The rate of photodegradation of two chelating agents, ethylenediaminetetraacetic acid (EDTA) and an isomeric mixture of ethylenediamine disuccinic acid (EDDS), was analysed in humic lake water and in distilled water using exposure to sunlight, and in the laboratory using lamps emitting UV radiation in the range 315-400 nm. Degradation was studied using Fe(III) complexes and sodium salts of chelates. Fe(III) complexes were illuminated at pH 3.1 and 6.5. The results demonstrated that the rate of photodegradation of Fe(III)-EDTA and Fe(III)-EDDS complexes seems to be pH dependent. In the laboratory experiments degradation occurred much faster when the original pH was 3.1 rather than 6.5. The photodegradation of the isomeric mixture of EDDS was markedly faster than the degradation of EDTA both in the laboratory and field experiments, and both in humic and distilled water. The results indicated that in natural waters photodegradation of EDDS is independent of initial speciation of EDDS, while degradation of EDTA is dependent on its existence as Fe(III)-EDTA species.  相似文献

Objective

We performed a 1-year evaluation of a novel strategy of simultaneously analyzing single nucleotide variants (SNVs), copy number variants (CNVs) and copy-number-neutral Absence-of-Heterozygosity from Whole Exome Sequencing (WES) data for prenatal diagnosis of fetuses with ultrasound (US) anomalies and a non-causative QF-PCR result.

Methods

After invasive diagnostics, whole exome parent-offspring trio-sequencing with exome-wide CNV analysis was performed in pregnancies with fetal US anomalies and a non-causative QF-PCR result (WES-CNV). On request, additional SNV-analysis, restricted to (the) requested gene panel(s) only (with the option of whole exome SNV-analysis afterward) was performed simultaneously (WES-CNV/SNV) or as rapid SNV-re-analysis, following a normal CNV analysis.

Results

In total, 415 prenatal samples were included. Following a non-causative QF-PCR result, WES-CNV analysis was initially requested for 74.3% of the chorionic villus (CV) samples and 45% of the amniotic fluid (AF) samples. In case WES-CNV analysis did not reveal a causative aberration, SNV-re-analysis was requested in 41.7% of the CV samples and 17.5% of the AF samples. All initial analyses could be finished within 2 weeks after sampling. For SNV-re-analysis during pregnancy, turn-around-times (TATs) varied between one and 8 days.

Conclusion

We show a highly efficient all-in-one WES-based strategy, with short TATs, and the option of rapid SNV-re-analysis after a normal CNV result.  相似文献

The population decline of the nominate lesser black-backed gull Larus fuscus fuscus in the Gulf of Finland (northern Baltic) is caused by an exceedingly high chick mortality due to diseases. The chick diseases include degeneration in various internal organs (primarily liver), inflammations (mainly intestinal), and sepsis, the final cause of death. The hypothesis of starvation causing intestinal inflammations (leading to sepsis) was tested by attempting to reproduce lesions in apparently healthy herring gull L. argentatus chicks in captivity. The herring gull chicks were provided a similar low food-intake frequency as observed for the diseased chicks in the wild. However, empty alimentary tract per se did not induce the intestinal inflammations and therefore, inflammations seem to be innate or caused by other environmental factors in the diseased lesser black-backed chicks. They had very high concentrations of PCB in their liver; but the concentrations were not significantly higher than those of the healthy herring gull chicks, indicating a common exposure area for both species (i.e. the Baltic Sea). When compared to NOEL and LOEL values for TEQs in bird eggs our TEQ levels clearly exceed most or all of the values associated with effects. Compared with published data on fish-eating waterbirds, the DDE concentrations in the diseased lesser black-backed chicks were well above the levels previously correlated with decreased reproduction, while the residues in apparently healthy herring gulls were below those levels. The DDE/PCB ratio in lesser black-backs was significantly elevated, indicating an increased exposure to DDTs as compared with most other Baltic and circumpolar seabirds. The possible exposure areas of DDT in relation to differential migration habits of the two gull species are discussed.  相似文献

ABSTRACT

A grading system was developed to rate the moisture damage profile of dwellings and to study the relationship between moisture-induced indoor air problems and occupant health. A total of 630 randomly selected houses and apartments, built between 1950 and 1989, were visually inspected. Moisture observations were standardized into three damage levels. Thus, a system to classify the homes into three grades was devised. The two grades of homes associated with the highest levels of damage were graded as index homes.

