Study of metabolites from the degradation of polycyclic aromatic hydrocarbons (PAHs) by bacterial consortium enriched from mangrove sediments 总被引:3,自引:0,他引:3
The PAH metabolites produced during degradation of fluorene, phenanthrene and pyrene by a bacterial consortium enriched from mangrove sediments were analyzed using the on-fiber silylation solid-phase microextraction (SPME) combining with gas chromatography–mass spectrometry (GC–MS) method. Seventeen metabolites at trace levels were identified in different PAH degradation cultures based on the full scan mass spectra. In fluorene degradation cultures, 1-, 2-, 3- and 9-hydroxyfluorene, fluorenone, and phthalic acid were detected. In phenanthrene and pyrene degradation cultures, various common metabolites such as phenanthrene and pyrene dihydrodiols, mono-hydroxy phenanthrene, dihydroxy pyrene, lactone and 4-hydroxyphenanthrene, methyl ester, and phthalic acid were found. The detection of various common and novel metabolites demonstrates that SPME combining with GC–MS is a quick and convenient method for identification as well as monitoring the real time changes of metabolite concentrations throughout the degradation processes. The knowledge of PAH metabolic pathways and kinetics within indigenous bacterial consortium enriched from mangrove sediments contributes to enhance the bioremediation efficiency of PAH in real environment. 相似文献

Leaching of atrazine, metolachlor and diuron in the field in relation to their injection depth into a silt loam soil 总被引:1,自引:0,他引:1
A field experiment was conducted on a Calcaric Cambisol soil to study the consequences of the penetration depth and properties of pesticides on the risk of subsequent leaching. Three pesticides with different mobility characteristics and bromide were injected at 30 cm (where soil organic matter (OM) was 2%) and 80 cm (soil OM 0.5%) on irrigated plots without a crop. The migration of injected solutes was assessed for two years by sampling the soil solution using six porous cups installed at 50 and 150 cm depth and by relating solute contents to drainage water flux estimated by the STICS model (Simulateur mulTIdisciplinaire pour les Cultures Standard). Pesticides injected at 30 cm were strongly retained so that no metolachlor or diuron was detected at 50 and 150 cm. The ratio of atrazine peak concentration in the soil solution to concentration in the injected solution (C/C0) was 1 × 10−3 and 0.2 × 10−3, respectively, at 50 and 150 cm. When injected at 80 cm, (C/C0) of atrazine, metolachlor and diuron were 10 × 10−3, 1 × 10−3 and 0.3 × 10−3 at 150 cm, respectively; 1/(C/C0) was correlated with Koc values reported from databases. The ratio of drainage volume to the amount of water at field capacity in the soil layer between the injection point at 30 cm and the water sampling level (V/V0) at 50 and 150 cm was 0.6 and 0.9, respectively, for bromide and 1.6 and 1.0 for atrazine. V/V0 of the injected solutes at 80 cm was for bromide, atrazine, metolachlor and diuron 0.6, 0.9, 1.2 and 1.7, respectively; pesticide V/V0 was correlated with Koc. The retardation factor was a good indicator of migration risk, but tended to overestimate retardation of molecules with high Koc. Atrazine desorption represented an additional leaching risk as a source of prolonged low contamination. The large variability in soil solution of bromide and pesticide concentrations in the horizontal plane was attributed to flow paths and clods in the tilled soil layer. This heterogeneity was assumed to channel water fluxes into restricted areas and thereby increase the risk of groundwater contamination. HCHs and DDTs in sediment-dwelling animals including mollusks and crabs from the Yangtze Estuary were determined by GC-ECD. Levels of t-HCH were in the range of 1.2-5.5 ng g(-1) and averaged 3.5 ng g(-1) in mollusks, while t-DDT concentrations ranged from 26.0 to 68.8 ng g(-1), with a mean of 34.5 ng g(-1). In crabs t-HCH concentrations varied from 2.0 to 25.7 ng g(-1) and averaged 13.8 ng g(-1), whereas the concentrations of t-DDT were in the range of 1.5-24.8 ng g(-1) with a mean value of 5.9 ng g(-1). The HCHs and DDTs levels depend on geographical position and sources, showing the high levels at fresh water area in the estuary, such as XP, CM and LHK sites, and lower at brackish water area, such as FX site, and little difference between species. Results also indicate there was no significant relationship between t-HCH (t-DDT) concentrations and lipid contents both in mollusks and crabs because of non-equilibrium state under a specific estuarine dynamics; smaller individuals accumulated more HCHs and DDTs than larger individuals of mollusks at LHK site, showing different uptake rate for these pesticides; moreover, HCHs and DDTs levels were lower in female crab bodies than male crab bodies suggesting that the release of spawning. BSAFs (Biota- Sediment Accumulation Factors) from sediment-dwelling animals for HCHs and DDTs show a significant "one high with two low" and "one low with two high" effect in the Yangtze Estuary. 相似文献

