We present the first case of a fetus with pure tetrasomy 20p proven by cord-blood sampling at 24 weeks of gestation. This case was diagnosed in utero with multiple congenital anomalies including occipital encephalocele, mega-cisterna magna, mesomelic shortening, and clubfeet. An analysis of GTG-banded chromosomes of 20 metaphase cells was performed. Female karyotype [47,XX, +i(20)(p10)] was revealed in all cells. Pure tetrasomy 20p was confirmed using fluorescent in situ hybridization (FISH) with a telomere probe for chromosome 20p in all seven metaphase cells. The pregnancy was terminated because of associated multiple anomalies and severe oligohydramnios. The postmortem examination confirmed the prenatal diagnosis. We report the prenatal diagnosis of a fetus with tetrasomy 5p mosaicism resulting from a supernumerary isochromosome for the short arm of chromosome 5. The pregnancy was terminated and fetal autopsy revealed minor facial abnormalities (long philtrum, up-slanting palpebral fissures and a broad nasal bridge). Cultures were established from fetal skin, kidney, and pancreas for cytogenetic analysis; the i(5p) was observed only in the skin fibroblasts. 以高磷污染河流沉积物为研究对象,利用液相31P核磁共振(31P-NMR)技术研究了沉积物中有机磷提取剂的选择和提取过程.结果表明:前提取以BD和EDTA效果最佳,TP和Po的提取量分别为(322.87±13.81)和(119.76±16.00)mg·kg-1、(24.91±15.25)和(20.90±2.58)mg·kg-1,2种提取剂均促进了NaOH的提取效率,前提取后NaOH提取液中TP和Po的提取量分别达到(2271.20±75.63)和(2477.40±112.71)mg·kg-1、(152.04±1.55)和(186.93±20.53)mg·kg-1;对顺磁性离子Fe和Mn而言,利用BD和EDTA+Na2S2O4作为前提取剂可以提取更多的Fe离子,提取液中的浓度分别为(5116.57±24.32)和(10677±215.61)mg·kg-1;EDTA和EDTA+Na2S2O4提取剂对Mn的去除效量较高,提取液中浓度分别为(243.02±2.80)和(266.61±1.61)mg·kg-1.液相31P-NMR图谱显示,上述提取剂处理后均可得到正磷酸盐、焦磷酸、磷酸单酯、磷脂和DNA 5种磷形态,其中EDTA可提高沉积物中正磷酸盐、焦磷酸盐、磷脂和DNA的含量.综合考虑:利用EDTA和NaOH两步提取可实现较高的Po提取量和较低的顺磁性离子干扰,并得到理想的核磁共振图谱,从而保证沉积物中有机磷液相31P-NMR分析的科学性和有效性. 相似文献

We describe the first case of mosaic supernumerary marker iso (8p) displaying a karyotype discordance between chorionic villi (CV) and amniotic fluid (AF) cultures during prenatal cytogenetic diagnosis. In the first trimester, cytogenetic analysis after chorionic villi sampling (CVS) was normal in all metaphases in the short-term cytotrophoblast cell culture, but an undefined supernumerary marker was detected in 60% of mesenchymal cells in the long-term CV culture. Informed of the mosaicism, the couple selected amniotic fluid sampling as a second-trimester confirmatory diagnostic procedure. The supernumerary marker was absent in all of the 25 available AF cells metaphases. The prospective parents received genetic counselling and were informed that the discordance could be interpreted as a placental confined mosaicism or as a true foetal mosaicism with low percentage of affected cells. The couple opted to continue the pregnancy. In the second month of life, the child had abnormal development with severe psychomotor delay and frequent episodes of epilepsy. Postnatal cytogenetic extensive re-evaluation discovered that the previously detected supernumerary marker was indeed an isochromosome (8p) rearrangement present at low frequency in 5% of the blood lymphocytes. The bioconcentration and distribution pattern of p,p′-DDT 1,1,1-1trichloro-2,2-bis(2-chlorophenyl-4-chlorophenyl)-ethane] and its main metabolites (p,p′-DDD [1,1-dichloro-2,2-bis (4-chlorophenyl) ethane] and p,p′-DDE [1,1-dichloro-2,2-bis (4-chlorophenyl) in adipose tissue, liver, brain, kidney, thymus, and testis were examined in young rats after 10 days of intraperitoneal injection of 50 and 100 mg of p,p′-DDT/kg of body weight. Analyses were performed by high-resolution gas chromatography. p,p′-DDT was found to be accumulated in a dose-dependent manner with the highest concentration in adipose tissue. However, in brain, the accumulation of pesticide was low and remained unchanged at the higher dose. This difference may relate to the protective role of the blood-brain barrier, which limits the access of the xenobiotic in the cerebral compartment, and to the differential tissue lipid composition. Although tissues concentration of p,p′-DDE and p,p′-DDD correlated positively to total p,p′-DDT levels, the active role in detoxification of pollutants may explain why p,p′-DDD is more abundant in liver than in the rest of organs. On the contrary, in brain, the concentration of p,p′-DDE is higher than that of p,p′-DDD, suggesting that the metabolism of the parent insecticide proceeds via more than one pathway. 相似文献

