The ability of cadmium uptake by metal-resistant yeast, Candida tropicalis, from the liquid medium and wastewater was evaluated. The minimum inhibitory concentration of Cd2?+? against C. tropicalis was 2,500 mg L???1. The yeast also showed tolerance toward Zn2?+? (1,400 mg L???1), Ni2?+? (1,000 mg L???1), Hg2?+? (1,400 mg L???1), Cu2?+? (1,000 mg L???1), Cr6?+? (1,200 mg L???1), and Pb2?+? (1,000 mg L???1). The yeast isolate showed typical growth curves, but lag and log phases extended in the presence of cadmium. The yeast isolate showed optimum growth at 30°C and pH 8. The metal processing ability of the isolate was determined in a medium containing 100 mg L???1 of Cd2?+?. C. tropicalis could decline Cd2?+? 70%, 85%, and 92% from the medium after 48, 96, and 144 h, respectively. C. tropicalis was also able to remove Cd2?+? 40% and 78% from the wastewater after 6 and 12 days, respectively. Cd produced an increase in glutathione (GSH) and nonprotein thiol levels by 135% and 134% at 100-mg L???1 concentration, respectively. An increase in the synthesis of GSH is involved in metal tolerance, and the presence of increasing GSH concentrations may be a marker for high metal stress in C. tropicalis. C. tropicalis, which is resistant to heavy metal ions and is adaptable to the local environmental conditions, may be employed for metal detoxification operations.  相似文献

The present study is aimed at assessing the ability of Bacillus sp.JDM-2-1 and Staphylococcus capitis to reduce hexavalent chromium into its trivalent form.Bacillus sp.JDM-2-1 could tolerate Cr(Ⅵ) (4800 μg/mL) and S.capitis could tolerate Cr(Ⅵ) (2800 μg/mL).Both organisms were able to resist Cd2+ (50 μg/mL),Cu2+ (200 μg/mL),Pb2+ (800 μg/mL),Hg2+ (50 μg/mL) and Ni2+ (4000 μg/mL).S.capitis resisted Zn2+ at 700 μg/mL while Bacillus sp.JDM-2-1 only showed resistance up to 50 μg/mL.Bacillus sp.JDM-2-1 and S.capitis showed optimum growth at pH 6 and 7,respectively,while both bacteria showed optimum growth at 37℃.Bacillus sp.JDM-2-1 and S.capitis could reduce 85% and 81% of hexavalent chromium from the medium after 96 h and were also capable of reducing hexavalent chromium 86% and 89%,respectively,from the industrial effluents after 144 h.Cell free extracts of Bacillus sp.JDM-2-1 and S.capitis showed reduction of 83% and 70% at concentration of 10 μg Cr(Ⅵ)/mL,respectively.The presence of an induced protein having molecular weight around 25 kDa in the presence of chromium points out a possible role of this protein in chromium reduction.The bacterial isolates can be exploited for bioremediation of hexavalent chromium containing wastes,since they seem to have the potential to reduce the toxic hexavalent form to its nontoxic trivalent form.  相似文献

Background, aim, and scope  

Soil contamination with heavy metals occurs as a result of both anthropogenic and natural activities. Heavy metals could have long-term hazardous impacts on the health of soil ecosystems and adverse influences on soil biological processes. Soil enzymatic activities are recognized as sensors towards any natural and anthropogenic disturbance occurring in the soil ecosystem. Similarly, microbial biomass carbon (MBC) is also considered as one of the important soil biological activities frequently influenced by heavy metal contamination. The polymerase chain reaction–denaturing gradient gel electrophoresis (DGGE) has recently been used to investigate changes in soil microbial community composition in response to environmental stresses. Soil microbial community structure and activities are difficult to elucidate using single monitoring approach; therefore, for a better insight and complete depiction of the soil microbial situation, different approaches need to be used. This study was conducted in a greenhouse for a period of 12 weeks to evaluate the changes in indigenous microbial community structure and activities in the soil amended with different application rates of Cd, Pb, and Cd/Pb mix. In a field environment, soil is contaminated with single or mixed heavy metals; so that, in this research, we used the selected metals in both single and mixed forms at different application rates and investigated their toxic effects on microbial community structure and activities, using soil enzyme assays, plate counting, and advanced molecular DGGE technique. Soil microbial activities, including acid phosphatase (ACP), urease (URE), and MBC, and microbial community structure were studied.  相似文献