The contribution of small watersheds to coastal pollution in Mediterranean areas is still poorly known, and presents great variations along an hydrological year. This work deals with the characterization and quantification of Al, Fe, Zn, Cu, Pb, Si, NO(3)(-) and organic carbon transported from the continent to the sea by a small coastal river in the French Mediterranean area. Transported species were characterized both during rainy and dry periods. During non-rainy periods (base-flow), waters showed a low content of contaminants, whereas during heavy rain events, following dry periods, some metals and organic carbon reached concentrations that could affect biological populations. These contaminants were mainly found in the particulate fraction, originating from the runoff of surface waters, which represents the main process of pollution in urban areas. 相似文献

The risks to Indo-Pacific Humpback Dolphins and Finless Porpoises associated with polychlorinated biphenyls (PCBs) were assessed. Stomach contents from twelve stranded Humpback Dolphins and sixteen stranded Finless Porpoises were collected. Concentrations of total and isomer-specific PCBs in the stomach contents were determined using dual-column gas chromatography equipped with electron capture detectors (GC-ECD). Risks due to the PCBs were assessed in three scenarios, based on total PCBs (summation of 41 PCB congeners), total toxicity equivalency (TEQs) and PCB 118, using the toxicity reference values (TRVs) as the threshold effects benchmarks. The calculated risk quotients (RQs) showed that risks due to PCBs were generally low or negligible. Specifically, RQs from total TEQs and total PCBs for Finless Porpoises are below one, suggesting that PCBs should be a low risk for the Finless Porpoise in Hong Kong waters. However, the Humpback Dolphin has RQs larger than 1 for total TEQs and total PCBs when the 95th percentile data were used in the evaluation. This indicates that further investigation may be needed to examine more closely the potential impact of toxic contaminants in the habitat of the Humpback Dolphin. 相似文献

Degradation of di-butyl-phthalate by soil bacteria 总被引:2,自引:0,他引:2
Twelve Gram-positive phthalate ester degraders were isolated from soil. Using Biolog GP2 plates, eight of them were identified as belonging to the Corynebacterium-Mycobacterium-Nocardia group, while the remaining four were unidentifiable. When cultured in the presence of di-butyl-phthalate (DBP) in basal salts solution, five of these isolates accomplished more than 90% of DBP degradation within 48 h (fast group), three were placed in the medium group, and the remaining four were placed in the slow group which caused less than 30% of DBP degradation within the same period of time. A 420 bp DNA fragment was amplified from six isolates and none of them fell within the slow group. When compared with the large subunit of phthalate dioxygenase gene (phtA) of Arthrobacter keyseri, 83% and 91% similarities were evident in the nucleotide and amino acid sequences, respectively. However, no correlation between cell surface hydrophobicity and phthalate degradation ability was evident. Six surfactants (Brij 30, Brij 35, Tergitoltype NP-10, Triton N-101, Triton X-100 and SDS) were tested for their abilities to increase degradation rate. When added at the critical micellar concentration (CMC), they all displayed strong growth inhibition against the three bacteria tested, with Brij 30 been the least toxic to isolates G2 and G11, and Brij 35 had the least inhibitory effect for G1. When half the CMC of Brij 30 was incorporated into the basal salts, the inhibitory effect on DBP degradation remained. Soil helped to minimize surfactant toxicity of surfactant and increase the degradation potential of some of the test bacteria. When DBP-amended soil had been aged for three months, decreases in bioavailability were observed but the effect varied tremendously between different organisms. For isolates G1, G2, G5, G7 and G17 the aging effects were almost non-exist. The present study indicates that selection of a suitable degrader may minimize the undesired effect of aging on bioremediation process. 相似文献

