A rapid and simple analytical method for the determination of organochlorines, such as polychlorinated biphenyls (PCBs) and selected organochlorinated pesticides (OCPs) and organobromines, such as polybrominated diphenyl ethers (PBDEs), in sediment samples was optimised using CRM 536 (PCBs in freshwater sediment). The method involved a hot Soxhlet extraction that reduced the extraction time to 2 h. Elemental sulphur, which is present in sediments and may interfere during the analysis, was removed by means of copper powder added to the sediment during extraction and into the clean-up cartridge. The analysis of PCBs and OCPs was accomplished by gas chromatography with electron capture or mass spectrometric detection. Similar quantitative results for PCB congeners in CRM 536 were obtained using a 50-m capillary column and a 10-m narrow bore column suited for fast analysis. The analysis of PBDEs was done by mass spectrometry in negative chemical ionisation mode. Concentrations of organic pollutants in two sediment cores (approximately 50 cm depth) from the Scheldt river (south of Antwerp, Belgium) showed a relative steady state for PCBs and DDTs, with a slight decrease in the top layers, suggesting a slight decline in their concentrations due to restrictions in their usage. On the contrary, PBDEs were showing an increase in their concentrations in the top layers (up to 270 and 8400 ng/g dry weight for sum of tri- to hexa-BDE congeners and for BDE 209, respectively). This suggests an increasing trend in the concentrations of PBDEs in the Belgian environment.  相似文献

The D2-protein is a neuronal membrane protein which has an immunochemically detectable soluble derivative in cerebrospinal fluid. We used rocket-on-line immunoelectrophoresis with an antiserum against rat synaptosomal membranes for demonstration of the human D2-protein in fetal cerebrospinal fluid and in amniotic fluids from pregnancies with fetal neural tube defects. D2 was neither found in normal amniotic fluids nor in cases of amniotic fluids from pregnancies with other malformations. Analysis of D2 in amniotic fluids may thus have a future role in the antenatal diagnosis of fetal neural tube defects.  相似文献

There is a great need for understanding the impact from dietary OHCs (organohalogen compounds) on bone mineral composition - and thereby osteoporosis - in especially arctic wildlife such as polar bears (Ursus maritimus) as well as humans. For that purpose, we measured BMD (bone mineral density) by DXA scanning (g/cm(-2)) in 15 age and weight normalized sledge dog (Canis familiaris) bitches and their 26 pups divided into a control group (n=26) given 50-200 g/day clean pork (Suis scrofa) fat and a treated group (n=15) given 50-200 g/day OHC polluted minke whale (Balaenoptera acutorostrata) blubber as main lipid sources. The results showed that BMD increased significantly with age (linear regression: p<0.0001, r(2)=0.83, n=41) while no sex difference was found in the F-generation (two-way ANOVA: all p>0.3). No differences in BMD(femur) or BMD(vertebrae) between exposed and control individuals in the bitch generation were found (linear mixed effect model: both p>0.38). Likewise, no difference between exposed and control subadults and juveniles in the F-generation was found (two-way ANOVA: all p>0.33). Correlation analyses between BMD(femur), BMD(vertebrae) and groups of OHCs, respectively, did not show any statistically significant relationships nor a clear or decreasing trend (Pearson's: p: 0.07-0.78; r: -0.2-0.59; n: 10-18). As the groups were similar regarding genetics, age and sex are the only factors that can explain this observation. Either the pollutants did not have an impact on BMD using the present time frame and OHC concentrations (threshold levels not reached), or the difference in food composition (mainly vitamins and n3 fatty acids) conceal the potential OHC impact on BMD. Such information is important when evaluating the positive and negative health consequences from eating polluted marine species.  相似文献

