Isolation of pathogenic bacteria from non-biological material of similar size is a vital sample preparation step in the identification of such organisms, particularly in the context of detecting bio-terrorist attacks. However, many detection methods are impeded by particulate contamination from the environment such as those from engine exhausts. In this paper we use dielectrophoresis--the induced motion of particles in non-uniform fields--to successfully remove over 99% of diesel particulates acquired from environmental samples, whilst letting bacterial spores of B. subtilis pass through the chamber largely unimpeded. We believe that such a device has tremendous potential as a precursor to a range of detection methods, improving the signal-to-noise ratio and ultimately improving detection rates.  相似文献

In the present study, selected advanced oxidation processes (AOPs)—namely, photo-Fenton (with Fe2+, Fe3+, and potassium ferrioxalate—FeOx—as iron sources), solar photo-Fenton, Fenton, and UV/H2O2—were investigated for degradation of the antineoplastic drug mitoxantrone (MTX), frequently used to treat metastatic breast cancer, skin cancer, and acute leukemia. The results showed that photo-Fenton processes employing Fe(III) and FeOx and the UV/H2O2 process were most efficient for mineralizing MTX, with 77, 82, and 90 % of total organic carbon removal, respectively. MTX probably forms a complex with Fe(III), as demonstrated by voltammetric and spectrophotometric measurements. Spectrophotometric titrations suggested that the complex has a 2:1 Fe3+:MTX stoichiometric ratio and a complexation constant (K) of 1.47 × 104 M–1, indicating high MTX affinity for Fe3+. Complexation partially inhibits the involvement of iron ions and hence the degradation of MTX during photo-Fenton. The UV/H2O2 process is usually slower than the photo-Fenton process, but, in this study, the UV/H2O2 process proved to be more efficient due to complexing of MTX with Fe(III). The drug exhibited no cytotoxicity against NIH/3T3 mouse embryonic fibroblast cells when oxidized by UV/H2O2 or by UV/H2O2/FeOx at the concentrations tested.  相似文献

This research work consists on valorizing poultry waste by biotransformation into biofertilizers, associating this agro-industrial waste with algae (abundant natural resources) and molasses (a by-product of the sugar refining industry) ensuring a good contribution of nutritional chemical elements and obtaining a balanced formulation. A total of seven different formulations of the above three components, were examined in a simplex centroid design. A fungal inoculum of Aspergillus niger was used as a fermentation agent for better quality of biotransformation. The monitoring of this biotransformation is ensured during 15 days by following the evolution of physicochemical and microbiological parameters and, to understand the bioconversion of the simple compounds of the biofertilizer mixture such as short aliphatic chains, sugar, and amino acids into soluble mineral compounds, a Fourier transform infrared spectroscopy (FTIR) analysis was carried out before and after biotransformation. Finally, germination and fertilization tests were performed to evaluate the efficiency of the final product on a barley crop. The overall results of the present study showed that the mixture which contained 68.75% poultry waste, 12.5% molasses, and 18.75% algae presented the better microbiological and chemical safety criteria required for a good biofertilizer according to NF U44-551 standard.  相似文献

