Background, Aim and Scope
Numerous herbicides and xenobiotic organic pollutants are detoxified in plants to glutathione conjugates. Following this enzyme
catalyzed reaction, xenobiotic GS-conjugates are thought to be compartmentalized in the vacuole of plant cells. In the present
study, evidence is presented for long range transport of these conjugates in plants, rather than storage in the vacuole. To
our knowledge this is the first report about the unidirectional long range transport of xenobiotic conjugates in plants and
the exudation of a glutathione conjugate from the root tips. This could mean that plants possess an excretion system for unwanted
compounds which give them similar advantages as animals.
Materials and Methods:
Barley plants (Hordeum vulgare L. cv. Cherie) were grown in Petri dishes soaked with tap water in the greenhouse.
- Fluorescence Microscopy. Monobromo- and Monochlorobimane, two model xenobiotics that are conjugated rapidly in plant cells
with glutathione, hereby forming fluorescent metabolites, were used as markers for our experiments. Their transport in the
root could be followed sensitively with very good temporal and spatial resolution. Roots of barley seedlings were cut under
water and the end at which xenobiotics were applied was fixed in an aperture with a thin latex foil and transferred into a
drop of water on a cover slide. The cover slide was fixed in a measuring chamber on the stage of an inverse fluorescence microscope
(Zeiss Axiovert 100).
- Spectrometric enzyme assay. Glutathione S-transferase (GST) activity was determined in the protein extracts following established
methods. Aliquots of the enzyme extract were incubated with 1-chloro-2,4-dinitrobenzene (CDNB), or monochlorobimane. Controls
lacking enzyme or GSH were measured.
- Pitman chamber experiments. Ten days old barley plants or detached roots were inserted into special incubation chambers, either
complete with tips or decapitated, as well as 10 days old barley plants without root tips. Compartment A was filled with a
transport medium and GSH conjugate or L-cysteine conjugate. Compartments B and C contained sugar free media. Samples were
taken from the root tip containing compartment C and the amount of conjugate transported was determined spectro-photometrically.
Results:
The transport in roots is unidirectional towards the root tips and leads to exsudation of the conjugates at rates between
20 and 200 nmol min-1. The microscopic studies have been complemented by transport studies in small root chambers and spectroscopic
quantification of dinitrobenzene-conjugates. The latter experiments confirm the microscopic studies. Furthermore it was shown
that glutathione conjugates are transported at higher rates than cysteine conjugates, despite of their higher molecular weights.
This observation points to the existence of glutathione specific carriers and a specific role of glutathione in the root.
Discussion:
It can be assumed that long distance transport of glutathione conjugates within the plant proceeds like GSH or amino acid
transport in both, phloem and xylem. The high velocity of this translocation of the GS-X is indicative of an active transport.
For free glutathione, a rapid transport-system is essential because an accumulation of GSH in the root tip inhibits further
uptake of sulfur. Taking into account that all described MRP transporters and also the GSH plasmalemma ATPases have side activities
for glutathione derivatives and conjugates, co-transport of these xenobiotic metabolites seems credible.
- On the other hand, when GS-B was applied to the root tips from the outside, no significant uptake was observed. Thus it can
be concluded that only those conjugates can be transported in the xylem which are formed inside the root apex. Having left
the root once, there seems to be no return into the root vessels, probably because of a lack of inward directed transporters.
Conclusions:
Plants seem to possess the capability to store glutathione conjugates in the vacuole, but under certain conditions, these
metabolites might also undergo long range transport, predominantly into the plant root. The transport seems dependent on specific
carriers and is unidirectional, this means that xenobiotic conjugates from the rhizosphere are not taken up again. The exudation
of xenobiotic metabolites offers an opportunity to avoid the accumulation of such compounds in the plant.
Recommendations and Perspectives:
The role of glutathione and glutathione related metabolites in the rhizosphere has not been studied in any detail, and only
scattered data are available on interactions between the plant root and rhizosphere bacteria that encounter such conjugates.
