Abstract

In an attempt to examine the chronic effect of low levels of cadmium on hepatic drug‐metabolizing enzyme system, an experiment was carried out in which growing male rats were given 0, 5, 10, and 2 0 ppm of cadmium in drinking water for a period of 8 weeks. An ip administration of a hypnotic dose of pentobarbital to the cadmium‐treated and the control rats 2 4 hr following the termination of the experiment exhibited that there was no significant difference in the drug metabolism in control and any of the treated groups. Next, liver microsomes were isolated from animals in all groups to study their ability to metabolize drugs in vitro. The results indicated that the activity of benzphetamine N‐demethylase and aniline hydroxylase, and the concentration of microsomal cytochrome P‐450 were almost identical in the control and treated groups. On the other hand, a single ip dose of cadmium (2 mg/kg) caused significant decrease in the in vivo and in vitro microsomal drug metabolism. These results suggest that although a single ip dose of cadmium (2 mg/kg) causes significant inhibition of drug metabolism, chronic exposure to cadmium up to 2 0 ppm in drinking water over a period of 8 weeks is unlikely to affect hepatic drug metabolism.  相似文献

Abstract

Deoxynivalenol (DON, vomitoxin), a Fusarium mycotoxin, is suspected of inducing its anorectic/feed refusal activity through a serotoninergic (5HT) mechanism, possible via 5HT2‐receptors. In this study the efficiency of cyproheptadine (CYP), a serotonin antagonist and known appetite stimulant, to attenuate the adverse effect of DON was investigated in mice. CYP was administered in the feed for two days before animals began receiving the DON, which was also added to the feed. Both agents were administered concurrently thereafter for a 12‐day period. Dosing levels included various combinations of the two compounds, ranging from 0–16 ppm DON and 0–20 ppm CYP.

Results showed that CYP could effectively offset the reduction in feed intake caused by dietary DON, but only when dose levels were optimized (CYP appeared to have a narrow effective dose range, which was also dependent on the DON concentration). In fact, optimum CYP doses were able to enhance feed intake in DON‐fed mice above control levels, indicating that more than just a direct block of the toxin's effect was occurring. Conversely though, the effect of CYP on weight gain (WG) was ambiguous. At the lower CYP doses (≤ 5 ppm), alone or in combination with the lowest DON level tested (4 ppm), a modest positive effect was noted, but at the higher DON concentrations (≥ 8 ppm) the overall WG generally remained depressed, and tended to be reduced further by increasing doses of CYP.

These results provide additional support that serotoninergic mechanisms probably play a role in mediating DON‐induced reduction in feed intake. Where and how DON actually initiates its anorectic effect, however, has not been resolved.  相似文献

Abstract

Human serum transferrin (HSTF), human serum albumin (HSA) and rat serum were compared for their interaction with AlCl3 , in a Tris‐HCl buffer solutions (pH 7.4). The AlCl3 was tested in series of concentrations in the range of 50 μM up to 500 μM . HSTF, HSA and their 1:1 mixture and rat serum were incubated at 37°C with series of AlCl3 concentrations. The protein profile of the incubated solutions were compared to control using SDS‐PAGE and FPLC tests. The results indicated that HSTF was more specifically responsive to AlCl3showing a characteristic increase in its UV absorption, peak and area dimensions. Simultaneously, HSA was less affected, but it showed a significant shift with an increase in molecular weight accompanied with a change in its profile. The respective bands of transferrin and albumin in rat serum behaved similarly. The SDS‐PAGE and FPLC data coincided and confirmed the preferential affinity of HSTF to bind with Al3+ . These results support the suggestion of using HSTF for monitoring levels of Al3+ in human blood samples of exposed population. The importance of further developing such a biomarker is the increased demand for early detection of the hazardous levels of Al3+ in relation to its long term neurotoxic adverse effects.  相似文献

