Chlorpyrifos (O,O-diethyl O-3,5,6-trichloro-2-pyridylphosphorothioate) is one of the organophosphate pesticides widely used in agricultural practices throughout world and irreversible inhibitor of cholinesterase in all animal species. Limited efforts have been made to study acute genotoxic effects of chlorpyrifos (CPF) in different tissues of fish using genotoxic biomarkers. Therefore, the present investigation was aimed to study the induction of DNA damage by CPF in freshwater teleost fish Channapunctatus using micronucleus assay (MN assay) and alkaline single-cell gel electrophoresis (comet assay). The value of LC(50) - 96 h of CPF was determined as 811.98 microgl(-1) for C. punctatus, in a semi-static system and on the basis of LC(50) value three acute concentrations viz., 203, 406 and 609 microgl(-1) were determined. The fishes were exposed to the different concentrations of CPF for 96 h and samplings were done at regular intervals for assessment of the MN frequencies and DNA damage. In general, significant effects (P<0.01) from both concentrations and time of exposure were observed in exposed fishes. It was found that the micronucleus induction was highest on 96 h at all concentrations in the peripheral blood. Similar trend was observed for the DNA damage measured in terms of the percentage of tail DNA in the lymphocyte and gill cells. This study explored the combined use of micronucleus assay and comet assay for in vivo laboratory studies using fresh water fish for screening the genotoxic potential of xenobiotics.  相似文献

In the present experiment the seeds of Cicer arietinum (L.) cv. Uday were inoculated with specific Rhizobium grown in sandy loam soil and were allowed to grow for 15 days. At this stage, the seedlings were supplied with 0, 50, 100 or 150 microM of cadmium in the form of cadmium chloride and sprayed with 0.01 microM of 28-homobrassinolide (HBL) at 30-day stage. The data indicated that plant fresh and dry mass, number of nodules, their fresh and dry mass, leghemoglobin content, nitrogen and carbohydrate content in the nodules, leaf chlorophyll content, nitrate reductase and carbonic anhydrase activities decreased proportionately with the increasing concentrations of cadmium but the content of proline and the activities of catalase, peroxidase and superoxide dismutase increased. The ill effect, generated by cadmium, was overcome if the stressed plants were sprayed with HBL.  相似文献

Corrosion deposits formed within drinking water distribution systems deteriorate drinking water quality and resultantly cause public health consequences. In the present study, an attempt was made to investigate the concurrent conditions of corrosion scales and the drinking water quality in selected water supply schemes (WSS) in districts Chitral, Peshawar, and Abbottabad, northern Pakistan. Characterization analyses of the corrosion by-products revealed the presence of α-FeOOH, γ-FeOOH, Fe3O4, and SiO2 as major constituents with different proportions. The constituents of all the representative XRD peaks of Peshawar WSS were found insignificant as compared to other WSS, and the reason could be the variation of source water quality. Well-crystallized particles in SEM images indicated the formation of dense oxide layer on corrosion by-products. A wider asymmetric vibration peak of SiO2 appeared only in Chitral and Abbottabad WSS, which demonstrated higher siltation in the water source. One-way ANOVA analysis showed significant variations in pH, turbidity, TDS, K, Mg, PO4, Cl, and SO4 values, which revealed that these parameters differently contributed to the source water quality. Findings from this study suggested the implementation of proper corrosion prevention measures and the establishment of international collaboration for best corrosion practices, expertise, and developing standards.  相似文献

The cement industry is one of the largest carbon dioxide (CO2) emitters in the Thai industry. The cement sector accounted for about 20,633 kilotonnes (ktonnes) CO2 emissions in 2005 in Thailand. A bottom-up CO2 abatement cost curve (ACC) is constructed in this study for the Thai cement industry to determine the potentials and costs of CO2 abatement, taking into account the costs and CO2 abatement of different technologies. The period of 2010–2025 is chosen as the scenario period. We analyzed 41 CO2 abatement technologies and measures for the cement industry. Using the bottom-up CO2 ACC model, the cost-effective annual CO2 abatement potential for the Thai cement industry during the 15 year scenario period (2010–2025) is equal to 3095 ktonnes CO2/year. This is about 15% of the Thai cement industry’s total CO2 emissions in 2005. The total technical annual CO2 abatement potential is 3143 ktonnes CO2/year, which is about 15.2% of the Thai cement industry’s total CO2 emissions in 2005. We also conducted a sensitivity analysis for the discount rate parameter.  相似文献