Summary Female threespine sticklebacks (Gasterosteus aculeatus) frequently raid male nests and eat all the eggs therein. We tested the hypothesis of Vickery et al. (1988) that females prefer to raid nests containing large numbers of eggs than ones with smaller numbers of eggs. This hypothesis is based on the finding that females spawning in nests containing many eggs will have reduced hatching success because of egg crowding. By consuming the male's eggs and forcing him to rebuild his nest, raiding females might obtain a new opportunity to spawn under better conditions. Our results were consistent with the first prediction of this hypothesis that females were more likely to spawn in nests containing fewer eggs than in nests with many eggs. However, this may be the result of males becoming less receptive to females as the number of eggs in their nests increases. Prediction 2 was that females should raid those nests containing the most eggs. Contrary to this prediction, males defending only one clutch were as likely to have their nests raided by groups of females as males defending several clutches of eggs. Female cannibalism is therefore unlikely to have evolved as a means of gaining access to a male defending a small number of eggs. We also examined the tactics used by males to counter female raids. Most raids occur when the male is courting, and nests are more vulnerable to shoals of females than to single females. Therefore, we hypothesized that males with eggs preferentially court a single female rather than large groups of females, and that males without eggs court both groups indiscriminately. We also predicted that males restrict the number of females they mate with when risk of having their nest raided is high. Our results indicate that: (1) both males with eggs and those without eggs minimize the risk of female cannibalism by courting solitary females rather than groups of females and (2) males limit the number of females that lay eggs in their nest when several potentially raiding females are present. The present study was designed to evaluate the effect of a natural prey (the crab Callinectes sp.) and an artificial diet (pellet with squid paste and offered as a paste) on the survival and assimilation efficiency of subadult octopuses with 486 g of initial live weight. In order to reach this goal, the effects of the type of diet on energetic balance were assessed by recording ingestion rate (C), respiratory rate (R = R routine, R rout + R apparent heat increment, R AHI), ammonia production rate (U = U routine, U rout + U post-prandial, U PP) and biomass production (P) of Octopus maya during its growing process. Energy lost from faeces (H) was calculated as H=C−(U+R+P) and assimilated energy (As) as R + P. Octopuses fed an artificial diet had almost five times higher ingestion rate compared to that observed in octopuses fed crab. However, growth rate and production (P) were high in octopuses fed crab in comparison to octopuses fed artificial diet. An inverse relation between faeces (H) and type of food was observed, indicating that animals lost 77% of the ingested energy when fed artificial diet and only 5% when fed crab. A higher assimilation and production efficiency were obtained in octopuses fed crab (P/As: 61%) than in animals fed the artificial diet (P/As: −5%). The routine O : N ratio for animals in fasting was 9.1 and 2.3 for octopuses being fed crabs and the artificial diet, respectively. The post-alimentary O : N ratio was 3.6 and 2.2 for animals fed crabs and the artificial diet, respectively. This indicates that animals fed on both diets rely almost exclusively on protein. Based on energy balance data, a value of 472 kJ week−1 kg−1 of live octopus was estimated as the energy needed to obtain a growth rate near 9 g day−1 (2.8% BW day−1) for O. maya subadults. The total crab biomass needed to obtain 1 kg of fed O. maya biomass was calculated. A comparison with other different energy balance measurements made in other octopus species indicates that O. maya and Enteroctopus megalocyathus (Pérez et al. 2006) tend to be more efficient by channelling more ingested energy to biomass production (P = 69.5% of C) than O. vulgaris (P = 23% of C; Petza et al. 2006) or Paraledone charcoti (P = 4% of C; Daly and Peck 2000).  相似文献

The locus of symbiotic dinoflagellates within host cells provides a habitat which could potentially be exploited by the alga through heterotrophic uptake of host-derived organic substrates. Using zooxanthellae (Symbiodinium sp.) isolated from the tropical sea anemone Aiptasia pulchella collected from Kaneohe Bay, Hawaii, the effect of various potential organic substrates on growth in vitro was assessed in Erdschreiber seawater medium (ES) supplemented with organic compounds. Zooxanthellae maintained at 5 to 7 E m-2 s-1 (below compensation irradiance) grew heterotrophically when supplied with 100 M glycerol, glycolate, acetate, malate, or propionate, and grew in darkness on 100 M propionate. Zooxanthellae exposed to irradiance below compensation were able to utilize carbon sources in the unsupplemented ES medium for slow growth, but generally the growth rate of cultured zooxanthellae was a function of incubation irradiance. Zooxanthellae incubated for 10 wk in unsupplemented ES at 5 to 7 E m-2 s-1 were capable of growth at this low irradiance, but were also capable of net photosynthetic oxygen production at higher irradiances. This suggests that zooxanthellae can be photoautotrophic or facultatively heterotrophic. An estimate for the duration of mitosis (t d ) is made on the basis of growth rate of cultured zooxanthellae in log-phase; this estimate of t d =4.88 h is less than half the estimated t d for zooxanthellae in situ.  相似文献

