In this study, Talinum triangulare Jacq. (Willd.) treated with different lead (Pb) concentrations for 7 days has been investigated to understand the mechanisms of ascorbate–glutathione metabolisms in response to Pb-induced oxidative stress. Proteomic study was performed for control and 1.25 mM Pb-treated plants to examine the root protein dynamics in the presence of Pb. Results of our analysis showed that Pb treatment caused a decrease in non-protein thiols, reduced glutathione (GSH), total ascorbate, total glutathione, GSH/oxidized glutathione (GSSG) ratio, and activities of glutathione reductase and γ-glutamylcysteine synthetase. Conversely, cysteine and GSSG contents and glutathione-S-transferase activity was increased after Pb treatment. Fourier transform infrared spectroscopy confirmed our metabolic and proteomic studies and showed that amino, phenolic, and carboxylic acids as well as alcoholic, amide, and ester-containing biomolecules had key roles in detoxification of Pb/Pb-induced toxic metabolites. Proteomic analysis revealed an increase in relative abundance of 20 major proteins and 3 new proteins (appeared only in 1.25 mM Pb). Abundant proteins during 1.25 mM Pb stress conditions have given a very clear indication about their involvement in root architecture, energy metabolism, reactive oxygen species (ROS) detoxification, cell signaling, primary and secondary metabolisms, and molecular transport systems. Relative accumulation patterns of both common and newly identified proteins are highly correlated with our other morphological, physiological, and biochemical parameters. 相似文献

Environmental toxicants viz lead or cadmium and phthalate esters (di(2-ethylhexyl) phthalate [DEHP], dibutyl phthalate [DBP], and diethyl phthalate [DEP]) widely found in different environmental strata are linked to deteriorating male reproductive health. The objective was to assess the relationships between the seminal lead, cadmium, and phthalate (DEHP, DBP, DEP) concentrations at environmental level and serum hormone levels and semen quality in non-occupationally exposed men and specify the effect of individual and combined exposure of toxicants on semen quality. A study of 60 male partners of couples attending the Andrology Laboratory of the Reproductive Biology Department, All India Institute of Medical Sciences (AIIMS), New Delhi, India for semen analysis to assess their inability to achieve a pregnancy was selected for the study. The results of univariate and stepwise multiple regression analysis in the unadjusted model showed a significant correlation between lead or cadmium and phthalates DEHP/DBP/DEP and sperm motility, sperm concentration, and DNA damage. After adjusting for potential confounders, an association with lead or DEHP was only observed. The present data shows that lead (Pb) or cadmium (Cd) or phthalates might independently contribute to decline in semen quality and induce DNA damage. Phthalates might influence reproductive hormone testosterone. These findings are significant in light of the fact that men are exposed to a volley of chemicals; however, due to the small sample size, our finding needs to be confirmed in a larger population. 相似文献

With growing concerns of fossil fuel resources availability and the volatility of crude oil price, it is becoming imperative day by day to utilize the renewable sources of energy in a sustainable, environment friendly and energy efficient manner. India is the world’s second largest producer of cotton after China. India also has several agricultural and forest residues, and cotton residue is one of the most abundant agricultural residues after rice and wheat residues. The hydropyrolysis of cotton residues has been carried out at various pressures (1, 20 and 40 bar) and temperatures (300, 350, 400 and 450 °C). The effects of temperature and pressure have been studied to understand their yield patterns, and it has been observed that 20 bar pressure and 400 °C are the optimum conditions. The thermogravimetric analysis shows that cotton residue has two significant decomposition temperatures. The SEM, XRD patterns and FT-IR spectra clearly indicate the decomposition of the macromolecular structure of the cotton residue and formation of low molecular weight hydrocarbons suitable for various applications. 相似文献

