Hepatitis A virus (HAV) is currently recognized as an important human food borne pathogen, and it is one of the most resistant
enteric RNA viruses, is highly infectious, and may lead to widespread outbreaks. The aim of this study was to optimize the
methods to detect HAV from artificially contaminated food. To this end, strawberry and lettuce were experimentally contaminated
with HAV suspension containing 6 × 106 copies/ml. After contamination, HAV persistence and washing procedure were evaluated at 0, 1, 3, 7, and 9 days of storage.
Five elution buffers (PBS (pH 7.4)/0.1% Tween80; 50 mM glycine/3% (wt/vol) beef extract (pH 9.5); PBS (pH 7, 4); 25 mM glycine/0.1
Tween80; and 1 M sodium bicarbonate) were used to elute the virus, and qualitative and quantitative PCR were used for HAV
detection. HAV was detected by qualitative and quantitative PCR using any of the five elution buffers, but PBS was the most
effective. Even after washing, HAV was detected up to 9 days after contamination by quantitative PCR. Quantitative PCR was
more sensitive than qualitative PCR since samples containing viral load lower than 1.4 × 103 copies/ml could not be detected by qualitative PCR. Quantitative PCR can be used for rapid detection of food borne viruses

The dynamic and residues of florasulam and flumetsulam in corn field ecosystem were investigated using quick, easy, cheap, effective, rugged, and safe (QuEChERS) procedure with high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The limits of quantification (LOQs) of the proposed method ranged from 0.005 to 0.01 mg/kg. Mean recoveries and relative standard deviations (RSDs) of the two compounds in all samples at three spiking levels ranged 94–110 % and 2.0–9.2 %, respectively. Florasulam and flumetsulam degradation followed first-order kinetics with half-lives 1.7–2.9 and 3.3–8.7 days in soil and 1.3–1.8 and 0.9–1.7 days in plant, respectively. The residues in all the samples were found to be less than the LOQs at preharvest intervals of 53 and 78 days. The results suggest that the combined use of florasulam and flumetsulam on corn is considered to be safe under the recommended conditions and can be utilized for establishing the maximum residue limit (MRL) of florasulam in corn in China. 相似文献

The present study investigates the use of combined methods of optical and acoustic sensors, in collaboration with direct in situ measurements, for the calibration and validation of a model transforming acoustic backscatter intensity series into suspended particulate matter (SPM) concentration datasets. The model follows previously elaborated techniques, placing particular attention to the parameterization of the acoustic absorption index as a function of water physical properties. Results were obtained from the annual deployment (during 2007–2008) of an upward-facing acoustic Doppler current profiler (ADCP) (307 kHz), equipped with a Wave Array, and an optical backscatter sensor (OBS), at the bottom of Thassos Passage near Nestos River plume (Thracian Sea, Northern Greece). The OBS was calibrated through linear regression, using 2007 and 2012 field sampling data, exhibiting an error of 13–14 % due to chlorophyll presence. The ADCP signal was calibrated through simultaneous measurements of backscatter intensity and turbidity profiles. Harmonic analysis on the model-produced SPM concentrations explained the tidal influence on their variability, especially during the summer. Empirical orthogonal functions analysis revealed the impact of waves and wave-induced currents on SPM variability. Finally, Nestos River sediment load was found uncorrelated to the SPM change in Thassos Passage, due to the dispersal and sediment deposition near the river mouth. 相似文献

Zinc, copper, iron, chromium and cobalt are essential elements for human health, showing toxicity only in high concentrations, while lead and cadmium are extremely toxic even as traces. Therefore, it is important to monitor the contents of toxic metals in vegetables. Large number of vegetables is grown and used in nutrition, in Kosovo. The concentrations of selected elements in vegetables (radish, onion, garlic and spinach) from Kosovo were determined using ICP-OES method. Oral intake of metals and health risk index were calculated. Statistical analysis indicated numerous positive correlations between concentrations of selected elements in vegetables. As a result of principal component analysis, 15 new variables were obtained which were characterized by eigenvalues. The sequence of health quotients for the heavy metals followed the decreasing order Zn?=?Mn?>?Pb?>?Cu?>?Ni?>?Fe?>?Cd?>?Co?>?Cr. The health quotients for all investigated heavy metals were below 1 (one), which is considered safe. The vegetables from Kosovo are mainly safe for use in everyday diet. 相似文献

Distinguishing between soluble and particulate lead in drinking water is useful in understanding the mechanism of lead release and identifying remedial action. Typically, particulate lead is defined as the amount of lead removed by a 0.45-μm filter. Unfortunately, there is little guidance regarding selection of filter membrane material and little consideration to the possibility of the sorption of dissolved lead to the filter. The objective of this work was to examine the tendency of 0.45-μm syringe filter materials to adsorb lead. Tests were performed with water containing 40 and 24 μg/L soluble lead at pH 7 buffered with 50 mg C/L dissolved inorganic concentration (DIC). The amounts of lead sorbed greatly varied by filter, and only two filter types, polypropylene and mixed cellulose esters, performed well and are recommended. Great care must be taken in choosing a filter when filtering soluble lead and interpreting filter results. 相似文献