High-frequency fluctuations due to wavefocusing are prominent characteristics of light in shallow marine environments. Effects of high-frequency (0.01 to 1 Hz) fluctuating light on growth rates of the red algaChondrus crispus Stackh., collected from Crane Neck Point, New York, USA, during July 1988, were determined by comparing plants grown under fluctuating and constant light regimes with similar daily irradiances. At high daily irradiance, growth rates were higher under fluctuating light than under constant light. Fluctuating light effects were frequency-dependent; growth was enhanced by fluctuations at 0.1 and 1 Hz, but not by fluctuations at 0.01 Hz. At low daily irradiance, growth rates were not affected by fluctuating light at any frequency tested. Enhancement of growth was not due to effects of high-frequency light fluctuations on photoacclimation responses ofC. crispus. Plants grown under fluctuating light at high daily irradiance actually exhibited lower photosynthetic capacity and efficiency (determined under constant light) than plants grown under constant light. These differences were attributable to variation in the density of Photosystem II reaction centers, which was low in plants grown under fluctuating light. Maximum turnover rate of whole-chain electron transport and activity of ribulose-1,5-bisphosphate carboxylase were affected by total daily irradiance, but not by high-frequency light fluctuations. Enhancement of growth under fluctuating light was partly attributable to reduced rates of dark respiration compared to rates of plants grown under constant light. The results also provided indirect evidence that high-frequency light fluctuations may enhance instantaneous photosynthetic rates. This effect could increase daily carbon gain and, therefore, stimulate growth ofC. crispus under high-frequency light fluctuations.  相似文献

Isozyme, randomly amplified polymorphic DNA, and ribosomal DNA analyses were used to evaluate the genetic distinctness and diversity of the endangered island endemic Malacothamnus fasciculatus var. nesioticus (Santa Cruz Island bush mallow). Analysis of amplified DNA profiles and electrophoretic data indicates that var. nesioticus is genetically distinct from other varieties within the species. No isozyme or amplified DNA variation was detected among individuals of nesioticus populations, but restriction site analysis of ribosomal DNA revealed the presence of three closely related genotypes within one of the nesioticus populations. Apparent clonal structuring was found in both nesioticus populations and in one mainland population, var. nuttallli . Results from these analyses provide the background necessary for the design of conservation efforts with recovery goals that insure the long-term survival of Malacothamnus fasciculatus var. nesioticus .  相似文献

As part of the Fluxes of Agrochemicals into the Marine Environment (FAME) project, the gross fluxes of selected pesticides (i.e. the herbicides atrazine, simazine, alachlor and metolachlor, the atrazine degradation product desethylatrazine, the insecticide dichlorvos and the antifouling agent Irgarol 1051) transported by the river Scheldt and the Canal Ghent-Terneuzen were determined from March 1995 through February 1997. In general, the observed temporal trends were related to the application period of the pesticides, except for metolachlor for which elevated concentrations were observed in the winter of 1995-1996. Relatively large gross fluxes were found for desethylatrazine compared with its parent compound. A study on the estuarine behaviour of pesticides showed distinct differences between the compound classes. The mixing plots of the organophosphorus insecticides dichlorvos and diazinon revealed clear evidence of estuarine loss processes which agrees with their low DT50 values reported for water/sediment systems, their relatively high Henry's law constants and, for diazinon, its relatively high Koc value. The mixing plots of the acetanilides alachlor and metolachlor were strongly influenced by an additional direct emission into the estuary, which was evident from a maximum in dissolved concentration near a salinity of 10@1000. An apparent conservative behaviour was observed for the triazine compounds atrazine and Irgarol 1051. This was in contrast to simazine, which showed an apparent non-conservative behaviour. However, the time profiles of the riverine concentrations of simazine did not exclude that the observed curvature was solely caused by estuarine losses; therefore, additional modelling is required. In a follow-up study a suitable hydrological model of the Scheldt estuary was constructed; the results will be presented in a forthcoming paper (Steen, R.J.C.A., Evers, E.H.G., Van Hattum, B., Cofino, W.P. and Brinkman, U.A.Th.
