Containment of genetically modified (GM) microorganisms such as Escherichia coli is a legal requirement to protect the environment from an unintended release and to avoid horizontal gene transfer (HGT) of recombinant DNA to native bacteria. In this study, we sampled the laboratory wastewater (LWW) at a large Swiss university from three sources over 2 years and cultured ampicillin-resistant, presumptive GM E. coli. From a total of 285 samples, 127 contained presumptive GM E. coli (45%) at a mean concentration of 2.8 × 102 CFU/ml. Plasmid DNA of 11 unique clones was partially or entirely sequenced. All consisted of cloning vectors harboring research-specific inserts. To estimate the chance of HGT between GM E. coli and native bacteria in LWW, we identified taxa representative for the bacterial community in LWW using 16S rRNA amplicon sequencing and measured conjugation frequencies of E. coli with five LWW isolates. At optimal conjugation conditions, frequencies were between 3.4 × 10−3 and 2.4 × 10−5. Given the absence of transferable broad-host range plasmids and suboptimal conjugation conditions in the LWW system, we conclude that the chance of HGT is relatively low. Still, this study shows that the implementation of robust containment measures is key to avoid the escape of GM microorganisms.

A southern Italian area that is characterized by large outcrops of rocks that are rich in titanium oxide (TiO2) phases were investigated to determine the mineralogical risk induced by the natural dispersion of TiO2 minerals. Rock, sediment and surface water samples were collected to determine the physicochemical and mineralogical factors (i.e., size distribution, morphology and alteration) indicative of potential TiO2 toxicity. X-ray diffraction data suggested that titanium oxides were present as rutile and anatase. Scanning electron microscopy images showed elongated TiO2 morphologies; fibres were found as either isolated or embedded/enclosed in flake-like phyllosilicates. The concentration of fibres in stream water ranged from 1.7 to 4.6 million fibres per litre. The highest fibre amounts in the sediments were in the <8-µm fraction, while single fibres were primarily concentrated in the <2-µm fraction. The results indicate that titanium oxide minerals represent a natural source of environmental risk and that the geomineralogical characterization of rich TiO2 areas is indispensable for understanding their geoavailability, dispersion and distribution.

- DOI: http://dx.doi.org/10.1065/espr2006.01.009 Background, Aims and Scope Most existing models used to describe the fate of chemicals in surface water and sediment generally consider a 'static scenario', in which a contaminant is discharged at a constant rate and environmental input parameters do not change during the simulation time. This approach is not suitable in environmental scenarios characterized by daily or periodic changes of several input parameters. The aim of this study is to estimate approximate emissions of DDT lo Lake Maggiore using a new surface water model, (DynA Model) that describes the fate of a chemical in a dynamic scenario. Methods The model is developed on the grounds of an existing and validated model (QWASI). A numerical solution was adopted to build the fully dynamic version of the model. Results and Discussion The model was applied to Lake Maggiore emitting DDT at a constant rate until steady-state was reached. Emissions were stopped and later sporadic 'pulse' emissions were added. This was done to calculate the amount of DDT needed to simulate concentrations close to those measured in water and sediments. This allowed the evaluation of the order of magnitude of emissions. An uncertainty analysis for sediment resuspension was also performed, given the lack of measured resuspension rates. Conclusion The model showed the time response of the Lake Maggiore system to varying emission scenarios and provided what are regarded as reasonable estimates of DDT emissions. The model demonstrated the importance of sediment-water exchange. Recommendation and Outlook In order to better calculate DDT concentrations the model should be run with different discharge scenarios to clarify the time trends of concentrations, possibly with the use of different sets of measured data (such as biota and sediment deposition/resuspension rates).  相似文献

The number and diversity of civil society or third sector sustainability organisations (TSSOs) have increased in recent decades. TSSOs play a prominent role in local approaches to sustainability. However, the contributions made by TSSOs are not fully understood, beyond a limited suite of quantifiable outputs and impacts. In this qualitative study, we examine how four TSSOs from two Australian regions, Tasmania and Queensland's Sunshine Coast, contribute to social transformation beyond discrete outputs. We examine the operation, ethos, scope and influence of these organisations over time. In so doing, we identify three common ways in which these organisations facilitate social change: by (i) enhancing social connectivity through boundary work; (ii) mobilising participatory citizenship and (iii) contributing to social learning. We conclude that TSSOs contribute significantly to the systemic social conditions that enable change for sustainability and the development of community resilience and well-being, but do so in ways undervalued by existing metrics, formal evaluation processes and funding models. Clearer recognition of, and strategic emphasis on, these qualitative contributions to social transformation is vital in ensuring that TSSOs remain viable and effective over the long term.  相似文献