Abstract

This study demonstrates that the growth of S. typhimurium in Luria Bertani broth supplemented with acetate, propionate, butyrate, or a mixture of the three SCFA, affected cell‐association and the ability to invade cultured HEp‐2 cells. Cell‐association and invasion was determined after growth for 4 h of growth in the presence of the SCFA at pH 6 and 7. The results suggest that the growth rate of the culture may have affected cell‐association and invasion since accompanying the significant decrease in growth rate in the presence of SCFA at pH 6 was a decrease in cell‐association and invasion. However, the results also suggest that the individual SCFA may play a role in modulating cell‐association and the invasion phenotype and the regulation of cell‐association and invasion by the SCFA was dependent on the concentration and the pH of the medium. Although the growth rates were similar for S. typhimurium in the SCFA mixture, butyrate (100 mM) and propionate (50 mM) at pH 6, differences in cell‐association and invasion were observed among these cultures. Also, at pH 7, differences were observed among the SCFA treatments even though the growth rates were similar.  相似文献

Abstract: A nontraditional application of the Hydrological Simulation Program – FORTRAN (HSPF) model to simulate freshwater discharge to upper Charlotte Harbor along Florida’s west coast was performed. This application was different from traditional HSPF applications in three ways. First, the domain of the model was defined based on the hydraulic characteristics of the landforms using small distributed parameter discretization. Second, broad wetland land forms, representing more than 20% of this area, were simulated as reaches with storage‐attenuation characteristics and not as pervious land segments (PERLNDs). Finally, the reach flow‐tables (F‐Tables) were configured in a unique way to be calibrated representing the uncertainty of the storage‐attenuation process. Characterizing wetlands as hydrography elements allows flow from the wetlands to be treated as a stage‐dependent flux. The study was conducted for the un‐gauged portion of the Peace and Myakka rivers in west‐central Florida. Due to low gradient tidal influences, a large portion of the basin is un‐gauged. The objective of this study was to simulate stream flow discharges and to estimate freshwater inflow from these un‐gauged areas to upper Charlotte Harbor. Two local gauging stations were located within the model domain and were used for calibration. Another gauge with a shorter period of record was used for verification. A set of global hydrologic parameters were selected and tested using the parameter optimization software (PEST) during the calibration. Model results were evaluated using PEST and well‐known statistical indices. The correlation coefficients were very high (0.899 and 0.825) for the two calibration stations. Further testing of this approach appears warranted for watersheds with significant wetlands coverage.  相似文献

