Biomonitoring studies have increased as a consequence of risks and effects to human health on exposure to environmental contaminants, mainly air pollutants. Genetic biomarkers are useful tools for the early assessment of exposure to occupational and environmental pollution. The objective of the present study was to investigate genotoxic effects on people residing and/or working downwind from an oil refinery in southern Brazil and the mutagenic activity of airborne particulate matter (PM10). Samples of peripheral blood and buccal mucosa cells were evaluated using the single-cell gel electrophoresis assay (comet assay) and the micronucleus (MN) assay, respectively. PM10 samples were collected in the target site and the organic matter extraced with dichloromethane was assessed for mutagenic activity in the Salmonella/microsome assay. The exposed group (n = 37) was compared to a reference group (n = 37) of subjects living in an urban area with limited traffic and industrial influence, located far from the main industrial areas. All PM10 organic extracts showed mutagenic positive responses and the effect decreased in the presence of S9 mix indicating that the predominant compounds present were direct-acting mutagens. The responses of YGs strains are consistent with aromatic amines and nitroarenes being present in the PM10 extracts. The group in the area under the influence of the oil refinery (exposed group) showed significantly higher DNA damage in lymphocytes than the reference group. The MN frequencies in buccal mucosa were very low for both groups and no difference between groups was observed. No association was found between age and tobacco smoking habit and level of DNA damages measured by the comet assay. The results indicate that the comet assay was a sensitive tool to detect DNA damage in subjects under the influence of an oil refinery, with marked genotoxic activity in the atmospheric environment.  相似文献

Following the Chernobyl accident, 137Cs contamination levels of wild boar in some districts of Southern Germany are still exceeding thousands of Bq kg−1. While the long term 137Cs concentration in forest plants, mushrooms, and roe deer meat has decreased significantly, for wild boar it has remained constant during the last decade. Between 1998 and 2008, we analysed the muscle meat of 656 wild boars shot in the district (“Landkreis”) Ravensburg. The 137Cs activity concentration showed considerable variability from less than 5 up to 8266 Bq kg−1 and it followed a seasonal pattern, which is attributed to changes in dietary habits, fodder availability, meteorological conditions and specific behaviour of 137Cs in wild boar organism. Tag values for wild boars from the district Ravensburg varied from 0.008 to 0.062 m2 kg−1 during 2000–2008.  相似文献

This special issue of AMBIO presents a new approach to sustainability science that goes beyond interdisciplinary research. Using coupled natural and human systems, or landscapes, as multiple case studies in Europe’s East and West knowledge production and learning toward transdisciplinary research was applied in Sweden, countries in Central and Eastern Europe, and Russia. First, the research group Forest-Landscape-Society summarizes the research program (2005–2012) behind this special issue of AMBIO and its development to participate in transdisciplinary research. Environmental objects (surface and groundwater, soil, bottom sediments, wastewater) are reservoirs in which large-scale multidirectional exchange of determinants of antibiotic resistance between clinical strains and natural bacteria takes place. The review discusses the results of studies on antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARG) isolated from environmental objects (water, soil, sewage, permafrost) of the Russian Federation. Despite the relevance of the topic, the number of available publications examining the resistomes of Russian water bodies and soils is small. The most studied environmental objects are surface waters (rivers, lakes), permafrost deposits. Soil resistomes are less studied. Data on ARG and ARB in wastewater are the least covered in publications. In most of the studies, antibiotic resistance of isolated pure bacterial cultures was determined phenotypically. A significant number of publications are devoted to the resistance of natural isolates of Vibrio cholerae, since the lower reaches of the Volga and Don rivers are endemic to cholera. Molecular genetic methods were used in a small number of studies. Geographically, the south of the European part of Russia is the most studied. There are also publications on the distribution of ARG in water bodies of Siberia and the Russian Far East. There are practically no publications on such developed regions of Russia as the center and northwest of the European part of Russia. The territory of the country is very large, anthropogenic and natural factors in its various regions vary significantly; therefore, it seems interesting to combine all available data in one work.

