Human pressure has been exponentially growing during recent decades in coastal areas, which have led to drastic losses of biodiversity in coastal ecosystems. The current conservation status of many coastal plant species is directly related to a lack of environmental criteria in the urban planning of coastal areas over recent decades. This study aimed to evaluate the evolution, over the last 9 years, of the conservation status of various populations of the endangered plant Glaucium flavum, exploring the extent to which human pressure and different management strategies practiced in the coastal areas where the populations are established have affected the conservation status of the species. The populations analysed have evolved in a different manner over the last 9 years, as have their threat factors, and a relationship was evident between their conservation status and the evolution of these different threat factors. Our results indicate that an appropriate planning of local management actions, such as the installation of walkways or the successful eradication of invasive species, may be determinant factors for successful conservation of the coastal vegetation. The presence of species that are sensitive to slight changes in the ecosystem, and the main factors that govern the plant performance of these species, must be given full consideration in decision-making processes of coastal planning and management.  相似文献

Background

The European Food Safety Authority proposed a concept for the environmental risk assessment of genetically modified plants in the EU that is based on the definition of thresholds for the acceptability of potential adverse effects on the environment. This concept, called Limits of Concern (LoC), needs to be further refined to be implemented in the environmental risk assessment of genetically modified organisms.

Methods

We analyse and discuss how LoC can be defined for the environmental risk assessment for three different types of genetically modified plants. We outline protection goals relevant to the genetically modified plants in question and discuss existing concepts and suggestions for acceptability thresholds from the environmental risk assessment of different regulatory areas. We make specific recommendations for the setting and use of LoC for each type of genetically modified plant.

Results

The LoC concept can be suitably applied for the environmental risk assessment of genetically modified organisms, if the different protection goals in agro-environments are specifically considered. Not only biodiversity protection goals but also agricultural protection goals need to be addressed. The different ecosystem services provided by weeds inside and outside agricultural fields have to be considered for genetically modified herbicide-tolerant crops. Exposure-based LoCs are suggested based on knowledge about dose–effect relationships between maize pollen and non-target Lepidoptera for insect-resistant maize. Due to the long-term nature of biological processes such as spread and establishment, LoCs for genetically modified oilseed rape should be defined for the presence of the genetically modified plant or its genetically modified traits in relevant protection goals.

Conclusions

When setting LoCs, the focus should be on protection goals which are possibly affected. Potential overlaps of the LoC concept with the ecosystem service concept have to be clarified to harmonise protection levels in the agro-environment for different stressors. If additional impacts on agro-biodiversity resulting from the cultivation of genetically modified plants are to be avoided, then high protection levels and low thresholds for acceptable effects (i.e. LoC) should be set.

In July 2018, the European Court of Justice (Case C-528/16) ruled that organisms obtained by directed mutagenesis techniques are to be regarded as genetically modified organisms (GMOs) within the meaning of Directive 2001/18. The ruling marked the next round of the dispute around agricultural genetic engineering in Europe. Many of the pros and cons presented in this dispute are familiar from the debate around the first generation of genetic engineering techniques. The current wave of enthusiasm for the new genetic engineering methods, with its claim to make good on the failed promises of the previous wave, seems to point more to an admission of failure of the last generation of genetic engineering than to a true change of paradigm. Regulation is being portrayed as a ban on research and use, which is factually incorrect, and the judges of the European Court of Justice are being defamed as espousing “pseudoscience”. Furthermore, this highly polarised position dominates the media reporting of the new techniques and the court’s ruling. Advocates of the new genetic engineering techniques appear to believe that their benefits are so clear that furnishing reliable scientific evidence is unnecessary. Meanwhile, critics who believe that the institution of science is in a serious crisis are on the increase not just due to the cases of obvious documented scientific misconduct by companies and scientists, but also due to the approach of dividing the world into those categorically for or against genetic engineering. In this construct of irreconcilable opposites, differentiations fall by the wayside. This article is a response to this one-sided and biased reporting, which often has the appearance of spin and lacks journalistic ethics that require journalists to report on different positions in a balanced and factual manner instead of taking positions and becoming undeclared advocates themselves.  相似文献

