以学校食堂餐饮废水为原料,通过发酵培养获得粘红酵母和地衣芽孢杆菌菌液,按照不同比例混合菌液制备微生物液体菌肥,在盆栽小油菜种植中检测其应用效果。利用餐饮废水发酵所得粘红酵母和地衣芽孢杆菌菌液pH为5.5~7.5,未检出杂菌且有效活菌数高于80亿/m L,N、P、K总养分大于4.6%,以体积比为4∶1混合施用于盆栽小油菜,其鲜重、叶绿素含量和过氧化氢酶活性与对照组相比分别提高了33.4%、34.1%和8.2%。  相似文献

Abstract

The larvicide activity of the experimental preparation SAN 402 I WDC containing B. Thuringiensis var. Israelensis and that of Abate 500 E (44 % temephos) were tested on the larvae of Aedes aegypti. The studies were performed on the following media : distilled water, pure and buffered at pH 9, 7 and 4 and pond water whether or not free from materials in suspension. The activity of both preparations is not influenced by the composition of the media, exception made for the presence of materials in suspension. The persistance varies according to the kind of the media and the concentration level. It is in general rather low. Both preparations behave rather similar, the microbial insecticide however is more influenced by the presence of materials in. suspension.  相似文献

Abstract

An industrial-scale, profitable method for production of the most widely used bioinsecticide, Bacillus thuringiensis (Bt), is challenging because of its widespread application. The aim of this study is to present a strategy to develop a low-cost, large-scale bioprocess to produce Bt H14.

This study was first focused on the design of a culture medium composed of economical and available components, such as glycerol and lysed Saccharomyces cerevisiae. The production goal of 1200 ITU was achieved using a medium composed of 20:20 g L?1of glycerol:lysed yeast in batch cultures. Efforts were subsequently focused on the design of an appropriate culture system, and an original two-stage culture system was proposed. First, yeast (the primary component of the culture medium) are cultivated using a minimal mineral medium and lysed, and in the second stage, Bt is cultivated in the same bioreactor using the lysed yeasts as culture medium (supplemented with a feeding pulse of 10 g L?1 glycerol). This system was called fed batch one pot (FOP). A new inoculation strategy is also presented in this study, since these Bt cultures were inoculated directly with heat pre-treated spores instead of vegetative bacteria to facilitate the bioprocess. This study was developed from the laboratory to production-scale bioreactors (measuring from 500 mL to 2500 L), and the efficiency of the proposed strategy was evident in LD50 tests results, achieving 1796 ITU in large-scale processes. Both the use of non-conventional sources and the process development for biomass production are important for cost-effective production of Bt-based insecticides in mosquito control projects.  相似文献

A study was conducted to investigate fenhexamid (FEX) behavior in soil and in water. FEX proved to be rather stable at acid pH but showed slight degradation at neutral and alkaline pH. After 101 days of FEX spiking of a soil sample, 94% at pH 4, 12% at pH 7 and 23% at pH 9 of the active ingredient was still present. In natural water the rate of FEX disappearance appeared to be slow which may be due to abiotic rather than biotic processes. The soil degradation tests showed low persistence of the active ingredient if a good microflora activity is guaranteed (DT50 about 1 day). Moreover, in absence of microorganisms, FEX proved to be stable. Humidities of 25 and 50% of Water Holding Capacity (WHC) influenced in equal measure the rate of degradation. From the same soil, a bacterium was isolated and identified as Bacillus megaterium, which was able to metabolize FEX with the hydroxylation of the cyclohexane ring. Moreover, FEX showed an elevated affinity for humic acid (73%), smectite (31%), and ferrihydrite(20%) and low affinity for vermiculite (11%) and kaolinite (7%).  相似文献

将来自苏云金芽胞杆菌李氏亚种的营养期杀虫蛋白基因vip83导入苏云金芽胞杆菌高毒菌株YBT-1520,得到广谱、高毒的工程菌BMB696B.室内生物测定显示该工程菌对棉铃虫和甜菜夜蛾具有很高的毒力.然而,国内外还未见有关于营养期杀虫蛋白的毒理学资料的报道,因此工程菌BMB696B的毒理学安全性也无法预测.为了对该工程菌的安全性进行合理的评价,并为该产品的生产和应用过程中安全防护措施的制定提供依据,根据国家标准GB 15670-1995<农药登记毒理学试验方法>,就BMB696B粉剂对实验动物进行了相关的毒理学试验.结果表明,工程菌对Wistar大鼠急性经口LD50大于5 000 mg/kg,急性经皮LD50大于2 000 mg/kg,均属低毒类;对兔皮肤刺激平均指数72 h后为0,皮肤刺激强度为无刺激性;眼刺激平均指数48 h后为O,眼刺激强度为无刺激性;对豚鼠致敏率为0,属Ⅰ级弱致敏物.由此证明,工程菌BMB696B没有经口和经皮的急性毒性、没有急性皮肤和眼刺激性、没有致敏性.  相似文献