Application of a constructed wetland for industrial wastewater treatment: a pilot-scale study 总被引:9,自引:0,他引:9
The main objective of this study was to examine the efficacy and capacity of using constructed wetlands on industrial pollutant removal. Four parallel pilot-scale modified free water surface (FWS) constructed wetland systems [dimension for each system: 4-m (L)x1-m (W)x1-m (D)] were installed inside an industrial park for conducting the proposed treatability study. The averaged influent contains approximately 170 mg l(-1) chemical oxygen demand (COD), 80 mg l(-1) biochemical oxygen demand (BOD), 90 mg l(-1) suspend solid (SS), and 32 mg l(-1) NH(3)-N. In the plant-selection study, four different wetland plant species including floating plants [Pistia stratiotes L. (P. stratiotes) and Ipomoea aquatica (I. aquatica)] and emergent plants [Phragmites communis L. (P. communis) and Typha orientalis Presl. (T. orientalis)] were evaluated. Results show that only the emergent plant (P. communis) could survive and reproduce with a continuous feed of 0.4m(3)d(-1) of the raw wastewater. Thus, P. communis was used in the subsequent treatment study. Two different control parameters including hydraulic retention time (HRT) (3, 5, and 7d) and media [vesicles ceramic bioballs and small gravels, 1cm in diameter] were examined in the treatment study. Results indicate that the system with a 5-d HRT (feed rate of 0.4m(3)d(-1)) and vesicles ceramic bioballs as the media had the acceptable and optimal pollutant removal efficiency. If operated under conditions of the above parameters, the pilot-plant wetland system can achieve removal of 61% COD, 89% BOD, 81% SS, 35% TP, and 56% NH(3)-N. Contamination source identification is a crucial step in environmental remediation. The exact contaminant source locations and release histories are often unknown due to lack of records and therefore must be identified through inversion. Coupled source location and release history identification is a complex nonlinear optimization problem. Existing strategies for contaminant source identification have important practical limitations. In many studies, analytical solutions for point sources are used; the problem is often formulated and solved via nonlinear optimization; and model uncertainty is seldom considered. In practice, model uncertainty can be significant because of the uncertainty in model structure and parameters, and the error in numerical solutions. An inaccurate model can lead to erroneous inversion of contaminant sources. In this work, a constrained robust least squares (CRLS) estimator is combined with a branch-and-bound global optimization solver for iteratively identifying source release histories and source locations. CRLS is used for source release history recovery and the global optimization solver is used for location search. CRLS is a robust estimator that was developed to incorporate directly a modeler's prior knowledge of model uncertainty and measurement error. The robustness of CRLS is essential for systems that are ill-conditioned. Because of this decoupling, the total solution time can be reduced significantly. Our numerical experiments show that the combination of CRLS with the global optimization solver achieved better performance than the combination of a non-robust estimator, i.e., the nonnegative least squares (NNLS) method, with the same solver. 相似文献

OBJECTIVES: Effective July 1, 2000, Florida's universal helmet law was amended to exclude riders ages 21 and older with insurance coverage providing at least 10,000 US dollars in medical benefits for injuries sustained in a motorcycle crash. Observed helmet use in Florida was reported to have declined from nearly 100% in 1998, before the law change, to 53% after. This study examined the effects of the law change on the likelihood of death, given involvement in a motorcycle crash. METHODS: Rates of motorcyclist deaths per crash involvement in Florida for 2001-2002 (after the law change) were compared with those for 1998-1999 (before the law change). Before/after death rate ratios (95% CIs) were examined, and logistic regression models estimated the effect of the helmet law change on the odds of death in a crash, while controlling for rider gender, age, and seating position, and number of vehicles. RESULTS: The motorcyclist death rate increased significantly after the law change, from 30.8 to 38.8 deaths per 1,000 crash involvements. Motorcyclist death rates increased for single- and multiple-vehicle crashes, for male and female operators, and for riders of all ages including those younger than 21. After controlling for gender and age, the likelihood of death given involvement in a motorcycle crash was 25% higher than expected after the law change. It is estimated that 117 motorcyclist deaths could have been avoided during 2001-2002 if Florida's universal helmet law had remained in place. CONCLUSIONS: This study provides evidence of the life-saving benefits of universal helmet laws. The results also suggest that age-specific helmet laws are not effective in protecting the youngest drivers. This is not surprising, as these laws are largely unenforceable. 相似文献

The behaviors of chlorine isotopes in relation to air-sea flux variables have been investigated through multivariate statistical analyses (MSA). The MSA technique provides an approach to reduce the data set and was applied to a set of 7 air-sea flux variables to supplement and describe the variation in chlorine isotopic compositions (delta37Cl) of ocean water. The variation in delta37Cl values of surface ocean water from 51 stations in 4 major world oceans--the Pacific, Atlantic, Indian and the Southern Ocean has been observed from -0.76 to +0.74 per thousand (av. 0.039+/-0.04 per thousand). The observed delta37Cl values show basic homogeneity and indicate that the air-sea fluxes act differently in different oceanic regions and help to maintain the balance between delta37Cl values of the world oceans. The study showed that it is possible to model the behavior of chlorine isotopes to the extent of 38-73% for different geographical regions. The models offered here are purely statistical in nature; however, the relationships uncovered by these models extend our understanding of the constancy in delta37Cl of ocean water in relation to air-sea flux variables. 相似文献
