Hematodinium perezi (genotype III) is a parasitic dinoflagellate that infects blue crabs along the eastern seaboard and Gulf of Mexico, USA. In order to examine the intra-specific genetic variation of this parasite, eleven microsatellite markers from H. perezi (III) were amplified from 227 infected blue crabs collected during 2008–2009 from six sites in Virginia. Simultaneous infections with multiple genetic types in a single-host individual were common and observed in 42 % of the samples. The remaining 58 % of samples had a single allele per locus at all eight polymorphic loci suggesting that the life history stages of the parasite in the host hemolymph are likely haploid. The composition and distribution of multi-locus genotypes (MLG) from samples with infections of a single genetic type indicated high genotypic variation along the Delmarva Peninsula, Virginia, with no evidence of population structure. The lack of linkage disequilibrium combined with the large number of unique MLGs (84 %) is strong evidence for recombination in the life cycle, but the sexual stages remain undetermined. This is the first evidence of ploidy level, infections by multiple genetic types in an individual host animal, high levels of genotypic variation, and sexual reproduction for any species of Hematodinium.  相似文献

Escolar (Lepidocybium flavobrunneum) is a large, mesopelagic fish that inhabits tropical and temperate seas throughout the world, and is a common bycatch in pelagic longline fisheries that target tuna and swordfish. Few studies have explored the biology and natural history of escolar, and little is known regarding its population structure. To evaluate the genetic basis of population structure of escolar throughout their range, we surveyed genetic variation over an 806 base pair fragment of the mitochondrial control region. In total, 225 individuals from six geographically distant locations throughout the Atlantic (Gulf of Mexico, Brazil, South Africa) and Pacific (Ecuador, Hawaii, Australia) were analyzed. A neighbor-joining tree of haplotypes based on maximum likelihood distances revealed two highly divergent clades (δ = 4.85%) that were predominantly restricted to the Atlantic and Indo-Pacific ocean basins. All Atlantic clade individuals occurred in the Atlantic Ocean and all but four Pacific clade individuals were found in the Pacific Ocean. The four Atlantic escolar with Pacific clade haplotypes were found in the South Africa collection. The nuclear ITS-1 gene region of these four individuals was subsequently analyzed and compared to the ITS-1 gene region of four individuals from the South Africa collection with Atlantic clade haplotypes as well as four representative individuals each from the Atlantic and Pacific collections. The four South Africa escolar with Pacific mitochondrial control region haplotypes all had ITS-1 gene region sequences that clustered with the Pacific escolar, suggesting that they were recent migrants from the Indo-Pacific. Due to the high divergence and geographic separation of the Atlantic and Pacific clades, as well as reported morphological differences between Atlantic and Indo-Pacific specimens, consideration of the Atlantic and Indo-Pacific populations as separate species or subspecies may be warranted, though further study is necessary.  相似文献

Phosphorus (P) loss in overland flow varies with spatial distribution of soil P, management, and hydrological pathways. The effect of flow time, flowpath length, and manure position on P loss in overland flow from two central Pennsylvania soils packed in boxes of varying length (0.5, 1.0, 1.5, 2.75, and 4.0 m long x 15 cm wide x 5 cm deep) were examined by collecting flow samples at 5-min intervals for 30 min (50 mm h(-1) rainfall) without and with 75 kg P ha(-1) applied as swine (Sus scrofa) manure over 0.5 m of the box slope length at distances of 0 to 3.5 m from the downslope collection point. Dissolved reactive P concentration was more closely related to the proportion of clay in sediment of overland flow before (r = 0.98) than after (r = 0.56) manure application. This was attributed to the transport of larger, low-density particles after applying manure. The concentration of dissolved and particulate P fractions decreased with increasing flowpath length, due to dilution rather than sorption of P by surface soil during overland flow. Total P loss (mainly as particulate P) from the Watson channery silt loam (fine-loamy, mixed, active, mesic Typic Fragiudult) was more than from Berks channery silt loam (loamy-skeletal, mixed, active, mesic Typic Dystrudept), even with manure applied. Thus, while P loss in overland flow is affected by where manure is applied relative to flowpath length, initial soil P concentration should not be discounted when looking at areas of potential P loss within a watershed.  相似文献