Because of the continuous production of large amount of waste tires, the disposal of waste tires represents a major environmental issue throughout the world. This paper reports the utilization of waste tires (hard-to-dispose waste) as a precursor in the production of activated carbons (pollution-cleaning adsorbent). In the preparation of activated carbon (AC), waste rubber tire (WRT) was thermally treated and activated. The tire-derived activated carbon was characterized by means of scanning electron microscope, energy-dispersive X-ray spectroscopy, FTIR spectrophotometer, and X-ray diffraction. In the IR spectrum, a number of bands centred at about 3409, 2350, 1710, 1650, and 1300–1000 cm?1 prove the present of hydroxyl and carboxyl groups on the surface of AC in addition to C═C double bonds. The developed AC was tested and evaluated as potential adsorbent removal of chromium (III). Experimental parameters, such as contact time, initial concentration, adsorbent dosage and pH were optimized. A rapid uptake of chromium ions was observed and the equilibrium is achieved in 1 h. It was also found that the adsorption process is pH dependent. This work adds to the global discussion of the cost-effective utilization of waste rubber tires for waste water treatment. 相似文献

It is the first report in which a novel psychrotrophic Pseudomonas putida SKG-1 strain was evaluated for simultaneous bioremediation of pentachlorophenol and Cr6+ under various cultural and nutritional conditions. Pentachlorophenol (PCP) dechlorination products, bacterial structure, and functional groups were characterized by gas chromatography and mass spectrometry (GC–MS), scanning electron microscope and energy dispersive X-ray spectroscopy (SEM–EDS), and Fourier-transform infrared (FTIR) techniques. The strain was extremely tolerant to excessively higher individual concentration of PCP (1,400 mg l?1) and Cr6+ (4,300 mg l?1). Increasing concentration of PCP and Cr6+ exerted inhibitory effect on bacterial growth and toxicants’ removal. The strain exhibited growth, and concomitantly remediated both the pollutants simultaneously over a broad pH (7.0–9.0) and temperature (28–32 °C) range; maximum growth, PCP dechlorination (87.5 %), and Cr6+ removal (80.0 %) occurred at optimum pH 8.0 and 30 °C (from initial PCP 100 mg l?1 and Cr6+ 500 mg l?1) under shaking (150 rpm) within 72 h incubation. Optimization of agitation (125 rpm) and aeration (0.4 vvm) in bioreactor further enhanced PCP dechlorination by ~10 % and Cr6+ removal by 2 %. A direct correlation existed between growth and bioremediation of both the toxicants. Among other heavy metals, mercury exerted maximum and cobalt minimum inhibitory effect on PCP dechlorination and Cr6+ removal. Chromate reductase activity was mainly associated with the supernatant and cytosolic fraction of bacterial cells. GC–MS analysis revealed the formation of tetrachloro-p-hydroquinone, 2,4,6-trichlorophenol, and 2,6-dichlorophenol as PCP dechlorination products. FTIR spectrometry indicated likely involvement of carbonyl and amide groups in Cr3+ adsorption, and SEM–EDS showed the presence of chromium on P. putida surface. Thus, our promising isolate can be ecofriendly employed for biotreatment of various industrial wastes contaminated with high PCP and Cr6+ concentrations. 相似文献

