local precipitation and land runoff. Changes in other environmental features such as temperature, salinity, dissolved oxygen, pH and alkalinity were small during the premonsoon period when the system remains marine-dominated, and large during the monsoon period when the estuary becomes freshwater-dominated. Seasonal changes in the hydrographical conditions of the adjoining coastal waters of the Arabian Sea which influence the backwater system have been discussed.  相似文献

In this paper we describe a cryopreservation protocol followed by the culture of Symbiodinium sp. isolated from the Caribbean gorgonian Pseudopterogorgia elisabethae as a potential renewable source of the dinoflagellate symbiont. Four different freezing protocols were designed: a controlled cooling device designed to cool at 1°C/min, a three-step protocol (−20°C for 2 h, −70°C for 2 h, liquid nitrogen-LN2), a two-step protocol (−70°C for 2 h, LN2), and a one-step protocol (LN2). All cells were stored in LN2 after cryopreservation. The cryoprotective agents (CPA) used were ethanol (EtOH) and methanol (MeOH) at 10 and 20%, and seawater (FSW) was used as a control. Viability measurements using cell counts showed that all cryopreservation protocols were relatively successful, and no trends were observed regarding freezing protocol or CPA used. After 19 weeks in culture the viability of samples which had high biomass was determined by the fluorescent assay CellTiter Blue™. The most viable cultures were those cryopreserved by a two-step protocol using 20% MeOH or 20% EtOH as a CPA. A genetic examination of the DNA of these samples using Symbiodinium-specific PCR primers confirmed that the composition of the culture had not changed. For the first time, we report that Symbiodinium sp. isolated from a gorgonian can be cryopreserved and subsequently cultured successfully. Lory Z. Santiago-Vázquez and Nealie C. In a previous study it was demonstrated that a lectin controls cell-cell interaction in the gorgonian Eunicella cavolinii (Koch) as a negative modulator. Now we describe the procedure to purify this lectin to homogeneity; its molecular weight is 23 400. The homologous proteoglycans were identified as positive modulators of cell-cell (and/or cell substrate) interaction. The purified single proteoglycan aggregates were 1200±700 nm long and the distance between the attachment points of the proteoglycan subunits was about 45 nm. The glycosaminoglycan residues of the gorgonian proteoglycans were identified as hyaluronic acid (35.5%), heparan sulfate (47.9%) and dermatan sulfate (14.1%). Binding studies with immobilized homologous proteoglycan preparations revealed that gorgonian cells attach to this substratum presumably via its glycosaminoglycans. Quantitative determinations of these two modulators of cell-cell recognition in the three regions of individual colonies revealed concentration gradients these concentration differences reflect a lower cell motility in the polyp-bearing branches and a higher motility of the cells in the polyp-free stem and in the basis, is discussed.  相似文献

In Dolioletta gegenbauri and Doliolum nationalis, collected in 1987 in the bay of Villefranche-sur-mer (French Mediterranean Sea), spermiogenesis is essentially the same. Early spermatids have a round head, a flagellum arising from a single centriole with short microtubules at 45° to its base, several mitochondria, and an acrosome 50 nm thick and 250 nm long with its long axis parallel to the plasma membrane. The acrosomal contents are dense, with a central denser plate. The nuclear envelope next to the acrosome is thickened and concave. In elongating nuclei, strands of chromatin become oriented parallel to the length of the nucleus and then twist helically. Although the mitochondria surround the nucleus, they remain relatively short and do not fuse into a single mitochondrion as in sperm of other tunicates. In very late doliolid spermatids, the acrosome undergoes exocytosis, and exposes fibrous material that stays associated with the tip of the sperm; no acrosomal tubule forms. Exocytosis at this stage may be triggered by fixation. If so, exocytosis probably occurs naturally at some time before fusion of sperm and egg. Sperm have elongate heads (1 m×10 m), with the anterior two-thirds of the nucleus surrounded by mitochondria. Spermiogenesis in doliolids, compared to that in other tunicates, is most like that in solitary members of the class Ascidiacea, except that in the latter the sperm mitochondria fuse and the acrosome appears incapable of exocytosis. In contrast, previous work has shown that salps (class Thaliacea) and colonial didemnid ascidians have an acrosomeless sperm with a spiral mitochondrion, while the class Appendicularia has a sperm with a midpiece, a compact head and an acrosome capable of exocytosis and acrosomal tubule formation. By outgroup comparison with echinoderms and acraniates, appendicularian sperm are plesiomorphic within the Tunicata. Thus, gamete morphology indicates that (1) solitary ascidians and doliolids had a common ancestor, (2) the popular idea that doliolids gave rise to appendicularians is incorrect, and (3) the Thaliacea are polyphyletic, doliolids having arisen very early from the ascidian lineage and salps having arisen later.  相似文献