Tuberous sclerosis (TSC) is a frequent autosomal-dominant condition (affecting 1 in 6000 individuals) caused by various mutations in either the hamartin (TSC1) or the tuberin gene (TSC2). This allelic and non-allelic heterogeneity makes genetic counseling and prenatal diagnosis difficult, especially as a significant proportion of TSC cases are due to de novo mutations. For this reason the identification of the disease causing mutation is mandatory for accurate counseling, yet current mutation detection methods such as single-strand conformation polymorphism (SSCP) or denaturing gradient gel electrophoresis (DGGE) are labor intensive with limited detection efficiency. Denaturing high-performance liquid chromatography (DHPLC) is a high-throughput, semi-automated mutation detection system with a reported mutation detection rate close to 100% for PCR fragments of up to 800 bp. We used a recently described DHPLC assay allowing the efficient detection of mutations in TSC1 to analyze the DNA extracted from a chorion villus sample in order to perform a prenatal diagnosis for TSC. The fetus was found not to have inherited the deleterious mutation and the DHPLC diagnosis was confirmed by haplotype analysis. In this paper, COSMO numerical weather prediction model is used to simulate land-sea breeze circulation in the north coast of Oman and to analyze the variability of the PBL depth. Typical summer day 1st August 2009 and winter day 1st January 2009 were simulated and verified by surface observations. The COSMO model was initialized using the German global model which runs as 40 km resolution and 40 vertical layers. The simulations show differences in the onset, strength, and inland penetration of the sea breeze. The high sea-land thermal contrast in the summer season induces earlier onset and progression of the sea breeze with larger horizontal and vertical extents. The summer and winter inland penetration is simulated around 150 and 65 km, respectively. The PBL depth was defined from the verticals profiles of the turbulence kinetic energy (TKE). The simulated PBL height varied at 1,600 m in summer and 700 m in the winter case. These results are in general in good agreement with the simulations of the National Centers for Environmental Prediction Global Data Assimilation System (NCEP-GDAS) model. The dispersive ability of the atmosphere was tested through the vertical mixing coefficient (VMC) generated by NCEP-GDAS simulations. The VMC during the summer simulated case is approximately four times larger than the respective one during the winter case. Consequently, during the advection of winter sea breeze, a shallow dome less than 100 m is shaped and forms an obstacle to the air-pollutant dispersion.  相似文献

In 1999, two earthquakes in northwest Turkey caused heavy damage to a large number of industrial facilities. This region is the most industrialized in the country, and heavy damage has a significant economic influence. Industrial storage tanks, ruptured by earthquakes, exascerbate damage through the spread of fire. Storage tanks are uniquely structured, tall cylindrical vessels, some supported by relatively short reinforced concrete columns. The main aim of this study is to evaluate the earthquake performance of Turkish industrial facilities, especially storage tanks in terms of earthquake resistance. Modeling a typical storage tank of an industrial facility helps to understand the structure’s seismic response. A model tank structure was modelled as a solid with lumped mass and spring systems. Performance estimation was done with 40 different earthquake data through nonlinear time history analyses. After the time history analyses, fragility analyses produced probabilistic seismic assessment for the tank model. For the model structure, analysis results were evaluated and compared. In the study, vulnerability of storage tanks in Turkey was determined and the probabilistic risk was defined with the results of the analysis.  相似文献

When agricultural lands are no longer used for agriculture and allowed to recover its natural vegetation, soil organic carbon can accumulate in the soil. Measurements of soil organic carbon and aggregate stability changes under various forms of land use are needed for the development of sustainable systems. Therefore, comparison of soil samples taken from both agricultural and nearby area close to land-mined fields where no agricultural practices have been done since 1956 can be a good approach to evaluate the effects of tillage and agriculture on soil quality. The objective of this study was to compare tillage, cropping and no tillage effects on some soil-quality parameters. Four different locations along the Turkey–Syria border were selected to determine effects of tillage and cropping on soil quality. Each location was evaluated separately because of different soil type and treatments. Comparisons were made between non-tilled and non-cropped fallow since 1956 and adjacent restricted lands that were tilled about every 2 years but not planted (T) or adjacent lands tilled and planted with wheat and lentil (P). Three samples were taken from the depths of 0–20 and 20–40 cm each site. Soil organic carbon (SOC), pH ,electrical conductivity, water soluble Ca++, Mg++, CO3-2{\rm CO}_{3}^{-2} and HCO3-{\rm HCO}_{3}^{-}, extractable potassium (K+) and sodium (Na+), soil texture, ammonium (NH4+{\rm NH}_{4}^{+}–N) and nitrate (NO3–N), extractable phosphorous and soil aggregate stability were determined. While the SOC contents of continuous tillage without cropping and continuous tillage and cropping were 2.2 and 11.6 g kg−1, respectively, it was 30 g kg−1 in non-tilled and non-planted site. Tillage of soil without the input of any plant material resulted in loss of carbon from the soil in all sites. Soil extractable NO3−N contents of non-tilled and non-cropped sites were greatest among all treatments. Agricultural practices increased phosphorus and potassium contents in the soil profile. P2O5 contents of planted soils were approximately 20 to 39 times greater than those of non-tilled and non-cropped soils at different sites. FTIR spectra showed that never tilled sites had greater phenol, carboxylic acid, amide, aromatic compounds, polysaccharide and carbohydrates than other treatments.  相似文献
