镉 (Cd) 是一种有毒重金属,可通过食物链传递到人体,对人体健康构成威胁。基于生理学的毒物动力学 (PBTK) 模型是量化人类暴露于环境元素的有用工具。基于PBTK模型,结合大面积区域调查数据,预测北方某小麦主产区居民因食用受Cd污染的食物而可能出现的尿镉 (U-Cd) 水平。研究结果显示,区域内57.8%小麦籽粒Cd含量超过国家食品安全限量标准 (0.1 mg·kg−1) 。区域人群中约35.1%的个体Cd摄入量超过JECFA的推荐安全值 (0.8 μg·(kg·d−1)) ,高达70.8%的个体超过EFSA的推荐值 (0.36 μg·(kg·d−1)) 。PBTK模型模拟结果显示,随着年龄增长,区域人群U-Cd含量呈上升趋势。70岁女性和男性U-Cd含量分别达到1.60 μg·g−1肌酐和0.80 μg·g−1肌酐。70岁女性U-Cd含量是男性的两倍且超过EFSA推荐的U-Cd含量安全限值 (1 μg·g−1肌酐) 。PBTK模型的应用能够预测不同暴露人群中U-Cd的累积水平,对于制定有效的区域Cd污染防治策略具有重要意义,并为食品安全风险评估提供科学依据。  相似文献

As important members of the zooplankton community and sources of food for fish, rotifers are used extensively in ecotoxicological research to assess the health of the environment and safety of compounds. However, most rotifer toxicity tests are only conducted using rotifer neonates derived from unexposed mothers, thus ignoring the potential transfer of contaminants from mother to offspring. To understand better the mother to offspring exposure, a multigenerational study was conducted using three successive generations (F0, F1 and F2) of the common freshwater rotifer Brachionus calyciflorus to investigate the toxic effects of the widely used organophosphate pesticide, dimethoate (O, O-dimethyl S-methylcarbamoylmethyl phosphorodithioate). When the F0 generation was exposed to five pesticide concentrations, the population growth rate (r) displayed symptoms of hormesis, characterized by the conversion of low-concentration stimulation to high-concentration inhibition. Despite this observation, the exposure to any given concentration of dimethoate reduced the population growth rates of the F1 and F2 generation rotifers. Significant differences existed between the F0, F1 and F2 rotifers for the population growth rate under dimethoate stress: F2 individuals were more sensitive than F1, whereas the F1 individuals were more sensitive than F0. The results indicated that the parental exposure to a given toxic stress could result in increased sensitivity and decreased fitness in the offspring. This study illustrates the utility of multigenerational toxicity tests, which may better reflect and more accurately predict the effects of long-term pesticide exposure to aquatic organisms at the population level.  相似文献

A cocktail of human pharmaceuticals pollute aquatic environments and there is considerable scientific uncertainty about the effects that this may have on aquatic organisms. Human drug target proteins can be highly conserved in non target species suggesting that similar modes of action (MoA) may occur. The aim of this work was to explore whether molecular docking offers a potential tool to predict the effects of pharmaceutical compounds on non target organisms. Three highly prescribed drugs, diclofenac, ibuprofen and levonorgestrel which regularly pollute freshwater environments were used as examples. Their primary drug targets are cyclooxygenase 2 (COX2) and progesterone receptor (PR). Molecular docking experiments were performed using these drugs and their primary drug target homologues for Danio rerio, Salmo salar, Oncorhynchus mykiss, Xenopus tropicalis, Xenopus laevis and Daphnia pulex. The results show that fish and frog COX2 enzymes are likely to bind diclofenac and ibuprofen in the same way as humans but that D. pulex would not. Binding will probably lead to inhibition of COX function and reduced prostaglandin production. Levonorgestrel was found to bind in the same binding pocket of the progesterone receptor in frogs and fish as the human form. This suggests implications for the fecundity of fish and frogs which are exposed to levonorgestrel. Chronic ecotoxicological effects of these drugs reported in the literature support these findings. Molecular docking may provide a valuable tool for ecotoxicity tests by guiding selection of test species and incorporating the MoA of drugs for relevant chronic test end points in environmental risk assessments.  相似文献

A chronic dietary risk assessment for pesticide residues was conducted for four age groups of the Argentinian population following the procedure recommended by the WHO. The National Theoretical Maximum Daily Intake (NTMDI) for 308 pesticides was calculated for the first time, using the Maximum Residue Limits (MRLs) from several Argentinean regulations and food consumption data from a comprehensive National Nutrition and Health Survey. The risk was estimated by comparing the TMDI with the Acceptable Daily Intakes (ADI) identified by various sources. Furthermore, for each of the compounds with a TMDI >65% of the ADI, a probabilistic analysis was conducted to quantify the probability of exceeding the ADI. In this study 27, 22, 10, and 6 active ingredients (a.i.) were estimated to exceed the 100% of the ADI for the different population groups: 6–23 month-old children, 2–5 year-old children, pregnant women, and 10–49 year-old women, respectively. Some of these ADI-exceeding compounds (carbofuran, diazinon, dichlorvos, dimethoate, oxydemeton-methyl and methyl bromide) were found in all four of these groups. Milk, apples, potatoes, and tomatoes were the foods that contributed most to the intake of these pesticides. The study is of primary importance for the improvement of risk assessment, regulations, and monitoring activities.  相似文献

