The characterization of microbial communities of different depth sediment samples was examined by a culture-independent method and compared with physicochemical parameters, those are organic matter (OM), total nitrogen (TN), total phosphorus (TP), pH and redox potential (Eh). Total genomic DNA was extracted from samples derived from different depths. After they were amplified with the GC-341 f/907r primer sets of partial bacterial 16S rRNA genes, the products were separated by denaturing gradient gel electrophoresis (DGGE). The profile of DGGE fingerprints of different depth sediment samples revealed that the community structure remained relatively stable along the entire 45 cm sediment core, however, principal-component analysis of DGGE patterns revealed that at greater sediment depths, successional shifts in community structure were evident. The principle coordinates analysis suggested that the bacterial communities along the sediment core could be separated into two groups, which were located 0-20 cm and 21-45 cm, respectively. The sequencing dominant bands demonstrated that the major phylogenetic groups identified by DGGE belonged to Bacillus, Bacterium, Brevibacillus, Exiguobacterium, γ-Proteobacterium, Acinetobacter sp. and some uncultured or unidentified bacteria. The results indicated the existence of highly diverse bacterial community in the lake sediment core.  相似文献

为了解煤在中等温度(150~400℃)下氧化的特点,对贫煤和褐煤进行了系列恒温加热.重点考察在超过加热温度后煤的升温速率,以分析其氧化放热反应的特点.不同条件实验中,贫煤的升温速率曲线都在相同的一些温度点附近出现峰值;褐煤也有相同的规律.这表明在特定温度附近氧化反应加强是由煤自身的特性决定的.另一方面煤样的厚度越小、粒度越大都使得升温速率数值增大,说明厚度小、粒度大有利于氧气的输运,而导致氧化反应加快.  相似文献

Spatial and temporal temperature variations exist in a compost pile. This study demonstrates that systematic temperature sampling of a compost pile, as is widely done, tends to underestimate these variations, which in turn may lead to false conclusions about the sanitary condition of the final product. To address these variations, a proper scheme of temperature sampling needs to be used. A comparison of the results from 21 temperature data loggers randomly introduced into a compost pile with those from 20 systematically introduced data loggers showed that the mean, maximum and minimum temperatures in both methods were very similar in their magnitudes. Overall, greater temperature variation was captured using the random method. In addition, 95% of the probes introduced systematically had attained thermophilic sanitation conditions (?55 °C for three consecutive days), as compared to 76% from the group that were randomly introduced. Furthermore, it was found that, from a statistical standpoint, readings from at least 47 randomly introduced temperature loggers are necessary to capture the observed temperature variation. Lastly, the turning of the compost pile was found to increase the chance that any random particle would be exposed to the temperature ?55 °C for three consecutive days. One turning was done during the study, and it increased the probability from 76% to nearly 85%. Using the Markov chain model it was calculated that if five turnings had been implemented on the evaluated technology, the likelihood that every particle would experience the required time–temperature condition would be 98%.  相似文献

Experimental investigations were carried out to investigate the effect of thermo-chemical exposures on the hydraulic performance of Compacted Clay Liners (CCLs) in landfills. Hydraulic conductivity of most CCL specimens was increased by two to three times their initial values when exposed to 55 °C for 75 days. CCL specimens also experienced increases in their hydraulic conductivities when exposed to leachate at room temperature. This behaviour could be due to the decrease in viscosity when the permeant was changed from tap water to leachate. However, as the leachate exposure time exceeded the first 15 days, hydraulic conductivity readings decreased to as much as one order of magnitude after 75 days of leachate permeation at room temperature. The gradual decrease in the CCLs hydraulic conductivities was most likely due to chemical precipitation and clogging of pore voids within the soils which seemed to reduce the effective pore volume. The rate of hydraulic conductivity reduction due to leachate permeation was slower at higher temperatures, which was attributed to the lower permeant viscosity and lower clogging occurrence. The observed hydraulic behaviours were correlated to the physical, mineral, and chemical properties of the CCLs and described below.  相似文献

Rising temperatures, a widespread consequence of climate change, have been implicated in enigmatic amphibian declines from habitats with little apparent human impact. The pathogenic fungus Batrachochytrium dendrobatidis (Bd), now widespread in Neotropical mountains, may act in synergy with climate change causing collapse in thermally stressed hosts. We measured the thermal tolerance of frogs along a wide elevational gradient in the Tropical Andes, where frog populations have collapsed. We used the difference between critical thermal maximum and the temperature a frog experiences in nature as a measure of tolerance to high temperatures. Temperature tolerance increased as elevation increased, suggesting that frogs at higher elevations may be less sensitive to rising temperatures. We tested the alternative pathogen optimal growth hypothesis that prevalence of the pathogen should decrease as temperatures fall outside the optimal range of pathogen growth. Our infection‐prevalence data supported the pathogen optimal growth hypothesis because we found that prevalence of Bd increased when host temperatures matched its optimal growth range. These findings suggest that rising temperatures may not be the driver of amphibian declines in the eastern slopes of the Andes. Zoonotic outbreaks of Bd are the most parsimonious hypothesis to explain the collapse of montane amphibian faunas; but our results also reveal that lowland tropical amphibians, despite being shielded from Bd by higher temperatures, are vulnerable to climate‐warming stress. 据1998年7月11日的29个点的温度观测资料与观测点不同半径内绿地率和建筑率分别建立了线性回归关系,结果发现,温度与绿地率存在负相关关系,温度与建筑率存在正相关关系,早上8:00和下午14:00线性相关关系不显著,晚上20:00显著,但绿地范围太小时也不显著,而建筑物在较小范围内就存在显著的线性相关关系。  相似文献