From the collated data relevant to discharges by the nuclear industry, it results that the input of beta activity (excluding Chernobyl fallout and tritium) into the OSPAR region decreased by a factor of 4 from 1986 to 1991, reaching by this date the same level as in the early 1950s. Over the same period the discharges of the alpha activity into the OSPAR region also decreased by a factor 3, the same trend has been seen also for tritium. Since 1986 the effective dose to members of the critical group in the vicinity of Sellafield and Cap de La Hague was consistently below the ICRP and EU limit of 1 mSv per year to members of the general public. The overall radiological impact from nuclear industry on the population of the European Union from the OSPAR area has decreased from 280 manSv y(-1) in 1978 to 14 manSv y(-1) in 2000.  相似文献

Due to the rise of functional foods,the distinction between foods and medicines hasbecome increasingly blurred. A new EUregulation covering health claims and otherclaims on food and drink products is on thedocks. A basic motive of legal regulation oflabeling and advertising is to inform andprotect the consumer. Promotion of informedchoice and consumer protection may, however, beconflicting objectives. A further problemsprings from the fact that choice, likeconsent, is a propositional attitude andtherefore opaque. Thus it is extremelydifficult for regulators to fasten onparticular formulations of claims. This paper presents results of a model test carried out within the framework of the COMETES project (EU). The aim of the work was to change the structure of the MOIRA lake model for radiocesium so that it can be applied more generally for, in principle, all types of radionuclides and heavy metals. This general lake model is used within the MOIRA decision support system (DSS; MOIRA and COMETES are acronyms for EU-projects). The model is based on a set of differential equations and a specific modelling structure. It incorporates all important fluxes to, from and within lakes in a general manner. Yet the model is driven by a minimum of variables accessible from standard maps and monitoring programs. The model can be separated into two parts, a general part with equations applicable for all types of water pollutants and a substance-specific part. This model has previously been validated for 137Cs from many lakes covering a wide domain and yielded excellent predictive power. The alterations discussed in this work are meant to be general and radiostrontium is used as a typical element. Radiostrontium is known to be more mobile than radiocesium and all abiotic parts of the model handling fixation and mobility have been altered. The new model for 90Sr has been critically tested using data from four lakes heavily contaminated with 90Sr from the Kyshtym accident in the Southern Urals, Russia, using empirical data from a period from 1958 to 1995 for 90Sr in fish (here goldfish), water and sediments.  相似文献

The Hanford test scenario described an accidental release of 131I to the environment from the Hanford Purex Chemical Separations Plant in September 1963. Based on monitoring data collected after the release, this scenario was used by the Dose Reconstruction Working Group of BIOMASS to test models typically used in dose reconstructions. The primary exposure pathway in terms of contribution to human doses was ingestion of contaminated milk and vegetables. Predicted mean doses to the thyroid of reference individuals from ingestion of 131I ranged from 0.0001 to 0.8 mSv. For one location, predicted doses to the thyroids of two children with high milk consumption ranged from 0.006 to 2 mSv. The predicted deposition at any given location varied among participants by a factor of 5-80. The exercise provided an opportunity for comparison of assessment methods and conceptual approaches, testing model predictions against measurements, and identifying the most important contributors to uncertainty in the assessment result. Key factors affecting predictions included the approach to handling incomplete data, interpretation of input information, selection of parameter values, adjustment of models for site-specific conditions, and treatment of uncertainties.  相似文献

Phytotoxicity, cytotoxicity and genotoxicity of pesticides with various mechanisms of targeted activity were studied in a hydroponic culture of 2-day-old seedlings of Triticum aestivum. All studied pesticides (with the exception of metribuzin) exhibited dose-dependent phytotoxicity (inhibited the growth of the main root and reduced the yield of root biomass). All studied pesticides did not affect mitotic index in the root apex meristem but did affect the duration of some phases of mitosis. Herbicides increased, while fungicides, on the contrary, decreased the duration of the cytokinesis phase. All pesticides (1 μg/mL) exhibited genotoxic activity: in the root apex meristem the number of cells with mitotic abnormalities was significantly higher than in the control variant (7–14 times). The genotoxic activity of metribuzin and tebuconazole was 2 times lower than for tribenuron-methyl, fenoxaprop-P-ethyl, epoxiconazole and azoxystrobin. The genotoxicity of the studied pesticides was combined: depending on the class of the pesticide, clastogenic or aneugenous effects dominated.
