The effect of Pb + Zn on coelomocyte riboflavin content in the epigeic earthworm Dendrodrilus rubidus inhabiting three metalliferous soils and one reference soil was measured by flow cytometry and spectrofluorimetry. A reciprocal polluted↔unpolluted worm transfer experiment (4-week exposure) was also performed. High proportions of autofluorescent eleocytes were counted in worms from all localities, but intense riboflavin-derived autofluorescence was detectable only in reference worm eleocytes. Other findings were: (i) fluorophore(s) other than riboflavin is/are responsible for eleocyte autofluorescence in residents of metalliferous soils; (ii) riboflavin content was reduced in the eleocytes of worms transferred from unpolluted to metal-polluted soil; (iii) the riboflavin content of D. rubidus eleocytes is a promising biomarker of exposure; (iv) COII mitochondrial genotyping revealed that the reference population is genetically distinct from the three mine populations; (v) metal exposure rather than genotype is probably the main determinant of inter-population differences in eleocyte riboflavin status. 相似文献
Species distribution models (SDMs) are increasingly used in conservation and land-use planning as inputs to describe biodiversity patterns. These models can be built in different ways, and decisions about data preparation, selection of predictor variables, model fitting, and evaluation all alter the resulting predictions. Commonly, the true distribution of species is unknown and independent data to verify which SDM variant to choose are lacking. Such model uncertainty is of concern to planners. We analyzed how 11 routine decisions about model complexity, predictors, bias treatment, and setting thresholds for predicted values altered conservation priority patterns across 25 species. Models were created with MaxEnt and run through Zonation to determine the priority rank of sites. Although all SDM variants performed well (area under the curve >0.7), they produced spatially different predictions for species and different conservation priority solutions. Priorities were most strongly altered by decisions to not address bias or to apply binary thresholds to predicted values; on average 40% and 35%, respectively, of all grid cells received an opposite priority ranking. Forcing high model complexity altered conservation solutions less than forcing simplicity (14% and 24% of cells with opposite rank values, respectively). Use of fewer species records to build models or choosing alternative bias treatments had intermediate effects (25% and 23%, respectively). Depending on modeling choices, priority areas overlapped as little as 10–20% with the baseline solution, affecting top and bottom priorities differently. Our results demonstrate the extent of model-based uncertainty and quantify the relative impacts of SDM building decisions. When it is uncertain what the best SDM approach and conservation plan is, solving uncertainty or considering alterative options is most important for those decisions that change plans the most. 相似文献
To assess the effects of exposure to complex mixtures of pesticides in farm workers from two communities from Rio Grande do Sul, Brazil, we evaluated the activities of butyrylcholinesterase (BChE) and delta-aminolevulinic acid dehydratase (ALA-D) enzymes, hematological, lipid parameters, and genotoxicity using two tests to detect DNA damage, the Comet assay in peripheral blood leukocytes and the micronucleus (MN) test in oral mucosa cells. The use of personal protective equipment (PPE), age and smoke habits were considered in the analysis. There was a significant decrease in the BChE and ALA-D activities in farm workers (n=37) relative to the control group (n=20) (P< or =0.05 and P< or =0.001, respectively). The Comet assay in peripheral blood leukocytes showed that the Damage index and Damage frequency observed in the exposed group were significantly higher in relation to the controls (P< or =0.001, and P< or =0.05, respectively). No differences were detected regarding the hematological parameters, lipids profile, and MN frequencies. In addition, no significant differences were observed between younger (< or =38 years) and older subjects (>38 years), or between smokers and non-smokers within the groups, either by Comet assay or MN test. However, the use of PPE seems to be important in the prevention of contamination, as suggested by BChE levels and Comet assay results. 相似文献

Some imidazolinone herbicides have been shown to be mobile in soil, raising concern about their possible movement to ground water. Three imidazolinone herbicides (imazamethabenz-methyl, 497 g ha(-1); imazethapyr, 14.7 g ha(-1); and imazamox, 14.7 g ha(-1)) commonly used in crop production on the Canadian prairies were applied to a tile-drained field to assess their susceptibility to leach when subjected to sprinkler irrigation using a center pivot. Tile-drain flow began when the water table rose above tile-drain depth, and peak flow rates corresponded to the greatest depths of ground water above the tile drains. Interception of irrigation water by the tile drains in each quadrant of the field varied from ~11 to 20% of the water applied. Under a worst-case scenario in which irrigation began the day after herbicide application and irrigation water was applied at 25 mm d(-1) for 12 d, there was evidence of preferential flow of all three herbicides and hydrolysis of imazamethabenz-methyl to imazamethabenz in the initial samples of tile-drain effluent. In subsequent samples, concentrations (analysis by LC-MS-MS) of the summation of imazamethabenz-methyl (25-24,000 ng L(-1)) plus its hydrolysis product imazamethabenz (63-26,500 ng L(-1)) greatly exceeded those of imazethapyr (<13-1260 ng L) and imazamox (19-599 ng L(-1)), thus reflecting relative application rates. In contrast, estimates of total transport of each herbicide from the root zone, which varied in each quadrant and ranged from 0.06 to 2.3% for imazamethabenz-methyl plus imazamethabenz, 0.71 to 3.1% for imazethapyr, and 0.61 to 2.8% for imazamox, did not reflect application rates. In shallow ground water (piezometer samples), there was inconsistent and infrequent detection all four compounds. With the frequency and amount of rainfall typically encountered in the prairie region of Canada, contamination of shallow ground water with detectable concentrations of the three imidazolinone herbicides would be unlikely. 相似文献

