E. coli and enterococci in recreational waters are monitored as indicators of fecal contamination, pathogen presence, and health risk. Quantitative polymerase chain reaction (qPCR) tests for fecal indicator bacteria can provide beach managers with same-day information about water quality, unlike culture methods which provide that information the following day. The abilities of qPCR measurements of indicator bacteria, as compared to culture measurements of indicator bacteria, as predictors of pathogen presence or density in surface waters are not well understood. The purpose of this study was to make such comparisons between water samples collected from Chicago area surface waters, including rivers, inland lakes, Lake Michigan, and the Chicago Area Waterways System, which is dominated by wastewater effluent. A total of 294 twenty-litre samples were collected and analyzed for Giardia and Cryptosporidium. qPCR and membrane filtration methods were used to quantify E. coli and enterococci. Correlation, logistic regression, and zero-inflated Poisson modeling were utilized to evaluate associations between indicators and parasites. qPCR and culture measures of the indicator bacteria were similar in their ability to predict parasite presence and density. Correlations between parasites and indicators were generally stronger at waters not dominated by effluent. Associations between indicator density and Giarida presence were observed more consistently than between indicator density and Cryptosporidium presence. Associations between enterococci and parasites were generally stronger than associations between E. coli and parasites. The use of qPCR monitoring in our setting would generate more timely results without compromising the ability to predict parasite presence or density.  相似文献

Ensuring the persistence of at-risk species depends on implementing conservation actions that ameliorate threats. We developed and implemented a method to quantify the relative importance of threats and to prioritize recovery actions based on their potential to affect risk to Mojave desert tortoises (Gopherus agassizii). We used assessments of threat importance and elasticities of demographic rates from population matrix models to estimate the relative contributions of threats to overall increase in risk to the population. We found that urbanization, human access, military operations, disease, and illegal use of off highway vehicles are the most serious threats to the desert tortoise range-wide. These results suggest that, overall, recovery actions that decrease habitat loss, predation, and crushing will be most effective for recovery; specifically, we found that habitat restoration, topic-specific environmental education, and land acquisition are most likely to result in the greatest decrease in risk to the desert tortoise across its range. In addition, we have developed an application that manages the conceptual model and all supporting information and calculates threat severity and potential effectiveness of recovery actions. Our analytical approach provides an objective process for quantifying threats, prioritizing recovery actions, and developing monitoring metrics for those actions for adaptive management of any at-risk species.  相似文献

This article presents an analysis of the projected performance of urban residential rainwater harvesting systems in the United States (U.S.). The objectives are to quantify for 23 cities in seven climatic regions (1) water supply provided from rainwater harvested at a residential parcel and (2) stormwater runoff reduction from a residential drainage catchment. Water‐saving efficiency is determined using a water‐balance approach applied at a daily time step for a range of rainwater cistern sizes. The results show that performance is a function of cistern size and climatic pattern. A single rain barrel (190 l [50 gal]) installed at a residential parcel is able to provide approximately 50% water‐saving efficiency for the nonpotable indoor water demand scenario in cities of the East Coast, Southeast, Midwest, and Pacific Northwest, but <30% water‐saving efficiency in cities of the Mountain West, Southwest, and most of California. Stormwater management benefits are quantified using the U.S. Environmental Protection Agency Storm Water Management Model. The results indicate that rainwater harvesting can reduce stormwater runoff volume up to 20% in semiarid regions, and less in regions receiving greater rainfall amounts for a long‐term simulation. Overall, the results suggest that U.S. cities and individual residents can benefit from implementing rainwater harvesting as a stormwater control measure and as an alternative source of water.  相似文献

This paper presents the chemical reaction engineering development of the H2O2/VisUV photo-oxidation process for treatment of hazardous waterborne substances, that occur in groundwater, leachates, and industrial wastewater. Reaction results, on benzene (BNZ), dichlorobenzene (DCB), trichloroethene (TCE), trichloroethane (TCA), and carbon tetrachloride (CTC), have been obtained, providing engineering data and models that can be used to size full-scale equipment. A photochemical flow stirred tank reactor (pcfSTR) and a photo-chemical tubular flow reactor (pcTFR) were used in the experimental work. Two experimental discoveries were made in the course of the work: (1) conventional thermal kinetics do not apply, the rate controlling variable is the photon flux, and (2) for the photo-chemical reactors used, the pcfSTR was more effective than the pcTFR. The following sub-topics are discussed: reaction mechanism, reactor hydrodynamics, photon flux effects, typical reaction data (on benzene and trichloroethane), and rate constants.  相似文献

We present the first confirmed case by molecular analysis of a metaphyseal chondrodysplasia, McKusick type, in a 22-week fetus. Two novel compound heterozygous mutations, 64T> A and 79G > T, were found in the highly conserved regions of the RMRP gene. Twenty-two heterozygous g.1018 T> C mutations, two homozygous g.1018 T> C mutations, two heterozygous insertion mutations g.799_g.800insC and one heterozygous insertion mutation g.849_g.850insT were found among 100 normal controls. Careful radiological examination of the fetus for skeletal dysplasia allowed definitive diagnosis, proper genetic counselling and future prenatal diagnosis. Gloucestershire County Council have developed a method for making annual Parish and Ward estimates which the authors claim combines robustness, economy and simplicity with a degree of rigour, using available data sources. Using an innovative, two-stage process to remediate uranium-contaminated soils, researchers at Los Alamos National Laboratory's (LANL's) Technical Area 33 (TA-33) successively reduced 218 cubic yards of contaminated soil to approximately 30 gallons of leachate solution and resins. In the first step, the contaminated soil is separated from the clean soil using the Thermo Nuclean (a division of the Thermo NUtech company) Segmented Gate System (SGS). Contaminated soil proceeds via conveyor belt to a separate storage bin to await further processing, while uncontaminated soil is returned to its original location. From the 218 cubic yards of soil excavated from the test site at TA-33, only seven cubic yards were found to contain uranium contamination above the criterion release limit, yielding an initial waste volume reduction of 97 percent. Using the containerized vat leaching (CVL) method, a technique borrowed from the mining industry, the uranium was then removed from the reduced volume of contaminated soil.