Furfural is a toxic metabolic inhibitor that is created during the conversion of lignocellulose to produce fuel, which can retard fermentation and increase production costs. thus, it is important for lignocellulosic conversion that the ability of the strain to resist furfural stress be improved. A cellulose-degrading bacterium BREC-11 with tolerance to furfural was isolated from the intestinal tract of Omphisa fuscidentalis hampson larvae via the addition of furfural in the medium. Based on analyses of morphological observations, physiological and biochemical characterizations, and 16S rDNA sequences, strain BREC-11 was shown to represent a member of the genus Bacillus and was named B. siamensis BREC-11. to study the tolerance concentration of strain BREC-11, a wide range of furfural formaldehyde concentrations were tested and strain BREC-11 was shown to grow in the mineral medium containing furfural up to 3.5 g/L. Cellulase activity of strain BREC-11 was determined at the tolerable concentration of 3.5 g/L furfural after incubation at 30 ℃ and 150 r/min for 2 days. Results indicated that filter paper enzyme, CMC-Na enzyme, and β-glucosidase activity was 0.1 U/mL, 0.21 U/mL, and 0.07 U/mL, respectively. BREC-11 is a cellulose-degrading bacterium with resistance to furfural, which has potential application in future bio-refinery processes. © 2018 Science Press. All rights reserved.  相似文献

菌剂的使用是目前促进园林废弃物堆肥的有效方法,但常用的菌剂往往为液态菌剂,其制作存在无菌操作要求高、产品存在运输不便等问题,拟尝试通过固体菌剂加以克服.以芽孢杆菌(Bacillus sp.)B01为研究对象,对其固态发酵过程中的载体、保护剂、接种量、装瓶量等常见工艺参数进行优化,制成固体菌剂,并与市售常见液体菌剂——EM菌进行对比,探究其对园林废弃物堆肥过程的影响.结果表明:B01固态发酵的最优载体配比为麦麸35%、米糠45%、乳糖6%、黄豆粉6%、硅藻土8%,保护剂为5%海藻糖,接种量为15%,装瓶量为15%,料水比为1:1.在该发酵条件下,B01固态发酵物活菌体数可达到2.62×1010 CFU/g(以干质量计),存储90 d后,25℃下的存活率为30.19%.与EM菌相比,固体菌剂——B01可使园林废弃物堆肥过程中纤维素降解率提高11.04%、木质素降解率提高15.47%、腐殖质含量提高19.91%.研究显示,通过固态发酵条件的优化成功地将B01制成固体菌剂,该固体菌剂具有较高的活菌数量和较长的保质期,还可促进园林废弃物堆肥过程中木质素、纤维素的降解以及腐殖质的合成,具有显著的应用潜力.   相似文献

选用巨大芽孢杆菌(Bacillus megatherium)作为实验菌种,以餐厨垃圾湿热处理脱出液为发酵培养基制作解磷液态菌肥。将巨大芽孢杆菌接种于餐厨垃圾湿热处理脱出液中进行培养并确定最佳发酵时间,考察初始p H值、接种量、培养温度、摇床转速、装液量和脱出液与水的混合比例对活菌数的影响。结果表明:巨大芽孢杆菌在餐厨垃圾湿热处理脱出液中最佳发酵时间为36 h,发酵最佳初始p H值、接种量、培养温度、摇床转速、装液量和脱出液与水的混合比例分别为7.5、2%、30℃、210 r/min、50 m L(250 m L锥形瓶)和1∶1。巨大芽孢杆菌在餐厨垃圾湿热处理脱出液中进行培养后可达到农业部规定的液态菌肥的活菌数(2×108cfu/m L)标准。  相似文献

微生物胞外聚合物(Extracellular Polymeric Substances,EPS)在废水中重金属的吸附去除过程中起着非常重要的作用.苯胺黑药高效降解菌(Bacillus vallismortis)对苯胺黑药有良好的降解能力,但对其吸附重金属的性能研究还不充分.因此,本文采用3种方法提取苯胺黑药高效降解菌的EPS,主要考察了p H、温度、底物浓度和时间对重金属去除效果的影响.结果表明,热提法提取的效率较高;p H对金属离子吸附影响很大,当p H7时,随着p H变大吸附量逐渐升高,而温度对吸附量影响不大.EPS对Cu~(2+)、Zn~(2+)的去除为快速表面吸附过程,在第8 min时对Cu~(2+)、Zn~(2+)的去除率分别达到了90.7%、52.3%,EPS对Cu~(2+)、Zn~(2+)的吸附表观上符合拟二级动力学规律.在单一体系中,根据Langmuir方程计算出EPS对Cu~(2+)的最大吸附量为2.155 mg·mg-1,对Zn~(2+)的最大吸附量为0.508 mg·mg-1;Cu~(2+)吸附过程与Freundlich方程拟合效果较好,Zn~(2+)吸附过程与Langmuir方程拟合效果较好.红外光谱分析结果表明,EPS表面的羟基、氨基、酰胺基团、羧基和C—O—C基团都参与了吸附,且Cu~(2+)和Zn~(2+)的吸附位点基本一致,本文的研究结果对工程实践具有一定的理论指导意义.  相似文献