According to the desperado sibling hypothesis, chicks of obligately siblicidal species kill their junior broodmates as early as possible because junior broodmates face dire ecological prospects and are consequently predisposed to aggressively overthrow elder broodmates by all-out aggression. The agonistic behavior of junior chicks is virtually unknown because they die so young. We investigated the agonistic behavior of hypothetical desperados by fostering junior brown boobies (Sula leucogaster, an obligately siblicidal species) into nests of the blue-footed booby (S. nebouxii, a facultatively siblicidal species) containing an older singleton blue-foot chick. Controls were junior blue-footed boobies fostered into the same situation. Junior brown boobies were 7 times more aggressive than controls and most of them dominated their elder and larger nestmates. Four of nine brown booby juniors showed relentless aggression, delivering up to 711 pecks, bites and pushes (including "expulsion pushes") per hour, thereby overwhelming nestmates 90% heavier and permanently expelling one of them from the nest. Similarly, in natural broods of two surviving brown booby chicks, the losing chick was 13 times as aggressive as blue-foot subordinates, up to at least age 7 weeks. Contrast of the two species of booby suggests the evolution of agonistic roles within broods may be partly driven by selection on potential victims to express a level and type of aggressiveness appropriate to their status-related ecological prospects.  相似文献

Hydropower developments along the main stem of the Mekong River and its tributaries cause transboundary effects within the Mekong Basin Region, which comprises parts of six countries. On the one hand, the provision of hydropower triggers economic development and helps to meet the rising energy demand of the Mekong riparian countries, especially China, Thailand, and Vietnam. On the other hand, the negative impact of dam construction, mainly altered water flow and sediment load, has severe impacts on the environment and the livelihoods of the rural Mekong population. Several discrepancies exist in the needs, demands, and challenges of upstream versus downstream countries. Against the common apprehension that downstream countries are powerlessly exposed to mainly negative impacts whereas upstream countries unilaterally benefit from hydropower, the authors argue that upstream–downstream relations are not really clear-cut. This conclusion is based on a consideration of the complex power play between Mekong riparians, with a focus on recent power trade interactions. The article investigates the consequences of hydropower dams for the Mekong region as well as the role of supranational players, such as the Mekong River Commission and the Greater Mekong Subregion Initiative, on the hydropower debate. It is not nations that are the winners or losers in the hydropower schemes in the Mekong, but rather parts of the riparian population: a few influential and powerful elites versus the large mass of rural poor.  相似文献

Hypochondroplasia (HCH) is a genetic skeletal dysplasia, inherited in an autosomal dominant fashion. About 50–70% of HCH patients have a mutation in FGFR3 gene and in the majority of cases it is a de novo mutation. Recent magnetic resonance imaging studies on relative large cohorts of HCH patients have showed a central nervous system involvement with a high incidence of characteristic temporal lobe and hippocampal abnormalities. To the best of our knowledge, this report shows the first magnetic resonance imaging prenatal detection of characteristic brain anomalies in a case of HCH, molecularly confirmed through postnatal FGFR3 analysis. © 2014 John Wiley & Sons, Ltd.  相似文献

This article reports a study of the economic viability of a separate household waste collection project, and considers its social and environmental benefits. The study was developed from the database obtained through the pilot project on separate collections in João Pessoa, the capital of Paraíba, in the northeast region of Brazil. The pilot project had been supported by the local municipal council since September 2000. The methodology applied to this study of economic viability is based on the economic quantification of the benefits and costs involved in the process of a separate collection of household waste. The benefits to the environment, owing to the reduction of the final disposal of household waste, were also evaluated. The results of this research are encouraging for the benefit cost relation (B/C) of both the quantified economic components and the environmental aspects. The values found for the relation B/C ranged from 1.27 to 1.77 depending on the economic quantification of the direct and indirect benefits to the components involved in the project, i.e., the waste collectors, the municipal council, the sellers of recycled material, and the recycling industry.  相似文献

Extreme weather and climate-related events affect human health by causing death, injury, and illness, as well as having large socioeconomic impacts. Climate change has caused changes in extreme event frequency, intensity, and geographic distribution, and will continue to be a driver for change in the future. Some of these events include heat waves, droughts, wildfires, dust storms, flooding rains, coastal flooding, storm surges, and hurricanes. The pathways connecting extreme events to health outcomes and economic losses can be diverse and complex. The difficulty in predicting these relationships comes from the local societal and environmental factors that affect disease burden. More information is needed about the impacts of climate change on public health and economies to effectively plan for and adapt to climate change. This paper describes some of the ways extreme events are changing and provides examples of the potential impacts on human health and infrastructure. It also identifies key research gaps to be addressed to improve the resilience of public health to extreme events in the future.