The exhaust emission of the polycyclic aromatic hydrocarbons (PAHs) considered toxic to human health were investigated on two spark ignition light duty vehicles, one being gasohol (Gasohol, in Brazil, is the generic denomination for mixtures of pure gasoline plus 20–25% of anhydrous ethyl alcohol fuel (AEAF).)-fuelled and the other a flexible-fuel vehicle fuelled with hydrated ethanol. The influence of fuel type and quality, aged lubricant oil type and use of fuel additives on the formation of these compounds was tested using standardized tests identical to US FTP-75 cycle. PAH sampling and chemical analysis followed the basic recommendations of method TO-13 (United States. Environmental Protection Agency, 1999. Compendium Method TO-13A – Determination of polycyclic Aromatic hydrocarbons (PAH) in Ambient Air Using Gas Chromatography/Mass Spectrometry (CG/MS). Center for environmental research information, Cincinnati, p. 78), with the necessary modification for this particular application.Results showed that the total PAH emission factor varied from 41.9 μg km?1 to 612 μg km?1 in the gasohol vehicle, and from 11.7 μg km?1 to 27.4 μg km?1 in the ethanol-fuelled vehicle, a significant difference in favor of the ethanol vehicle. Generally, emission of light molecular weight PAHs was predominant, while high molecular weights PAHs were not detected. In terms of benzo(a)pyrene toxicity equivalence, emission factors varied from 0.00984 μg TEQ km?1 to 4.61 μg TEQ km?1 for the gasohol vehicle and from 0.0117 μg TEQ km?1 to 0.0218 μg TEQ km?1 in the ethanol vehicle.For the gasohol vehicle, results showed that the use of fuel additive causes a significant increase in the emission of naphthalene and phenanthrene at a confidence level of 90% or higher; the use of rubber solvent on gasohol showed a reduction in the emission of naphthalene and phenanthrene at the same confidence level; the use of synthetic oil instead of mineral oil also contributed significantly to a decrease in the emission of naphthalene and fluorene. In relation to the ethanol vehicle, the same factors were tested and showed no statistically significant influence on PAH emission.  相似文献

Results showed that emission factors of PCDD/Fs for the.gasohol vehicle varied from undetected to 0.068 pg international toxic equivalency (I-TEQ) km(-1) (average of 0.0294 pg I-TEQ km(-1)), whereas in the ethanol vehicle they varied from 0.004 to 0.157 pg (I-TEQ) km(-1) (average of 0.031 pg I-TEQ km(-1)). In the gasohol-powered vehicle, the use of fuel additive diminished the emission of Octachlorodibenzo-p-dioxin (OCDD) significantly, whereas in the ethanol vehicle no significant associations were observed between the investigated variables and the emissions.  相似文献

IMPLICATIONS The objective of this work was to analyze differences in emissions from a traditional fossil fuel (gasoline) and an alternative renewable fuel (ethanol from sugarcane), and the influence of fuel additives and lubricant oils on the formation of chlorinated dioxins and furans in spark-ignition light-duty gasohol and ethanol vehicles. Renewable fuels are very important in terms of climate change but the risk to the population's health must not increase. Thus the results of this work could help in the development of environmental impact studies as well as orienting policy-makers in formulating strategies for air pollution control.  相似文献

A novel multivariate method based on principal component analysis of pre-processed sections of chromatograms is used to characterize the complex PAH pollution patterns in sediments from Guanabara Bay, Brazil. Five distinct sources of 3- to 6-ring PAHs could be revealed. The harbour is the most contaminated site in the bay, its plume stretches in a South West to North East direction and the chemical profile indicates mainly pyrogenic sources mixed with a fraction of high-molecular-weight petrogenic PAHs. Rio São João de Meriti is the second largest source of PAHs, and introduces mainly a fraction of low-molecular-weight petrogenic PAHs from the western region of Rio de Janeiro. The sites close to the ruptured pipeline at the Duque de Caxias Refinery show a distinctive pollution pattern indicating a heavy petroleum fraction. The method also led to the identification of new potential indicator ratios also involving coeluting peaks (e.g., triphenylene and chrysene).  相似文献

Lake Vela, located in the Portuguese littoral-centre, is a temperate shallow lake exhibiting a high trophic status. This aquatic ecosystem has been potentially exposed to contamination generated by agricultural and livestock activities held in their drainage basin, which seriously compromise their health. This work summarizes some background information and presents the problem formulation step of the ERA. Therefore, it evaluates the characteristics of the stressor(s), describes the ecological system and receptors and suggests a conceptual model, which predicts the potential exposure pathways, relating suspected sources to the defined endpoints. This introductory step also described an analysis plan on the entire study, which includes a delineation of the assessment design, data needs, measures, and methods for conducting the analysis phase of the risk assessment process.  相似文献