Soxhlet extraction of polychlorinated dibenzo‐p‐dioxins/dibenzofurans (PCDD/F) from sediment was studied using a designed experimental set‐up. Three variables were studied: three solvent combinations n‐hexane/methanol, dichloromethane/acetone and toluene/methanol; two extraction times, 8 and 16 h, and addition of bulk substances (sodium carbonate). The extraction efficiency was almost identical in all extractions, with the exception of extraction where toluene/ methanol was combined with sodium carbonate. In these samples a major effect was observed, due to formation of Hx‐, Hp‐ and OCDD isomers. Composition of formed PCDD was similar to PCDD isomers found in pentachlorophenol products. 相似文献

The molecular mechanisms underlying proliferation and apoptosis induced by p-nonylphenol (NP) through regulation of mRNA and protein expression of c-myc, p53 mRNA and the protein was investigated using ovarian cancer PEO4 cells. PEO4 cells were grown in the Dulbecco's modified Eagle medium (DMEM), 3(4,5-dimethylthiazol-2-yl) 2,5-diphenyltetrazolium bromide (MTT) assay was employed to investigate cell proliferation. RT-PCR and immunohistochemistry was performed to determine mRNA and protein expression of PEO4 c-myc and p53. Within the range of approximately 8–9×10?7 mol L?1, NP stimulated proliferation in PEO4 cells in a concentration-and time-dependent manner. RT-PCR and immunohistochemistry analyses revealed that treatment with 32 × 10?7 mol L?1 NP for 72 hr resulted in increased mRNA and protein expression of PEO4 c-myc and p53. Evidence indicates that NP exerted estrogenic actions and stimulated proliferation in estrogen responsive ovarian cancer PEO4 cells by up-regulation of c-myc mRNA and protein expression. 相似文献

The biodegradation of electrospun nano-fibers of poly(-caprolactone) (PCL) was initially investigated with respect to the environmental application of PCL non-woven fabrics, using pure-cultured soil filamentous fungi, Aspergillus oryzae, Penicillium caseicolum, P. citrinum, Mucor sp., Rhizopus sp., Curvularia sp., and Cladosporium sp. Three kinds of non-woven PCL fabrics with different mean fiber diameters (330, 360, and 510 nm) were prepared by changing the viscosities of the pre-spun PCL solutions (150, 210, and 310 cPs, respectively). All of the pure-line soil filamentous fungi tested grew on the two fiber materials. Electron microscopy was used to observe the biodegradation processes revealing remarkable growth of two fungi, Rhizopus sp. and Mucor sp., along with the accompanying collapse of the nano-fiber matrices. In the biochemical oxygen demand (BOD) test, the biodegradation of the 330 nm PCL nano-fibers by Rhizopus sp. and Mucor sp. exceeded 20 and 30% carbon dioxide generation, respectively. The biodegradability of the PCL non-woven fabrics decreased with the mean fiber diameter and the 330 nm PCL nano-fiber that was made from 150 cPs solution (concentration, 7 wt%) exhibited the highest biodegradability. These results might offer some clues for the applications of the PCL non-woven fabrics having the controlled biodegradability in the environmental uses. 相似文献