The final fate of these compounds in the root zone has also not been addressed so far. It will be interesting to study effects
of the exuded metabolites on the biology of rhizosphere bacteria and fungi. 相似文献
In many species reproduction and embryonic development have been shown to be sensitive to environmental contaminants. Understanding embryonic exposure to environmental contaminants is thus highly important. In this study concentrations of brominated flame retardants (BFRs) were measured in zebrafish eggs after parental exposure for 42 days via the diet. Zebrafish were exposed to two doses of eleven structurally-diverse BFRs. Eight BFRs were detected in the female zebrafish and maternal transfer to eggs was evident for all eight compounds. The highest concentrations in eggs were observed for hexabromocyclododecane (HBCD) and 2,4,4'-tribromodiphenyl ether (BDE 28), followed by 2,2',3,4,4',5',6-heptabromodiphenyl ether (BDE 183) and tetrabromobisphenol A 2,3-dibromopropyl ether (TBBPA DBPE). Five potential BFR metabolites were tentatively identified in female fish and maternal transfer was observed also for these compounds. The lipid adjusted concentrations in eggs were significantly higher than the concentrations in female fish for several of the BFRs. Further, the results showed a generally higher transfer in the lower exposure level and also indicated a dependency on the physico-chemical properties of the compounds. 相似文献
Hexabromocyclododecanes (HBCDs) are high production volume chemicals (>20000 ty(-1)) used as flame retardants for plastics and textiles. Lately, we reported on the stereoselective isomerization of beta-HBCDs. Herein we present insights into the mechanism and kinetics of (+)gamma- to (+)alpha- and of (-)gamma- to (-)alpha-HBCD isomerization. Only two of the six bromine atoms migrated, indicating that rearrangements of gamma- to alpha-HBCDs are regio- and stereoselective as well. The apparent first-order isomerization rate constants increased from 0.0013 to 0.0031 to 0.0070 min(-1) at 120, 130, and 140 degrees C, respectively, corresponding to half-lives of 540, 230, and 99 min. Thus, a thermal treatment of materials containing gamma-HBCDs at temperatures >100 degrees C may induce the formation of alpha-HBCDs and, hence, may alter the diastereomeric ratio of a HBCD mixture. The inversion of vicinal dibromides in like-configurations (RR/SS) prevailed, whereas unlike-configurations (RS/SR) were not affected. An intramolecular, stereoselective migration of neighboring bromine atoms via a four-center transition state would explain the observed stereoisomer pattern and first-order kinetics. Despite the fact that vicinal dibromides in HBCDs prefer synclinal (gauche) conformations, antiperiplanar (staggered) conformations are assumed to facilitate concerted 1.2-shifts of both bromine atoms. A conformation analysis revealed that under kinetic control, only those bromine atoms in the more flexible part of the molecules are migrating, whereas those in the conserved triple-turn motive were not affected. Thus, this structural motive, common to all alpha-, beta-, and gamma-HBCDs, is more rigid and less reactive than the flexible part, containing the reacting dibromides in like-configurations. 相似文献

The Three-North Shelter Forest Program is the largest afforestation reconstruction project in the world. Remote sensing is a crucial tool to map land use and land cover change, but it is still challenging to accurately quantify the change in forest extent from time-series satellite images. In this paper, 30 Landsat MSS/TM/ETM+ epochs from 1974 to 2012 were collected, and the high-quality ground surface reflectance (GSR) time-series images were processed by integrating the 6S atmosphere transfer model and a relative reflectance normalization algorithm. Subsequently, we developed a vegetation change tracking method to reconstruct the forest change history (afforestation and deforestation) from the time-series Landsat GSR images based on the integrated forest z-score (IFZ) model by Huang et al. (2009a), which was improved by multi-phenological IFZ models and the smoothing processing of IFZ data for afforestation mapping. The mapping result showed a large increase in the extent of forest, from 380,394 ha (14.8 % of total district area) in 1974 to 1,128,380 ha (43.9 %) in 2010. Finally, the land cover and forest change map was validated with an overall accuracy of 89.1 % and a kappa coefficient of 0.858. The forest change time was also successfully retrieved, with 22.2 % and 86.5 % of the change pixels attributed to the correct epoch and within three epochs, respectively. The results confirmed a great achievement of the ecological revegetation projects in Yulin district over the last 40 years and also illustrated the potential of the time-series of Landsat images for detecting forest changes and estimating tree age for the artificial forest in a semi-arid zone strongly influenced by human activities. 相似文献
This study aimed to compare different methods to analyse the contribution of individual river characteristics to predict the
abundance of Asellus (Crustacea, Isopoda). Six methods which provide the relative contribution and/or the contribution profile of the input variables
of artificial neural network models were therefore compared: (1) the ‘partial derivatives’ method; (2) the ‘weights’ method;
(3) the ‘perturb’ method; (4) the ‘profile’ method; (5) the ‘classical stepwise’ method; (6) the ‘improved stepwise’ method.