Overall, 51% of the sample had some kind of moisture fault in them and one in every three homes (33%) was classified as an index home. The mean number of damage incidents in the index dwellings varied from 1.4 to 2.6. The mean number of damage incidents in the reference homes was 0.28. Prevalence of index dwellings was significantly higher (p < 0.01) in houses (38%) than in apartments (26%). There was no major difference in the prevalence of index buildings in houses built in any particular decade (30-35%). Moisture was observed in 28% of bathrooms, in 10% of kitchens, and in 17% of other spaces. Indoor relative humidity (RH) levels were low in most homes.  相似文献

Natural organic matter (NOM) is abundant in natural waters in Finland and in many ways affects the unit operations in water purification. In this study, the organic matter content in water in different stages of a full-scale treatment process over 1 year was measured. The full-scale treatment sequence, studied at the Rusko water treatment plant in Tampere, Finland, consisted of coagulation, flocculation, clarification by sedimentation or flotation, activated carbon (AC) filtration, and disinfection. High-performance size exclusion chromatography (HPSEC) was used for separation to determine changes in the humic substances content during the purification process. In addition, total organic carbon (TOC), KMnO4-number, and UV-absorbance at wavelength 254 nm (UV254) were measured. High molecular weight (HMW) matter was clearly easier to remove in coagulation and clarification than low molecular weight (LMW) matter. Furthermore, depending on the regeneration of the activated carbon filters, activated carbon filtration was effective to a degree but did not remove most of the lowest molecular weight compounds. Significant correlation was established among HPSEC, KMnO4, UV254 absorbance, and TOC. HPSEC proved to be a fast and relatively easy method to estimate NOM content in water and, in fact, gave more information than traditional methods on the type of NOM in a water sample. Polychlorinated biphenyls (PCBs) have been used in diverse purposes such as indoor paints. Removal of these paints with dust creating techniques, like sandblasting, will result in contamination of building surfaces with PCB-containing dust. Objectives of this study was to analyze the PCB concentrations on surfaces after sandblasting with silica using wipe samples and estimate the resulting health risks and further calculate the risk based acceptable PCB surface concentrations that do not cause incremental lifetime cancer risk higher that 10(-5) or does not cause immunosupression effects in residential use or in occupational settings. Both deterministic and probabilistic approaches were used. The total PCB concentrations on surfaces ranged from 10 to 1100 microg/m(2). Estimated cancer risk was 1.2 x 10(-4) for childhood exposure, 1.3 x 10(-5) for adult residents and 1.5 x 10(-5) for occupational exposure. Probabilistic risk assessment revealed that point estimates were quite reasonable and located between 45th and 79th percentiles on probabilistic distribution of risk. The noncancer risks were calculated as hazard quotients (HQ) which ranged from 3.3 to 35 depending on the exposure scenario. Acceptable surface concentrations based on noncancer effects that are protective for 95% of exposed population were 7 microg/m(2) for residential use, 65 microg/m(2) for residential use if only adults will be exposed and 140 microg/m(2) for occupational use. Preliminary cleanup experiment revealed that when contaminated dust was carefully removed with industrial vacuum cleaner and further washed with terpene containing liquid the surface concentration dropped below the acceptable levels calculated in this study.  相似文献

This study is concerned with how car and steel workers in different countries see their work and what features of their work they feel important. The material is drawn from two international projects, namely the car and steel industry studies, both coordinated by the Vienna Centre. The former study was carried out in 15 countries, comprising in all 126 production units from 38 plants and 3214 interviews of workers. The latter study was carried out in 10 countries, in 24 plants and 47 production units, 1576 workers were interviewed. Clear differences were found between countries in how critically the workers were oriented to the burdens and challenges of their work. This was interpreted to reflect country-specific work assessment standards. However, differences should be seen as differences in emphasis rather than as manifestations of work cultures, which substantially differ from each other. The order of importance among work characteristics was roughly the same from country to country: matters related to physical and economic security were widely considered the most important aspects of work.  相似文献

