The aim of this study was to determine oxidative alterations leading to cellular dysfunctions in Pb-exposed subjects by evaluating damage to all major classes of biomolecules in the cell, lipid peroxidation, protein and DNA damage and determine relationships between parameters of Pb toxicity and specific biomarkers of oxidative damage.

Analysis was conducted of smelter workers with high blood Pb and urine aminolevulinic acid levels and slightly elevated values of coproporphyrin and erythrocyte protoporphyrin IX. Significant decreases of thiol groups and increases in carbonyl groups as protein degradation end products, and of nitrite were detected. Elevated rates of lipid peroxidation and rises in the activities of the antioxidant enzymes Cu–Zn superoxide dismutase and catalase were also observed. Both enzymes showed positive correlations with the blood lead levels and urine coproporphyrin, while thiol groups correlated negatively with the same indices. The genotoxic potential of lead was manifested through an increased number of DNA-damaged cells. Increased activities of serum lactate dehydrogenase isoenzymes indicated cellular damage in the lungs, kidneys, and liver. These lead-induced impairments should be taken into consideration in the assessment of Pb-related health hazards.  相似文献

In this study, the performance and the efficiency of physico-chemical effluent treatment in a paper mill was investigated, with the aim of evaluating the treatment rationality and running stability. The effluent treatment plant (ETP) uses alum as a coagulant and polyacrylamide as a flocculant in multi-stage water purification. Inadequate efficiency and some operational problems were observed and their occurrences were investigated. Several simple measures inside the mill and some technical measures can prevent or postpone expensive end-of-pipe treatment investments, such as biological treatment. Chemically enhanced primary treatment under optimal operating conditions can be a stand-alone technique for achieving high quality effluent. Furthermore, reductions in wastewater generation in combination with fiber and filler recovery provide environmental benefits as well as significant economic savings in production. The advantages of the cleaner production approach in existing treatment units compared to expensive biological end-of-pipe technologies are obvious. This study demonstrated that the examined mill could easily and cost-effectively change its water system toward strict discharge limits by continuing to use the existing treatment units.  相似文献

Genotoxicity of freshwater pollution was assessed by measuring DNA damage in haemocytes of caged freshwater crayfish Astacus leptodactylus by the means of Comet assay and micronucleus test, integrated with the measurements of physiological (total protein concentration) and immunological (total haemocyte count) haemolymph parameters as biomarkers of undergone stress. Crayfish were collected at the reference site (River Mre?nica) and exposed in cages for 1 week at three polluted sites along the Sava River (Zagreb, Sisak, Krapje). The long term pollution status of these locations was confirmed by chemical analyses of sediments. Statistically significant increase in DNA damage measured by the Comet assay was observed at all three polluted sites comparing to the crayfish from reference site. In addition, native crayfish from the mildly polluted site (Krapje) cage-exposed on another polluted site (Zagreb) showed lower DNA damage than crayfish from the reference site exposed at the same location indicating adaptation and acclimatisation of crayfish to lower levels of pollution. Micronuclei induction showed similar gradient of DNA damage as Comet assay, but did not reach the statistical significance. Observed increase in total haemocyte count and total protein content in crayfish from polluted environments in the Sava River also confirmed stress caused by exposure to pollution. The results of this study have proved the applicability of caging exposure of freshwater crayfish A. leptodactylus in environmental genotoxicity monitoring using Comet assay and micronucleus test.  相似文献

Despite that the use of DDT has been restricted for more than 40 years to malaria affected areas, low doses of this pesticide and its metabolites DDE and DDD can be found in the environment around the world. Although it has been shown that these pollutants induce cell and DNA damage, the mechanisms of their cytogenotoxic activity remains largely unknown. This study looks into their possible genotoxic effects, at doses that can be found in body fluids, on human lymphocytes using the cytokinesis-block micronucleus assay and the comet assay. After exposure for 1, 6, and 24 h compounds p,p′-DDT (0.1 μg mL−1), p,p′-DDE (4.1 μg mL−1), and p,p′-DDD (3.9 μg mL−1) showed increase in DNA damage. The most significant results were observed at exposure period of 24 h where number of micronucleated cells increased from control 2.5 ± 0.71 to 23.5 ± 3.54, 13.5 ± 0.71, and 16.5 ± 6.36 for DDT, DDE, and DDD, respectively. Similar effect was observed using comet test where the percentage of DNA in comets tail increased from control 1.81 ± 0.16 to 17.24 ± 0.55, 11.21 ± 0.56 and 9.28 ± 0.50 for each compound, respectively. At the same time Fpg-comet assay failed to report induction of oxidative DNA damage of these pollutants. Additionally, the type of cell death was determined using diffusion assay and necrosis dominated. Our findings suggest that even at low concentrations, these pesticides could induce cytogenetic damage to human peripheral blood lymphocytes and in that manner have the impact on human health as well.  相似文献

The aim of this study was to determine the possible effects of meteorological factors on the crown condition of Norway spruce in habitats with different site humidity. In many cases the correlation between meteorological indices and crown condition was stronger and more reliable in habitats with normal humidity than in overmoist habitats. Our results suggest that meteorological conditions during the vegetation period play more significant role compared with dormant period for spruce crown condition.  相似文献

Although there are tendencies to develop a single common index which would describe an overall air quality status within an area, constructed from a choice of measurements of individual pollutants, indices describing individual pollutants themselves have several potentials which can be used in ways which are not possible with pollutant concentrations. On the case of Belgrade, Serbia, we investigated possibilities of using such indices for comparisons between pollutants, characterization of monitoring sites, and extending their use to include elements of population exposure. A methodology of adjusting the results obtained at monitoring stations located in severe pollution conditions, like street canyons, is proposed and used.  相似文献

We report an unusual case of the complete Currarino triad diagnosed in a fetus at 21 weeks' gestation by means of prenatal ultrasonography (US). The highly suspicious findings in the fetus were accompanied by analogous US findings in the mother who suffered from mild symptoms of up to that time unrecognized Currarino triad. Consecutively, magnetic resonance imaging (MRI) confirmed the findings simultaneously in the fetus and in her mother. This is the first report describing the prenatal diagnosis of Currarino triad without the background of positive family history.
