Effects of auxin as plant hormones are widespread; in fact in almost all aspects of plant growth and development auxin plays
a pivotal role. Although auxin is required for propagating cell division in plant cells, its effect upon cell division is
least understood. If auxin is depleted from the culture medium, cultured cells cease to divide. It has been demonstrated in
this context that the addition of auxin to auxin-starved nondividing tobacco BY-2 cells induced semisynchronous cell division.
On the other hand, there are some cell lines, named habituated cells, that can grow without auxin. The cause and reason for
the habituated cells have not been clarified. A habituated cell line named 2B-13 is derived from the tobacco BY-2 cell line,
which has been most intensively studied among plant cell lines. When we tried to find the difference between two cell lines
of BY-2 and 2B-13 cells, we found that the addition of culture filtrated from the auxin-habituated 2B-13 cells induced semisynchronous
cell division in auxin-starved BY-2 cells. The cell division factor (CDF) that is responsible for inducing cell division in
auxin-starved BY-2 cells was purified to near-homogeneity by sequential passage through a hydroxyapatite column, a ConA Sepharose
column and a Sephadex gel filtration column. The resulting purified fraction appeared as a single band of high molecular weight
on sodium dodecyl sulfate-polyacrylamide gel electrophoresis gels by silver staining and was able to induce cell division
in auxin-starved BY-2 cells. Identification of the protein by MALD-TOF-MS/MS revealed that it is structurally related to P-glycoprotein
from Gossypioides kirkii, which belongs to ATP-binding cassette (ABC)-transporters. The significance of CDF as a possible ABC-transporter is discussed
in relationship to auxin–autotrophic growth and auxin-signaling pathway. 相似文献

Serum levels of alpha-fetoprotein (AFP), human chorionic gonadotropin (hCG), and un-conjugated oestriol (uE3) were measured in serum samples of 4131 non-smoking and 1018 smoking women during the second trimester of pregnancy. The levels of all three analytes decreased with increasing body weight. The AFP median was significantly increased in smokers in a dose-response association; hCG decreased by 21 per cent and uE3 decreased by 3 per cent in smokers in a non-dose-related fashion. Regression functions for adjustment of serum levels for weight and smoking should be considered in risk estimation for Down syndrome in order to give a woman's individual risk more precisely. 相似文献

The Ecosystem Management Decision Support (EMDS) system is an application framework for knowledge-based decision support of ecological assessments. EMDS integrates geographic information system and knowledge base system technologies to provide an analytical tool for environmental assessment and monitoring. The basic objective of EMDS is to improve the quality and completeness of environmental assessments and the efficiency with which they are performed. The USDA Forest Service and the U.S. Environmental Protection Agency have cooperatively developed an EMDS knowledge base for watershed condition assessment. Specifically, this knowledge base evaluates watershed processes, patterns, general effects of human activity, and fisheries habitat suitability. Such assessments are based on spatially explicit input data concerning current conditions and reference conditions which are subsequently interpreted by user-defined "fuzzy" membership functions. Mature Lumbricus terrestris were host soils and leaf litter were collected from a former arsenic mine in Devon, UK (Devon Great Consols), a former gold mine in Ontario, Canada (Deloro), and an uncontaminated residential garden in Nottingham, UK. Arsenic concentrations determined by inductively coupled plasma-mass spectrometry (ICP-MS) in soils were 16-348 mg kg−1, 6.0-239 mg kg−1 in the earthworms and 8.6 mg kg−1 in leaf litter sampled at Deloro (all dry weight). High performance liquid chromatography (HPLC-ICP-MS) analysis revealed arsenite (AsIII), arsenate (AsV) and five organoarsenic species; arsenobetaine (AB), methylarsonate (MAV), dimethylarsinate (DMAV), arsenosugar 1 (glycerol sugar), arsenosugar 2 (phosphate sugar), and trimethylarsineoxide (TMAO) in field-collected L. terrestris. Differences were observed in the variety of organoarsenic species present between field sites. Several organoarsenic species were observed in the leaf litter (DMAV, arsenosugar 2 and TMAO) but not AB. Depuration resulted in higher concentrations of inorganic As being detected in the earthworm whereas the concentration or variety of organoarsenic species was unchanged. Commercially sourced L. terrestris were exposed to As contaminated soil in laboratory mesocosms (1.0, 98, 183, 236, 324 and 436 mg kg−1) without leaf litter and were additionally analyzed using X-ray absorption near edge structure (XANES). Only inorganic AsIII and AsV was observed. It is proposed that ingestion of leaf litter and symbiotic processes in the natural soil environment are likely sources of organoarsenic compounds in field-collected L. terrestris. 相似文献

An intensive field survey, with 6-h measurement intervals, of concentrations of chemical species in particulate matter and gaseous compounds was carried out at coastal sites on the Sea of Japan during winter. The concentration variation of SO2(g) and HNO3(g) were well correlated, whereas the NH3(g) concentration variation had no correlation with those of SO2(g) and HNO3(g). The NH4

Long distance dispersal may have important consequences for gene flow and community structure. The dispersal of many plants depends on transport by vertebrate seed dispersers. The shapes of seed shadows produced by vertebrates depend both on movement patterns of the dispersers and on the dynamics and effects of passage through the disperser’s gut (i.e. the retention time, survival and germination of ingested seeds). A combination of experiments with captive waterbirds and aquatic plant seeds was used to analyse the following: (a) the effects of inter- and intra-specific variation in seed size and duck species on seed retention time in the gut and (b) the relationship between retention time and the percent germination and germination rates of seeds. Among the three Scirpus species used, those with smaller seeds showed higher survival after ingestion by birds and longer retention times inside their guts than those with larger seeds. For Potamogeton pectinatus, only seeds from the smaller size class (<8 mg) survived ingestion. Retention time affected the percent germination and germination rate of Scirpus seeds but in a manner that varied for the different plant and bird species studied. We recorded both linear and non-linear effects of retention time on percent germination. In addition, germination rate was positively correlated with retention time in Scirpus litoralis but negatively correlated in Scirpus lacustris. Small seed size can favour dispersal over larger distances. However, the effects of retention time on percent germination can modify the seed shadows produced by birds due to higher percent germination of seeds retained for short or intermediate periods. The changes in dispersal quality associated with dispersal distance (which is expected to be positively related to retention time) will affect the probability of seedling establishment over longer distances and, thus, the spatial characteristics of the effective seed shadow. 相似文献