The paper provides a complex, critical assessment of heavy metal soil pollution using different indices. Pollution indices are widely considered a useful tool for the comprehensive evaluation of the degree of contamination. Moreover, they can have a great importance in the assessment of soil quality and the prediction of future ecosystem sustainability, especially in the case of farmlands. Eighteen indices previously described by several authors (Igeo, PI, EF, Cf, PIsum, PINemerow, PLI, PIave, PIVector, PIN, MEC, CSI, MERMQ, Cdeg, RI, mCd and ExF) as well as the newly published Biogeochemical Index (BGI) were compared. The content, as determined by other authors, of the most widely investigated heavy metals (Cd, Pb and Zn) in farmland, forest and urban soils was used as a database for the calculation of all of the presented indices, and this shows, based on statistical methods, the similarities and differences between them. The indices were initially divided into two groups: individual and complex. In order to achieve a more precise classification, our study attempted to further split indices based on their purpose and method of calculation. The strengths and weaknesses of each index were assessed; in addition, a comprehensive method for pollution index choice is presented, in order to best interpret pollution in different soils (farmland, forest and urban). This critical review also contains an evaluation of various geochemical backgrounds (GBs) used in heavy metal soil pollution assessments. The authors propose a comprehensive method in order to assess soil quality, based on the application of local and reference GB.  相似文献

Spherical TiO2 nanoparticles (npTiO2) were prepared by controlled hydrolysis of tetraethoxy orthotitanate under a nitrogen atmosphere. ZnO nanoparticles (npZnO) were prepared using hydrothermal methods. The crystal structure, chemical, thermal and morphological properties of npZnO and npTiO2 were characterised using Fourier Transform Infrared Spectrometer, enery-dispersive X-ray spectroscopy, X-ray diffraction, and scanning electron microscope techniques. The short- and long-term experiments were started with neonates taken from the same culture and laboratory condition. In the acute experiments, npTiO2, npZnO, and cocktail concentrations were applied. 96h-LC50 values were 1.8, 0.7, and 0.1?mg?L?1, respectively (p?<?.05). For the chronic experiments, different npTiO2 concentrations were performed. 21d-LC50 value was 1.0?mgL?1 (p?<?.05). Morphometry became progressively worse in concentrations of more than 1?mgL?1 npTiO2. Neonate and young individuals were more sensitive to death because of their low tolerance. This result was affected by population progeny and growth rates (p?<?.05). While control and 0.5?mgL?1 npTiO2 groups were determined as growing population, 1.5 and 2?mgL?1 npTiO2 groups had decreased population size as R0 values. Consequently, the relationships between nanoparticle accumulation within Daphnia magna and its population structure and body morphometry for each concentration were important indicators. Its tolerance level to nanoparticles under laboratory conditions reflected its replacement and behaviour in the ecosystem.  相似文献