The nitrifying activity and the effect of fertilization with urea and methylene urea were studied in a landfarming site. The site has been operative over 20 years and maintained by heavy nitrogen fertilization. The landfarming soil contained 4-6% (w/w) oil. The nitrate accumulation was 20-50mg NO3-N day(-1)kg(-1) observed after methylene urea fertilization of 889 g Nm(-2). Nitrification ex situ (in laboratory conditions) was 8.8 mg NO3-N day(-1) kg(-1) in the presence of 380 mg kg(-1) NH4+-N. The half-saturation concentration of nitrification was more than 200 mg NH4+-N kg(-1). The results show that nitrification was active in soil with high oil concentration. Urea fertilization of 893 g Nm(-2) caused an increase of soil NH4+-N concentration up to 5500 mg kg(-1) and pH>8.5. This led to inhibition of nitrification, which persisted after NH4+ concentration decreased below 200mg NH4+ kg(-1).  相似文献

The environmental fate of ethylenediaminetetraacetic acid (EDTA) has been extensively studied, while much less is known about the environmental behaviour of diethylenetriaminepentaacetic acid (DTPA). In this study, it was confirmed that DTPA is persistent toward biodegradation. The biodegradability of DTPA was investigated in the absence and in the presence of Fe(III) by using CO2 evolution test and Manometric respirometry test. The CO2 evolution and oxygen uptake of iron-free (DTPA was added as free acid) and Fe(III)DTPA were less than in inoculum blank. Possible inhibitor effect was analysed by testing biodegradation of sodium benzoate with and without iron-free or Fe(III)DTPA in the Manometric respirometry test. Only slight inhibition was observed when DTPA was added as free acid. Photodegradation of iron-free DTPA and Fe(III)-DTPA complex was studied by using sunlight and UV radiation at the range 315-400 nm emitted by black light lamps. The results indicate that DTPA added as free acid degrades photochemically in humic lake water. Fe(III)DTPA was shown to be very photolabile in humic lake water in the summer; the photochemical half-life was below one hour. Photodegradation products were identified by the mass spectrometric technique (GC-MS). It was shown that photodegradation of Fe(III)DTPA does not result in total mineralization of the compound. Diethylenetriaminetetraacetic acid, diethylenetriaminetriacetic acid, ethylenediaminetriacetic acid, N,N'- and/or N,N-ethylenediaminediacetic acid, iminodiacetate, ethylenediaminemonoacetic acid and glycine were identified as photodegradation products of Fe(III)DTPA. Based on these observations, we propose a photodegradation pathway for Fe(III)DTPA.  相似文献

ABSTRACT

This study presents the heavy metal results obtained during Finnish interlaboratory comparison (ILC) measurements made during 2019. The aim of this measurement campaign was to verify the skills of accredited emission measurement teams and also to evaluate the challenges that stack testing teams face in the future when emission levels decrease. ILCs have been organized in Finland since the 1970s. ILCs provide an important platform for stack testing teams so that they can verify their measurement skills and also for the dissemination of knowledge. The knowledge about the measurement standards and their requirements has improved among stack testing teams during past years in Finland. As emission levels get lower, they still need to pay more attention to some quality assurance procedures, e.g. to method and field blanks. Based on the observations of this ILC for heavy metals it can be noted that the challenges that stack testing teams face are related to the fact that no guidance is given in the standard reference methods EN 13211 and EN 14385 for example on the calculation of measurement uncertainties and how results below limit of quantification should be taken into account. These reference methods were suitable for their purpose at the time they were validated. However, emission levels are now more stringent and it is challenging to measure them with acceptable uncertainty criteria. As a consequence, there is a clear need for harmonized approaches in Europe for consistent implementation of standards and regulations. Key issues where guidance should be provided include realistic measurement uncertainties at low concentration levels, reporting low concentrations and guidance on how measurement uncertainties should be taken into account when the results are used for compliance assessment. The overall aim is to ensure that even with low emission levels, the emission measurement results would be transparent and robust throughout the EU.

Implications: Interlaboratory comparison measurements between stack testing teams are the most important tool to verify the quality of the measurements. Participation in an appropriate ILC is often mandatory to successfully achieve accreditation under ISO/IEC 17025. Such campaigns also provide an efficient platform for dissemination of knowledge. In addition, ILCs can be used to clarify the challenges that teams nowadays face when measuring low emission levels, thus creating important information for the revision work of standards.  相似文献