Animal‐mediated seed dispersal is important for sustaining biological diversity in forest ecosystems, particularly in the tropics. Forest fragmentation, hunting, and selective logging modify forests in myriad ways and their effects on animal‐mediated seed dispersal have been examined in many case studies. However, the overall effects of different types of human disturbance on animal‐mediated seed dispersal are still unknown. We identified 35 articles that provided 83 comparisons of animal‐mediated seed dispersal between disturbed and undisturbed forests; all comparisons except one were conducted in tropical or subtropical ecosystems. We assessed the effects of forest fragmentation, hunting, and selective logging on seed dispersal of fleshy‐fruited tree species. We carried out a meta‐analysis to test whether forest fragmentation, hunting, and selective logging affected 3 components of animal‐mediated seed dispersal: frugivore visitation rate, number of seeds removed, and distance of seed dispersal. Forest fragmentation, hunting, and selective logging did not affect visitation rate and were marginally associated with a reduction in seed‐dispersal distance. Hunting and selective logging, but not fragmentation, were associated with a large reduction in the number of seeds removed. Fewer seeds of large‐seeded than of small‐seeded tree species were removed in hunted or selectively logged forests. A plausible explanation for the consistently negative effects of hunting and selective logging on large‐seeded plant species is that large frugivores, as the predominant seed dispersers for large‐seeded plant species, are the first animals to be extirpated from hunted or logged forests. The reduction in forest area after fragmentation appeared to have weaker effects on frugivore communities and animal‐mediated seed dispersal than hunting and selective logging. The differential effects of hunting and selective logging on large‐ and small‐seeded tree species underpinned case studies that showed disrupted plant‐frugivore interactions could trigger a homogenization of seed traits in tree communities in hunted or logged tropical forests. Despite widespread recognition of the major threat to tropical forest biological diversity and local food security posed by unsustainable bushmeat hunting, virtually no long‐term studies tracking the socioecological dynamics of hunting systems have been conducted. We interviewed local hunters and collected detailed hunting data to investigate changes in offtake and hunter characteristics over 10 years (2001–2010) in Dibouka and Kouagna villages, central Gabon, in the context of hunter recollections of longer term trends since the 1950s. To control for changes in hunter behavior, such as trap location and characteristics, we report hunting offtake data per trap. Our results suggest the hunting area was already highly depleted by 2001; local hunters reported that 16 large‐bodied prey species had become rare or locally extirpated over the last 60 years. Overall, we observed no significant declines in hunting offtake or changes in species composition from 2001 to 2010, and offtakes per trap increased slightly between 2004 and 2010. However, trapping distance from the villages increased, and there was a switch in hunting techniques; a larger proportion of the catch was hunted with guns in 2010. The number of hunters declined by 20% from 2004 to 2010, and male livelihood activities shifted away from hunting. Hunters with the lowest hunting incomes in 2004 were more likely than successful hunters to have moved away from the village by 2010 (often in response to alternative employment opportunities). Therefore, changes in trap success (potentially related to biological factors) were interacting with system‐level changes in hunter number and composition (related to external socioeconomic factors) to produce a relatively static overall offtake. Our results highlight the importance of understanding the small‐scale context of hunting to correctly interpret changes or apparent stasis in hunting effort and offtake over time. Conservation conflicts are increasing on a global scale and instruments for reconciling competing interests are urgently needed. Multicriteria decision analysis (MCDA) is a structured, decision‐support process that can facilitate dialogue between groups with differing interests and incorporate human and environmental dimensions of conflict. MCDA is a structured and transparent method of breaking down complex problems and incorporating multiple objectives. The value of this process for addressing major challenges in conservation conflict management is that MCDA helps in setting realistic goals; entails a transparent decision‐making process; and addresses mistrust, differing world views, cross‐scale issues, patchy or contested information, and inflexible legislative tools. Overall we believe MCDA provides a valuable decision‐support tool, particularly for increasing awareness of the effects of particular values and choices for working toward negotiated compromise, although an awareness of the effect of methodological choices and the limitations of the method is vital before applying it in conflict situations. Abstract: Assessment of abundance, survival, recruitment rates, and density (i.e., population assessment) is especially challenging for elusive species most in need of protection (e.g., rare carnivores). Individual identification methods, such as DNA sampling, provide ways of studying such species efficiently and noninvasively. Additionally, statistical methods that correct for undetected animals and account for locations where animals are captured are available to efficiently estimate density and other demographic parameters. We collected hair samples of European wildcat (Felis silvestris) from cheek‐rub lure sticks, extracted DNA from the samples, and identified each animals’ genotype. To estimate the density of wildcats, we used Bayesian inference in a spatial capture‐recapture model. We used WinBUGS to fit a model that accounted for differences in detection probability among individuals and seasons and between two lure arrays. We detected 21 individual wildcats (including possible hybrids) 47 times. Wildcat density was estimated at 0.29/km2 (SE 0.06), and 95% of the activity of wildcats was estimated to occur within 1.83 km from their home‐range center. Lures located systematically were associated with a greater number of detections than lures placed in a cell on the basis of expert opinion. Detection probability of individual cats was greatest in late March. Our model is a generalized linear mixed model; hence, it can be easily extended, for instance, to incorporate trap‐ and individual‐level covariates. We believe that the combined use of noninvasive sampling techniques and spatial capture‐recapture models will improve population assessments, especially for rare and elusive animals.  相似文献

Abstract

Effects of the herbicide metsulfuron‐methyl on soil microorganisms and their activities in two soils were evaluated under laboratory conditions. Measurements included their populations, soil respiration, and microbial biomass. In the clay soil, bacterial populations decreased with increasing concentration of metsulfuron‐methyl during the first 9 days of incubation but exceeded that of the control soil from day 27 onward. In the sandy loam soil, the herbicide reduced bacterial populations during the first 3 days after application, but these increased to the level of untreated controls after 9 days’ incubation. Fungal populations in both soils increased with increasing metsulfuron‐methyl concentrations, especially in the sandy loam soil. CO2 evolution was stimulated in both soils in the presence of the herbicide initially, but decreased during days 3 to 9 of the incubation period before increasing again afterward. The presence of metsulfuron‐methyl in the soil increased microbial biomass, except in sandy loam soil at the first day of incubation.  相似文献