To determine the population exposure to PM(10) in Chongqing, China, we developed an indirect model by combining information on the time activity patterns of various demographic subgroups with estimates of the PM(10) concentrations in different microenvironments (MEs). The spatial and temporal variations of the exposure to PM(10) were illustrated in a geographical information system (GIS). The population weighted exposure (PWE) for the entire population was 229, 155 and 211 microg/m(3), respectively, in winter, summer and as the annual average. Indoor PM(10) level at home was the largest contributor to the PWE, especially for the rural areas where high pollution levels were found due to solid fuels burning. Elder people had higher PM(10) exposure than adults and youth, due to more time spent in indoor MEs. The highest health risk due to particulate was found in the city zone and northeast regions, suggesting that pollution abatement should be prioritized in these areas.  相似文献

Organochlorine contamination in the Mediterranean coastal environment of Egypt was assessed based on 26 surface sediments samples collected from several locations on the Egyptian coast, including harbors, coastal lakes, bays, and estuaries. The distribution and potential ecological risk of contaminants is described. Organochlorine compounds (OCs) were widely distributed in the coastal environment of Egypt. Concentrations of PCBs, DDTs, and chloropyrifos ranged from 0.29 to 377 ng g−1 dw, 0.07 to 81.5 ng g−1 dw, and below the detection limit (DL) to 288 ng g−1 dw, respectively. Other organochlorinated pesticides (OCP) studied were 1–2 orders of magnitude lower. OCP and PCBs had higher concentrations at Burullus Lake, Abu Qir Bay, Alexandria Eastern Harbor, and El Max Bay compared to other sites. OCP and PCB contamination is higher in the vicinity of possible input sources such as shipping, industrial activities and urban areas. PCB congener profiles indicated they were derived from more than one commercially available mixture. The ratios of commercial chlordane and heptachlor metabolites indicate historical usage; however, DDT and HCHs inputs at several locations appear to be from recent usage. The concentrations of PCBs and DDTs are similar to those observed in sediments from coastal areas of the Mediterranean Sea. Ecotoxicological risk from DDTs and PCBs is greatest in Abu Qir Bay, Alexandria Harbor, and El-Max Bay.  相似文献

为明确酸雨条件下飞灰基胶凝固砷体的长期稳定性,模拟2种酸度的酸雨对3种粒径状态下的固砷体进行动态淋溶实验,并运用改进多级连续提取法(Sequential Extraction Procedure, SEP)和潜在风险评估指数对固砷体进行长期稳定性风险评价。结果表明,随着浸出时间的增加,固砷体中的Al3+溶出并与溶液中的OH反应生成Al(OH)3胶体,且废渣中的硫化矿物氧化生成H2SO4、Fe2(SO4)3等氧化剂,进一步加剧硫化物的氧化溶解,使得浸出液pH呈下降趋势;而且,由于飞灰固砷体粒径大小不同,表面积大小和吸附位点变化,使砷浸出过程呈现出初始、快速释放和慢速释放3个阶段的浸出特征,最高浸出质量浓度为2.42 mg·L−1,累计释放量达133.78 mg·kg−1,累计释放率为2.32%。SEP实验发现,原渣中的还原态砷大幅度降低,酸可提取态砷和残渣态砷增加,有利于降低固砷体的风险。潜在生态风险评价表明,在Ⅰ类和Ⅱ类建设用地筛选值为背景时,潜在生态危害程度由中等危害转为轻微危害。本研究结果可为飞灰固砷体在不同侵蚀作用下的安全处置提供参考。  相似文献

Abstract

A sudden increase in serum creatinine after paraquat intoxication has been reported in several clinical studies. However, this dramatic change of creatinine may be possibly due to an interconversion of creatine-creatinine in relation to paraquat toxicity. In order to investigate the creatine-creatinine relationship, a liquid chromatography tandem mass spectrometry in combination with electrospray ionization was developed and validated for simultaneous determination of creatine and creatinine in the serum. The chromatographic separation was achieved on a Gemini® C6-Phenyl column with a gradient elution consisting of 0.1% formic acid in ultrapure water and methanol as the mobile phase. The method yielded suitable levels of specificity and selectivity, and calibration curves of creatine and creatinine in serum were linear over the concentration range of 0.5–200?µg mL?1. The limit of quantification of both compounds was 0.5?µg mL?1, and the method was accurate within the recovery range of 96.23–102.75%, indicating the robustness of the method. The method was successfully applied to toxicological samples from paraquat-intoxicated patients, and the concentrations of creatine and creatinine were quantified. High creatine concentrations in serum samples were observed which may lead to high serum creatinine despite normal kidney function as creatine is converted to creatinine in proportion to its concentration.  相似文献