The agricultural non-point source pollution by nitrogen (N) and phosphorus (P) loss from typical paddy soil (whitish soil, Bai Tu in Chinese) in the Taihu Lake region was investigated through a case study. Results shown that the net load of nutrients from white soil is 34.1 kg ha–1 for total nitrogen (TN), distributed as 19.4 kg ha–1, in the rice season and 14.7 kg ha–1in the wheat season, and for total phosphorus (TP) 1.75 kg ha–1, distributed as 1.16 kg ha–1 in the rice season and 0.58 kg ha–1 in the wheat season. The major chemical species of N loss is different in the two seasons. NH4-N is main the form in the rice season (53% of TN). NO3-N is the main form in wheat season (46% of TN). Particle-P is the main form in both seasons, (about 56% of TP). The nutrient loss varied with time of the year. The main loss of nutrients happened in the 10 days after planting, 64% of TN and 42% of TP loss, respectively. Rainfall and fertilizer application are the key factors which influence nitrogen and phosphorus loss from arable land, especially rainfall events shortly after fertilizer application. So it is very important to improve the field management of the nutrients and water during the early days of planting.  相似文献

A short outline is given of the floristic composition, structure and distribution of coastal dune vegetation found at Malindi Bay, Kenya. The area was studied by air photo interpretation and field sampling to determine the relationship of plants to aeolian features. TWINSPAN classification was used to distinguish geomorphological units on the basis of their species composition. In this paper, an inventory and first quantitative analysis of vegetation distribution is presented. We identified 174 plant species from 62 families in the sand dunes and several plant communities are distinguished based on the species content and the connection with morphological units.Papilionaceae with 18 species andPoaceae with 17 species were the most represented families. A distinct zonal distribution of the plant communities was found. The most important plant species are the pioneer vegetation consisting ofHalopyrum mucronatum, Ipomoea pescaprae andScaevola plumieri. The woody shrub species which have colonized the established primary dunes and hummock dunes areCordia somaliensis, Pluchea discoridis, Tephrosia purpurea (dunensis). Succulent herbs were identified in the dune slacks and salt marsh that are moist and damp environments.  相似文献

The role of hydroxyl radicals on the degradation of lignins during a cellulosic pulp bleaching process including a photocatalytic stage, was assessed using peroxyformic acid lignins EL1 and REL1 and two phenolic niphenyl lignin models 1 and 2. The irradiations were performed in the absence of photocatalyst TiO2 and H2O2 (condition a), in the presence of TiO2 (condition b) and in the presence of H2O2 (condition c). The experiments were conducted in alkaline (pH approximately 11) aqueous ethanol solutions with oxygen bubbling. The relative phenolic content of the irradiated solutions, which is indicative of the involvement of hydroxyl radicals, was determined by ionization absorption spectroscopy. The results obtained show that the catalyzed reaction involves both degradation of the phenolate groups by electron transfer and hydroxylation of the lignin aromatic structure. Benzyl alcohol structural elements in sodium borohydride reduced lignin REL1 and compound 2 were also found as good trapping agents for the hydroxyl radicals. The degradation of EL1 was studied by measuring its fluorescence emission by comparison to the fluorescence of compound 2. The emission spectra indicate that some biphenyl phenolate anions in EL1 are reacting under UV/visible irradiation and some others, probably polyphenolic chromophores emitting less fluorescence, are formed.  相似文献

A cheap and simple but very sensitive and specific detection tube has been developed for the detection of cholinesterase-inhibiting compounds in natural and waste water disinfected with chlorine-containing oxidants. After the oxidants have been quantitatively destroyed by a mixture of NaNO(2) and NaHCO(3), the horse-serum cholinesterase catalyses the hydrolysis of the orange-red 2,6-dichloro-indophenyl acetate producing the blue 2,6-dichloroindophenol. The colour transformation does not occur when the enzyme is inactivated by the cholinesterase-inhibiting compounds. By making use of a chemical heater the detection tube can complete a check job at an environmental temperature as low as about 0 degrees C in only 5 min. The plastic-made detection tube is composed of an inner tube for use in chemical reaction and an outer tube for use in chemical heating. DDVP can be detected at concentrations as low as 2 microg ml(-1), when the sampled water volume is 0.5 ml.  相似文献