Soil phosphorus (P) quantity-intensity (q-i) relationships, based on common extraction methods, may potentially be used to estimate the risk of P loss in overland flow and subsurface drainage water. Some workers have used nonlinear q-i relationships to derive thresholds in soil test P (STP; a quantity factor) above which the risk of P loss increases, while others find linear relationships and no threshold. We present here a simple modelling exercise (based on Langmuir adsorption theory) along with data from literature to explain the behaviour of q-i relationships, and to give an explanation for this apparent discrepancy. The data indicate that q-i relationships are dependent upon the soil to solution ratio of the P intensity parameter, adsorption capacity (Qmax) and strength (K) of the soil, and the total range in STP. In turn, this affects the calculation of a threshold in STP. The q-i relationship tends towards linearity under conditions of a narrow total range of STP and/or when using a wide soil to solution ratio for estimating the P intensity parameter. Under such conditions, a threshold is difficult to detect, and uncertain. We conclude that the sensitivity of thresholds to experimental conditions and soils needs to be considered if thresholds are to be successful in environmental management to decrease P loss to surface waters.  相似文献

ABSTRACT: Cattle grazing practices in the western United States have contributed to widespread riparian degradation resulting in unstable channel morphologies and the loss of fish habitat. Because of prolonged disturbance, numerous riparian areas on both public and private lands have been fenced to exclude cattle in order to promote vegetation establishment and riparian improvement. We selected four gravel-bedded, steep alluvial streams in eastern Oregon with cattle exclosures greater than 14 years old for an analysis of geomorphic adjustments following the removal of cattle grazing. We compare channels inside exclosures and in adjacent grazed reaches to identify the salient stream channel properties that respond to the removal of riparian stresses and to document the magnitude of these changes. Results indicate that significant changes occur, with reductions in bankfull dimensions and increases in pool area being the most common and identifiable changes. At all four sites, bankfull widths are narrower by 10 to 20 percent, and the percentage of channel area occupied by pools is higher in the exclosure by 8 to 15 percent. The increase in pool area is primarily offset by a reduction in the percent glide area. Not all of the channel properties demonstrate adjustment, indicating that perhaps 14 years is an insufficient duration for these variables to adjust.  相似文献

The USEPA standards (40 CFR Part 503) for the use or disposal of sewage sludge (biosolids) derived risk-based numerical values for Mo for the biosolids --> land --> plant --> animal pathway (Pathway 6). Following legal challenge, most Mo numerical standards were withdrawn, pending additional field-generated data using modern biosolids (Mo concentrations <75 mg kg(-1) and a reassessment of this pathway. This paper presents a reevaluation of biosolids Mo data, refinement of the risk assessment algorithms, and a reassessment of Mo-induced hypocuprosis from land application of biosolids. Forage Mo uptake coefficients (UC) are derived from field studies, many of which used modern biosolids applied to numerous soil types, with varying soil pH values, and supporting various crops. Typical cattle diet scenarios are used to calculate a diet-weighted UC value that realistically represents forage Mo exposure to cattle. Recent biosolids use data are employed to estimate the fraction of animal forage (FC) likely to be affected by biosolids applications nationally. Field data are used to estimate long-term Mo leaching and a leaching correction factor (LC) is used to adjust cumulative biosolids application limits. The modified UC and new FC and LC factors are used in a new algorithm to calculate biosolids Mo Pathway 6 risk. The resulting numerical standards for Mo are cumulative limit (RPc)=40 kg Mo ha(-1), and alternate pollutant limit (APL) = 40 mg Mo kg(-1) We regard the modifications to algorithms and parameters and calculations as conservative, and believe that the risk of Mo-induced hypocuprosis from biosolids Mo is small. Providing adequate Cu mineral supplements, standard procedure in proper herd management, would augment the conservatism of the new risk assessment.  相似文献
