Statistical evaluation procedures for monitoring data at facilities permitted under the Resource Conservation and Recovery Act (RCRA) are frequently established before monitoring begins. Selecting the statistical method before background data have been collected often leads to the use of statistical procedures that are inappropriate for the actual monitoring data. Such was the case for unsaturated zone monitoring at a permitted land treatment unit in the Gulf Coast area of Texas. Due to the large number of “not detected” results in the background database for lysimeters, statistical evaluation procedures specified in the original RCRA permit yielded an artificially low standard deviation, resulting in background values that were strongly biased on the low side. An alternate statistical procedure based on probability plots was developed and was accepted by the state environmental regulatory agency. This technique, which has wide applicability for many types of environmental monitoring data, significantly reduced the chasing of false positives, thus saving potentially expensive investigations and remediations.  相似文献

A column containing four concentric layers of progressively finer-grained glass beads (graded column) was used to study the transport of the bacteriophage T7 in water flowing parallel to layering through a fining-upwards (FU) sedimentary structure. By passing a pulse of T7, and a conservative solute tracer upwards through a column packed with a single bead size (uniform column), the capacity of each bead type to attenuate the bacteriophage was determined. Solute and bacteriophage responses were modelled using an analytical solution to the advection-dispersion equation, with first-order kinetic deposition simulating bacteriophage attenuation. Resulting deposition constants for different flow velocities indicated that filtration theory-determined values differed from experimentally determined values by less than 10%. In contrast, the responses of solute and bacteriophage tracers passing upwards through graded columns could not be reproduced with a single analytical solution. However, a flux-weighted summation of four one-dimensional advective-dispersive analytical terms approximated solute breakthrough curves. The prolonged tailing observed in the resulting curve resembled that typically generated from field-based tracer test data, reflecting the potential importance of textural heterogeneity in the transport of dissolved substances in groundwater. Moreover, bacteriophage deposition terms, determined from filtration theory, reproduced the T7 breakthrough curve once desorption and inactivation on grain surfaces were incorporated. To evaluate the effect of FU sequences on mass transport processes in more detail, bacteriophage passage through sequences resembling those sampled from a FU bed in a fluvioglacial gravel pit were carried out using an analogous approach to that employed in the laboratory. Both solute and bacteriophage breakthrough responses resembled those generated from field-based test data and in the graded column experiments. Comparisons with the results of simulations using averaged hydraulic conductivities show that simulations employing averaged parameters overestimate bacteriophage travel times and underestimate masses recovered and peak concentrations.  相似文献

Isochrysis galbana Parke, Strain CCAP 927/1, was grown in ammonium-limited batch culture under a 12 h light: 12 h dark illumination cycle. Samples were taken every 12 h over the 26 d period from lag phase through exponential into stationary phase (no net carbon fixation), with more frequent sampling at points of interest. Exponential cell-specific growth rate was 0.3 to 0.4d-1. Cell division occurred during the dark phase, while cell volume increase, ammonium uptake, and pigment synthesis occurred during the light. Stationary phase cells were small, and the lag phase was long (5 d) even though the C:N ratio had returned from 18 to 6.5 within 2 d, followed by synthesis of chlorophyll a. Net chlorophyll synthesis ceased within 4 d of exhaustion of the nitrogen source. The chlorophyll c: chlorophyll a ratio remained constant during increasing nitrogen deprivation. Biovolume and carotenoids correlated with carbon biomass. Levels of chlorophyll a correlated poorly with carbon fixation and carbon biomass once the nitrogen source had been exhausted. Except after the addition of ammonium to nitrogen-deprived cells (refeeding), the content of intracellular glutamine and the glutamine: glutamate ratio were low during the dark phase, rising to a plateau within the first 1 h of illumination. Refeeding of cells which had only just exhausted the extracellular nitrogen source resulted in a much smaller increase in glutamine than refeeding of nitrogen-starved (stationary-phase) cells. Nitrogen biomass correlated with the presence of an unidentified intracellular amine.  相似文献

Despite the fact that coccolithophorids such as Emiliania huxleyi are suspected to play an important role in carbon-cycling, there are few data from which to deduce how these organisms may respond to CO2-induced global warming. the nitrogen and phosphorus nutrient physiology of these organisms, together with its interaction with photosynthesis, needs to be studied especially in connection with the selection for coccolith forming individuals and the quantity and quality of the CaCO3 deposited in the coccoliths. Without such data, it will not be possible to model the contributions that these microalgae may play in arresting the increase in levels of atmospheric CO2.  相似文献