Estimation of essential and toxic element intakes is crucial to evaluate the risks of deficiency or toxicity. The purpose of this study was to investigate and also to compare the dietary intakes of zinc (Zn) and copper (Cu) by adults living in Shiraz, Iran, determined by two procedures: duplicate portion sampling of 21 hospital meals combined with either instrumental analysis (voltammetric measurement) or the use of food composition tables (FCTs). The level of exposure to lead (Pb) and cadmium (Cd) was evaluated as well. The daily Zn intakes of both methods were not significantly different and were higher than the RDA values except the value measured by the instrumental analysis which was lower than the RDA established for a male adult. Daily intake of Cu determined by instrumental analysis was significantly lower and closer to RDA for adults compared with the value estimated by FCTs. The dietary intakes of Pb and Cd were 313 and 61 % of the respective provisional tolerable weekly intakes (PTWIs), respectively. The accuracy of two methods used for estimation of Zn intake was similar. In the case of Cu, the use of FCTs, in which the influence of environmental conditions and dietary habits of meal preparation is not taken into account, overestimated dietary intake. The risk of zinc deficiency was found in adult males. Moreover, the estimated intake of Pb, but not Cd, could be a cause of concern for Shiraz population. Thus, conducting regular periodic studies to assess the dietary intake of mentioned elements are recommended. 相似文献

Dissipation and decontamination of chlorantraniliprole (Coragen 18.5 SC) in brinjal and okra fruits were studied following field application at single and double doses of 30 and 60 g ai ha?1, and the residues of the insecticide was estimated using LC-MS/MS. Initial residues of chlorantraniliprole at single and double doses on the fruits of brinjal were 0.72 and 1.48 mg kg?1, while on okra fruits, the residues were 0.48 and 0.91 mg kg?1, respectively. The residues reached below detectable level of 0.01 mg kg?1 on the 10th day. Half-life of chlorantraniliprole at 30 and 60 g ai ha?1 on brinjal was 1.58 and 1.80 days with the calculated waiting period of 0.69 and 2.38 days, whereas on okra, the values were 1.60 and 1.70 and 0 and 1.20 days, respectively. The extent of removal of chlorantraniliprole using simple decontaminating techniques at 2 h and 3 days after spraying was 40.99–91.37 % and 29.85–89.12 %, respectively, from brinjal fruits and 47.78–86.10 % and 41.77–86.48 %, respectively, from okra fruits. 相似文献

The contribution presents numerical simulation of gypsum particles, lifting from a gypsum landfill. First, particle characteristics
are presented, resulting from different technologies of gypsum depositing. Next, a laboratory experiment parameter validation
tests are described, which served as a means of determination of mass flow of particles from the landfill. The background
of the numerical simulations, used in the assessment of landfill impact on the environment, is also described. Simulations
consist of two parts: simulation of a long term impact of the particles on the surrounding area, performed by implementation
of the Gaussian dispersion model based computer code ISC3, and second, a CFD based simulation for assessing the flow and mass
concentration fields in the vicinity of the landfill for several pre-selected flow cases. The results of both computational
approaches are presented and compared. In the conclusions, a relation of the simulation results with existing environmental
pollution levels is made, and recommendations for landfill management are drawn. 相似文献

A 2D analytical turbulent diffusion model for particle dispersion and deposition at different heights along the pipe flow and circumferential deposition has been developed. This liquid–solid turbulent diffusion model presented in this paper has emanated from an existing gas–liquid turbulent diffusion model. This model can be used as a handy tool for quick estimation one and two-dimensional deposition fluxes of particles in water distribution networks. A comprehensive 3D numerical investigation has been carried out using multiphase mixture model available in “Fluent 6.2” to verify the above analytical model. Different particles sizes and densities were used for 3D numerical investigations. The deposition was studied as a function of particle diameter, density, and fluid velocity. The deposition of particles, along the periphery of the pipe wall and at different depths, was investigated. Both the models findings matched with qualitative phenomena such as deposition of heavier particles at the bottom of the pipe wall were higher at lower velocities and lower at higher velocities. The lighter particles were found mostly suspended with homogeneous distribution. Smaller particles were also suspended with marginal higher concentration near the bottom of the pipe wall. This marginal higher concentration of the smaller particles was found to be slightly pronounced for lower velocity. These analogies of particles are well discussed with the ratio between free-flight velocity and the gravitational settling velocity. Extended analytical model results were compared with the 3D computational fluid dynamics simulation results. Discrepancies in the model results were discussed. 相似文献

Groundwater flow modelling is an important tool in simulating and predicting hydraulic behaviour of groundwater transporting
in the domain consisted of groundwater flow zone (aquifer) and permeable reactive barriers (PRBs). The aquifer regime is modelled
using the Darcy equation, whereas PRBs are simulated by the Brinkman equation. By combining the above equations, the present
paper is devoted to analyse the effect of permeability of aquifer (K
aq
) and reactive media (K
p
), groundwater flow velocity, and barrier size as well as assembled style of barriers on key barrier design parameter, such
as the hydraulic capture zone width. The simulations indicate that the capture zone width generally decreases with a decrease
in K
p
at a given K
aq
; however, there is relatively little decrease in capture zone width when the K
p
of freshly installed reactive media is roughly 10 times higher than the K
aq
. For a given barrier system, the absolute capture zone width increases with an increase in barrier size, whereas the increment
of relative capture zone width decreases dramatically. Although the capture zone width remains relatively unchanged as groundwater
flow velocity increases, it results in a shorter residence time in the reactive media that may require greater barrier thickness
so that contaminant levels can be reduced to regulatory limits. In addition, the effect of parallel barriers interaction on
the capture zone width should be incorporated into the design. The present paper is based on the continuous reactive barrier
and homogeneous barrier system. Similar methods can be used to analyse the hydrodynamics of funnel-and-gate system and heterogeneous