An ordered hierarchical meso/macroporous monoclinic bismuth vanadate (BiVO4) particle was fabricated for the first time by a simple two-step melamine template hydrothermal method followed by calcination. The physiochemical parameters of as-prepared porous materials were characterized by means of X-ray diffraction, scanning electron microscopy, high-resolution transmission electron microscopy, Raman, Barrett–Emmett–Teller, and UV–vis techniques. The nitrogen adsorption–desorption measurement and pore size distribution curve suggest that meso/macropores exist in these hierarchical microarchitectures. Further, it is found that melamine plays a significant role in the formation of porous BiVO4 particles, and when a known amount of melamine was added, the surface area and pore size of such porous BiVO4 particles were increased. The photocatalytic activities of the as-prepared hierarchical BiVO4 samples were measured for the photodegradation of Congo red aqueous dye solution under visible light irradiation. Surprisingly, the porous BiVO4 particles showed outstanding photocatalytic activities than polycrystalline BiVO4 sample. Indiscriminate release of metal oxide nanoparticles (NPs) into the environment due to anthropogenic activities has become a serious threat to the ecological system including plants. The present study assesses the toxicity of nano-CuO on rice (Oryza sativa cv. Swarna) seedlings. Three different levels of stress (0.5 mM, 1.0 mM and 1.5 mM suspensions of copper II oxide, <50 nm particle size) were imposed and seedling growth performance was studied along control at 7 and 14 d of experiment. Modulation of ascorbate–glutathione cycle, membrane damage, in vivo ROS detection, foliar H2O2 and proline accumulation under nano-CuO stress were investigated in detail to get an overview of nano-stress response of rice. Seed germination percentage was significantly reduced under stress. Higher uptake of Evans blue by nano-CuO stressed roots over control indicates loss of root cells viability. Presence of dark blue and deep brown spots on leaves evident after histochemical staining with NBT and DAB respectively indicate severe oxidative burst under nano-copper stress. APX activity was found to be significantly increased in 1.0 and 1.5 mM CuO treatments. Nevertheless, elevated APX activity might be insufficient to scavenge all H2O2 produced in excess under nano-CuO stress. That may be the reason why stressed leaves accumulated significantly higher H2O2 instead of having enhanced APX activity. In addition, increased GR activity coupled with isolated increase in GSH/GSSG ratio does not seem to prevent cells from oxidative damages, as evident from higher MDA level in leaves of nano-CuO stressed seedlings over control. Enhanced proline accumulation also does not give much protection against nano-CuO stress. Decline in carotenoids level might be another determining factor of meager performance of rice seedlings in combating nano-CuO stress induced oxidative damages. 相似文献

Chlorpyrifos (O,O-diethyl O-3,5,6-trichloro-2-pyridylphosphorothioate) is one of the organophosphate pesticides widely used in agricultural practices throughout world and irreversible inhibitor of cholinesterase in all animal species. Limited efforts have been made to study acute genotoxic effects of chlorpyrifos (CPF) in different tissues of fish using genotoxic biomarkers. Therefore, the present investigation was aimed to study the induction of DNA damage by CPF in freshwater teleost fish Channapunctatus using micronucleus assay (MN assay) and alkaline single-cell gel electrophoresis (comet assay). The value of LC(50) - 96 h of CPF was determined as 811.98 microgl(-1) for C. punctatus, in a semi-static system and on the basis of LC(50) value three acute concentrations viz., 203, 406 and 609 microgl(-1) were determined. The fishes were exposed to the different concentrations of CPF for 96 h and samplings were done at regular intervals for assessment of the MN frequencies and DNA damage. In general, significant effects (P<0.01) from both concentrations and time of exposure were observed in exposed fishes. It was found that the micronucleus induction was highest on 96 h at all concentrations in the peripheral blood. Similar trend was observed for the DNA damage measured in terms of the percentage of tail DNA in the lymphocyte and gill cells. This study explored the combined use of micronucleus assay and comet assay for in vivo laboratory studies using fresh water fish for screening the genotoxic potential of xenobiotics. 相似文献

The aim of this paper was to obtain the first screening ecotoxicological risk evaluation in the Sunderban wetlands, the largest prograding delta in the estuarine phase of the River Ganges. The characterization of exposure was conducted by means of an extensive survey of several persistent organic pollutants (PAHs, PCBs, DDTs, PBDEs, HCHs, HCB) measured in seven core sediments from the Sunderban wetlands, obtaining a dataset with more than 2200 analyses. The pollutant effects were assessed by the use of three different sediment quality guidelines (SQGs) previously developed in the literature to evaluate toxicity induced in sediment-dwelling organisms. The three different approaches chosen for risk assessment of the Sunderban were the consensus SQGs obtained by TEC (threshold effect concentration), PEC (probable effect concentration) and EEC (extreme effect concentration), the threshold/probable effect level (TEL/PEL) approach and, finally, the ERL-ERM guidelines, including the m-ERM-Q (mean ERM quotient). The evaluation of the toxicity induced by a mixture of the target pollutants indicated the importance of gamma-HCH contamination in the Sunderban sediments despite the very low concentrations measured in core sediments. A different sensitivity for toxicity assessment due to quality guidelines was obtained, as the consensus SQGs based on TEC were less conservative and protective than the TEL and ERL approaches, while the use of m-ERM-Q seems to be the most powerful tool to predict the toxicity related to a contaminant mixture. 相似文献