The reasons why a study of public attitudes can be an important factor in planning an air pollution control program are briefly summarized. Acetaminophen (ACT) is one of the most frequently detected pharmaceuticals in aqueous environments, and treatment of ACT were generally carried out by photocatalytic degradations under high energy UV irradiation. In this study, potassium ferricyanide was utilized as a quadruple-elemental dopant in a TiO2 photocatalyst in order to enhance its visible-light activity. Two critical parameters (amounts of dopants and durations of calcination) of the synthesis of the photocatalyst by a sol–gel method were systematically evaluated. Crystal structure of the doping TiO2 was examined by X-ray diffraction while the effects of the two parameters on the photocatalytic activity were elucidated by various characterizations. Increasing the amount of dopant or the duration of calcination red-shifted the UV–vis DRS of the doped TiO2. The estimated band gap energy of the doped TiO2 decreased slightly as the amount of dopant increased, but it increased as the duration of calcination increased. The FT-IR yielded characteristic peaks that revealed the effects of the two parameters, whereas the SEM images revealed the morphological evolutions of each effect. The photocatalyst, synthesized at optimum conditions was able to remove 99.1 % acetaminophen with rate constant of 7.9 × 10−3 min−1, which was 4.88 times greater than virgin TiO2. In general, this study not only optimized synthetic conditions of the new visible-light active photocatalyst for ACT degradation but also presented characterizations conducted by SEM, XRD, UV–vis DRS, and FTIR to elucidate the relationship between modified structure and the photocatalytic activity.
Effects of doping amounts of K3[Fe(CN)6] and calcunation duration on visible light absorbance of TiO2 photocatalysts
An analytical method was developed for determining organophosphate pesticides (OPP) and pyrethroid pesticides (PYR) in duplicate-diet solid food. The method consisted of pressurized liquid extraction (PLE) with dichloromethane followed by cleanup with gel permeation and solid phase extraction columns and gas chromatography/mass spectrometry (GC/MS) analysis. Quantitative recoveries (73–117 %) of the target pesticides were obtained for spiked duplicate-diet food samples. The percent standard deviation (% RSD) of replicate food samples was within ± 20 %. Another method was developed for determining a common OPP metabolite, 3, 5, 6-trichloro-2-pyridinol (TCP) in duplicate-diet food. The method consisted of a PLE with methanol followed by liquid-liquid partitioning, derivatization, and GC/MS analysis. Recoveries of TCP ranged from 83 to 101 % for spiked duplicate-diet food samples. The % RSD of replicate food samples was within ± 15 %. The results confirmed that these methods are reliable and robust, and that they can be used in routine analysis. In addition, a storage stability study for a common OPP, chlorpyrifos (CPF), in solid food samples was performed. The fortified 15N-13C-labeled CPF was stable over 16 mo storage at ?20° C in the dark. The developed analytical methods were successfully applied to 278 duplicate-diet food samples from preschool children, demonstrating that these methods are robust and suitable for routine analysis in future exposure monitoring studies. 相似文献
Harmful or nuisance algal blooms can cause economic damage to fisheries and tourism. Additionally, toxins produced by harmful algae and ingested via contaminated shellfish can be potentially fatal to humans. The seas around the Orkney Islands, UK currently hold a number of toxic algal species which cause shellfishery closures in most years. Extensive and costly monitoring programs are carried out to detect harmful microalgae before they reach action levels. However, the ability to distinguish between toxic and non-toxic strains of some algae is not possible using these methods. The microarrays for the detection of toxic algae (MIDTAL) microarray contains rRNA probes for toxic algal species/strains which have been adapted and optimized for microarray use. In order to investigate the use of the chip for monitoring in the Orkney Islands, samples were collected between 2009 and 2011 from Brings Deep, Scapa Flow, Orkney Islands, UK; RNA was extracted and hybridized with generation 2 and 3.1 of the chip. The data were then compared to cell counts performed under light microscopy and in the case of Alexandrium tamarense to qPCR data targeting the saxitoxin gene and the LSU-rRNA gene. A good agreement between cell numbers and microarray signal was found for A. tamarense, Pseudo-nitzschia sp., Dinophysis sp. (r?<?0.5, for all) in addition to this there the chip successfully detected a large bloom of Karenia mikimotoi (r?<?0.70) in August and September 2011. Overall, there was good improvement in probe signal between generation 2 and generation 3.1 of the chip with much less variability and more consistent results and better correlation between the probes. The chip performed well for A. tamarense group I signal to cell numbers in calibrations (r?>?0.9). However, in field samples, this correlation was slightly lower suggesting interactions between all species in the sample may affect signal. Overall, the chip showed it could identify the presence of target species in field samples although some work is needed to improve the quantitative nature of the chip before it would be suitable for monitoring in the Orkney Islands. 相似文献