Consequently, the key variables which affect the habitat preferences of Asellus could be identified. To evaluate the performance of the artificial neural network model, the model predictions were compared
with the results of a multiple linear regression analysis. The dataset consisted of 179 samples, collected over a 3-year period
in the Zwalm catchment in Flanders, Belgium. Twenty-four environmental variables as well as the log-transformed abundance
of Asellus were used in this study. The different contribution methods seemed to give similar results concerning the order of importance
of the input variables. Nevertheless, their diverse computation led to differences in sensitivity and stability of the methods
and the derived outcomes on the habitat preferences. From an ecological point of view, the environmental variables describing
the stream type (width, depth, stream order and distance to mouth) were the most significant variables for Asellus in the Zwalm catchment during the period 2000–2002 for all applied methods. Indirectly, one can conclude that the water quality
is not the limiting factor for the survival of Asellus in the Zwalm catchment. 相似文献
Nine dust storms in south-central Arizona were simulated with the Weather Research and Forecasting with Chemistry model (WRF-Chem) at 2 km resolution. The windblown dust emission algorithm was the Air Force Weather Agency model. In comparison with ground-based PM10 observations, the model unevenly reproduces the dust-storm events. The model adequately estimates the location and timing of the events, but it is unable to precisely replicate the magnitude and timing of the elevated hourly concentrations of particles 10 µm and smaller ([PM10]).Furthermore, the model underestimated [PM10] in highly agricultural Pinal County because it underestimated surface wind speeds and because the model’s erodible fractions of the land surface data were too coarse to effectively resolve the active and abandoned agricultural lands. In contrast, the model overestimated [PM10] in western Arizona along the Colorado River because it generated daytime sea breezes (from the nearby Gulf of California) for which the surface-layer speeds were too strong. In Phoenix, AZ, the model’s performance depended on the event, with both under- and overestimations partly due to incorrect representation of urban features. Sensitivity tests indicate that [PM10] highly relies on meteorological forcing. Increasing the fraction of erodible surfaces in the Pinal County agricultural areas improved the simulation of [PM10] in that region. Both 24-hr and 1-hr measured [PM10] were, for the most part, and especially in Pinal County, extremely elevated, with the former exceeding the health standard by as much as 10-fold and the latter exceeding health-based guidelines by as much as 70-fold. Monsoonal thunderstorms not only produce elevated [PM10], but also cause urban flash floods and disrupt water resource deliveries. Given the severity and frequency of these dust storms, and conceding that the modeling system applied in this work did not produce the desired agreement between simulations and observations, additional research in both the windblown dust emissions model and the weather research/physicochemical model is called for.
Implications: While many dust storms can be considered to be natural, in semi-arid climates such storms often have an anthropogenic component in their sources of dust. Applying the natural, exceptional events policy to these storms with strong signatures of anthropogenic sources would appear not only to be misguided but also to stifle genuine regulatory efforts at remediation. Those dust storms that have resulted, in part, from passage over abandoned farm land should no longer be considered “natural”; policymakers and lawmakers need to compel the owners of such land to reduce its potential for windblown dust. 相似文献