To explore the role of endophytic fungi in the decomposition of litter, the endophytic fungi Penicillium sp. strain CG2 (A), Fusarium flavum strain AY13 (B), and Talaromyces strain AJ14 (C) of Cunninghamia lanceolata were added to experimental pots in different forms (mycelium, sterilized fermentation broth, single fungus, and mixed fungi), and a control treatment (CK) was set up (no fungi added). At 10, 30, 60, 90, and 120 days after litter decomposition, a study on the decomposition dynamics of C. lanceolata litter under different treatments was performed. The results showed that the rate of leaf mass loss was the highest in the sterilized fermentation broth treatment A after 120 days, and that there was a significant difference (P < 0.05) between the mycelium treatment AC and the control treatment after 60 days (23.97% higher than the control group). On day 60, the litter carbon content from the mycelium treatment A was significantly different from that of the control (P < 0.05), showing a 16.74% lower value, whereas the litter carbon content of the mycelium treatment B was 21.13% lower than that of the control after 90 days. The nitrogen content of the litters of most mycelium and sterilized fermentation broth treatments was increased compared to that of the control group; there was significant difference (P < 0.05) between the sterilized fermentation broth treatment A and the control (P < 0.05), with a 17.05% higher value than that of the control. Similar to nitrogen, the litter phosphorus content also increased; there was a statistically significant difference between the mycelium treatment A and the control group, with treatment A showing a 46.67% higher value than the control group. The potassium content was 28% lower than that of the control group under the sterilized fermentation broth treatment C, a result that was significantly different from that of the control group (P < 0.05). After treatment for 90 days, the ratio of carbon to nitrogen was the lowest under the treatments with the mycelium A and the mycelium B, with values 25.54% and 25.11% lower than that of the control group, respectively, and a statistically significant difference from that of the control group (P < 0.05). The ratio of carbon to phosphorus was the lowest under the treatment with mycelium A after 60 days, and the result was significantly different from that of the control (P < 0.05), with a 43.05% lower value than the control. Thus, the three endophytic fungi had different effects on the mass loss rate and nutrient content of the litter. The Penicillium sp. strain CG2 (A) had statistically significant effects on the mass loss and nutrient content of leaf litter, which was within the range of fungi fertilizer reference values for the breeding of C. lanceolata. © 2018 Science Press. To study the effect of flow velocity on drinking water distribution systems, bulk water quality was monitored over 28 days, biomass was measured, and 16S rDNA was sequenced on the 28th day using a water distribution simulation system. The relationship between bulk water quality and biofilm was statistically analyzed. Flow velocity of 0.5 m/s yielded the most total organic carbon (TOC) (5.26 ± 0.17 mg/L) in the bulk water, the most bulk water bacteria (lg (n+1/mL-1) = 4.79 ± 0.02), the worst bulk water quality, and the most biofilm bacteria (lg (n+1/cm-2) = 5.48 ± 0.06). A Pearson correlation analysis showed the total number of biofilm bacteria was positively correlated with conductivity (R = 0.73, P < 0.01), turbidity (R = 0.87, P < 0.001), TOC (R = 0.94, P < 0.001), and total bacteria (R = 0.92, P < 0.001), and was negatively correlated with residual chlorine (R = -0.68, P < 0.05). Biofilm diversity was high under the low (0.1 m/s) and high (2.5 m/s) flow rates, but the bacterial diversity of biofilm was the lowest at the 0.5 m/s flow rate, in which Proteobacteria dominated the biofilm community structure. These results suggest that flow velocity affects bulk water quality and biofilm population structure, and water quality and biofilm population structure are interrelated, which provides the theoretical basis for research on biofilms in drinking water distribution systems. © 2018 Science Press. BRI1-ASSOCIATED RECEPTORKINASE1(BAK1), a leucine-rich repeat (LRR) receptor protein kinase, plays a significant role in brassinosteroid (BR) signaling. Furthermore, it combines with other LRR-RLKs protein to initiate immune response in plants. The objective of this study was to (1) investigate the function of the Populus euphratica BAK1;1 gene in the resistance of transgenic tobacco to Pseudomonas syringae pv. tomato DC3000 (Pst DC3000) and (2) discuss the regulation pathway of PeBAK1;1 involved in the resistance to plant pathogen. We cloned the cDNA sequence of the P. euphratica PeBAK1;1 gene, constructed the pBI121-35S::PeBAK1;1 over-expression vector, and then transformed it into wild-type tobacco by Agrobacterium-mediated transformation to obtain PeBA K 1;1-overexpressing transgenic tobacco plants. The bioinformatic analysis showed that the PeBAK1;1 protein contained all the structural features of the plant SERK family. The phylogenetic tree showed that PeBAK1;1 has the highest sequence homology with PtBAK1. The gene expression profile results indicated that the expression of PeBAK1;1 in the root was higher than that in the leaf and stem. The wild-type tobacco plants showed an obvious susceptibility to Pst DC3000, whereas the transgenic plants exhibited enhanced resistance to Pst DC3000. Compared with that of the wild-type (WT), the real-time PCR and quantitative real-time PCR analysis revealed that the expression of pathogenesis-related genes (including PR1, PR3, PR4, and PR5), BAK1-interacting receptor kinase 1 gene, and BONZAI1 gene was upregulated in 35S::PeBAK1;1 transgenic tobacco plants. In conclusion, the PeBAK1;1 gene plays a positive regulatory role in 35S::PeBAK1;1 transgenic tobacco against Pst DC3000, which can enhance the resistance of plants to pathogen. © 2018 Science Press. Cytochrome P450 (CYP450s) is a type of heme-mercaptide protein superfamily, which is distributed widely in animals, plants, and microorganisms. CYP450s can oxidize and degrade many exogenous compounds such as drugs, herbicides, pesticides, some persistent organic pollutants, and so on. Based on recent researches, this paper reviews the nomenclature, classification, structure, and catalytic mechanism of P450 enzymes, and summarizes the research progresses in the metabolism and biodegradation of xenobiotics using P450 enzymes from microorganisms. The nomenclature and classification of the P450 gene superfamily mainly rely on the similarities of amino acid sequences. Although the structures of P450 are conserved, their recognition sites towards to the substrates are variable. This is also the structural basis for the catalytic diversities of P450 enzymes. Few P450 enzymes from bacteria and fungi can metabolize and degrade xenobiotics such as polycyclic aromatic hydrocarbons, herbicides, and so on. However, these P450 enzymes are less likely be used in practical applications because of their low catalytic activities. In the future, more P450 enzymes with high degradable efficiencies towards xenobiotics are needed to be obtained using multiple omics tools or modifying the existing P450 enzymes, to achieve the bioremediation of the environment. © 2018 Science Press.
