An evaluation of the applicability of microarrays for monitoring toxic algae in Irish coastal waters

The applicability of microarrays to monitor harmful algae across a broad range of ecological niches and toxic species responsible for harmful algal events has been one of the key tasks in the EU Seventh Framework Programme (FP7)-funded Microarrays for the Detection of Toxic Algae project. The technique has a strong potential for improving speed and accuracy of the identification of harmful algae and their toxins to assist monitoring programmes. Water samples were collected from a number of coastal sites around Ireland, including several that are used in the Irish National Phytoplankton and Biotoxin Monitoring Programme. Ribosomal RNA was extracted from filtered field samples, labelled with a fluorescent dye, and hybridised to probes spotted in a microarray format on a glass slide. The fluorescent signal intensity of the hybridisation to >120 probes on the chip was analysed and compared with actual field counts. There was a general agreement between cell counts and microarray signal. Results are presented for field samples taken from a range of stations along the Irish coastline known for harmful algal events during the first field trial (July 2009–April 2010). 相似文献

The body of information presented in this paper is directed to researchers in stack testing methodology and to those concerned with reduction of emissions through equipment upgrading programs. Extensive testing was done using the U.S. Environmental Protection Agency’s Method 5 stack sampling train to obtain emission factors for existing apartment house boilers and incinerators in the City of New York. In addition to calculating emission factors, stack emission data were examined to compare results of simultaneous emission tests and to compare the dry particulate catch of the sampling train with the total particulate catch which included the impinger catch. Conclusions reached as a result of the testing were that published emission factors for boilers burning moderately high-sulfur residual oil are applicable to New York City boilers burning low-sulfur residual oil. In addition, it was found that the back half of the sampling train—the impinger section—collects a relatively constant amount of material when sampling oil-fired boilers. This may be due to absorption of S02 and S03 in the impingers and the subsequent formation of sulfuric acid. Comparison of simultaneous boiler tests indicated that the sampling train may be sensitive to variations in operating personnel, sampling conditions, and boiler operation. From tests of on-site incinerators, it was determined that previously published emission factors may be too high for well maintained and properly operated incinerators. Anthropogenic increase in atmospheric nitrogen (N) deposition in nature areas results in nitrate leaching to groundwater, threatening its quality. Member states of the European Union are obliged to reduce groundwater nitrate concentrations and to monitor this reduction. The relationship between N deposition and groundwater nitrate concentrations is quantified using a field survey and geographical information. Nitrate concentrations of the uppermost metre of groundwater in nature areas in the sandy regions in 1990 were related to geographical data by means of regression analysis. In this way nitrate concentrations could be explained by potential ammonia deposition, soil type, vegetation and land use. We found that about 35% of 54 kg ha(-1) a(-1) atmospheric N deposition was leached to the upper groundwater as nitrate, resulting in a mean NO3 concentration of about 30 mg L(-1). The critical N load for exceeding the EC limit value (50 mg L(-1)) in the sandy regions of The Netherlands composed of natural vegetation will be about 80 kg ha(-1) a(-1). Leaching is less than expected for nature areas but comparable with leaching of N surpluses in pastures in The Netherlands. Atmospheric CO2 concentration (Ca) is rising, predicted to cause global warming, and alter precipitation patterns. During 1994, spring barley (Hordeum vulgare L. cv. Alexis) was grown in a strip-split-plot experimental design to determine the effects that the main plot Ca treatments [A: Ambient at 370 μmol (CO2) mol−1; E: Enriched with free-air CO2 enrichment (FACE) at ∼550 μmol (CO2) mol−1] had on several gas exchange properties of fully expanded sunlit primary leaves. The interacting strip-split-plot irrigation treatments were Dry or Wet [50% (D) or 100% (W) replacement of potential evapotranspiration] at ample nitrogen (261 kg N ha−1) and phosphorous (29 kg P ha−1) fertility. Elevated Ca facilitated drought avoidance by reducing stomatal conductance (gs) by 34% that conserved water and enabled stomata to remain open for a longer period into a drought. This resulted in a 28% reduction in drought-induced midafternoon depression in net assimilation rate (A). Elevated Ca increased A by 37% under Dry and 23% under Wet. Any reduction in A under Wet conditions occurred because of nonstomatal limitations, whereas under Dry it occurred because of stomatal limitations. Elevated Ca increased the diurnal integral of A (A′) that resulted in an increase in the seasonal-long integral of A′ (A″) for barley leaves by 12% (P = 0.14) under both Dry and Wet - 650, 730, 905 and 1020 ± 65 g (C) m−2 y−1 for AD, ED, AW and EW treatments, respectively. Elevated Ca increased season-long average dry weight (DWS; crown, shoots) by 14% (P = 0.02), whereas deficit irrigation reduced DWS by 7% (P = 0.06), although these values may have been affected by a short but severe pea aphid [Acyrthosiphon pisum (Harris)] infestation. Hence, an elevated-Ca-based improvement in gas exchange properties enhanced growth of a barley crop. 相似文献
