苯并[a]芘和菲对缢蛏血细胞DNA损伤的研究   总被引:1,自引:0,他引:1  
为研究苯并[a]芘和菲对缢蛏的毒性效应,将缢蛏(Sinonovacula constricta)分别暴露于浓度为0.45 mg·L-1、0.15 mg·L-1、0.05 mg·L-1苯并[a]芘溶液和0.45 mg·L-1、0.15 mg·L-1、0.05 mg·L-1菲的溶液中,采用单细胞凝胶电泳实验(彗星实验)技术检测不同暴露时间缢蛏血淋巴细胞的DNA损伤程度,对照组为清洁海水。结果显示,高浓度(0.45 mg·L-1)苯并[a]芘溶液和(0.45 mg·L-1)菲溶液在短期(7 d)内即可导致缢蛏血细胞显著的DNA损伤,并且随苯并芘[a]和菲浓度的增大和暴露时间的延长,DNA损伤程度增加。21 d恢复实验后,各浓度组DNA损伤又均有不同程度的恢复,但中高浓度组(0.45 mg·L-1和0.15 mg·L-1)与对照组仍显著性差异。两种多环芳烃物质对缢蛏血细胞的DNA损伤作用均存在较显著时间-剂量-效应关系。其中,苯并芘[a]对缢蛏血细胞的DNA损伤作用要高于菲。  相似文献

为研究太原市冬季灰霾天气下大气PM2.5对肺泡巨噬细胞(AM)的毒性作用,采用采集于2011年12月30—31日的灰霾PM2.5悬浮液体外处理大鼠AM(终浓度分别为0、33、100、300μg·m L-1),用MTT法检测细胞活力,用酶标仪测定胞内超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)及Ca2+浓度,并用流式细胞仪测定胞内活性氧(ROS)和细胞凋亡状况.结果表明:随着PM2.5浓度增大,AM存活率、SOD和GSH-Px活性下降,MDA及ROS含量和Ca2+浓度升高,均呈现剂量-效应关系,细胞早期凋亡率也随着染毒浓度的增加呈现升高趋势,揭示了太原市灰霾PM2.5可使AM产生氧化应激,引起细胞脂质过氧化损伤和凋亡发生.  相似文献

The aim of this in vitro study was to examine the secretion activity, markers of proliferation and apoptosis in porcine ovarian granulosa cells (GCs) after deoxynivalenol (DON) addition. Ovarian granulosa cells were incubated with DON for 24h: 10, 100 and 1000 ng/mL, while the control group received no DON. The secretion of insulin-like growth factor I (IGF–I) and progesterone was determined by radioimmunoassay (RIA) and expression of cyclin B1, PCNA and caspase-3 by immunocytochemistry. IGF–I release by GCs was inhibited by DON, while progesterone release and the expression of cyclin B1 was stimulated by DON (at 1000 ng/mL but not at 10 and 100 ng/mL). PCNA expression was stimulated by DON (at 100 and 1000 ng/mL but not at 10 ng/mL). Caspase-3 expression was not influenced by DON treatment (at all doses). In conclusion, our results indicate, (1) a direct effect of DON on secretion of growth factor IGF-I and steroid hormone progesterone, (2) expression of markers of proliferation (cyclin B1 and PCNA) but not on the (3) expression of marker of apoptosis (caspase-3) in porcine ovarian granulosa cells. This in vitro study suggests the dose-dependent association of DON on porcine ovarian functions.  相似文献

以去除碳酸根后的镁铝水滑石正电胶团簇(LDHEPCL)作骨架,合成新型水滑石接枝污泥处理乳剂.用硫酸铈作引发剂,通过程序升温,实现了油包水多面液胞(W/O PHC)中丙烯酰胺(AM)、二甲基二烯丙基氯化铵(DMDAAC)与去除碳酸根后的LDHEPcL接枝聚合,制得污泥处理乳剂--LDHEPCL-PAM-PDMDAAC共聚物乳剂.聚合升温速度为2.5℃/min,最高温度为(985±1)℃.各配料最佳摩尔比为LDHEPCL:AM:DMDAAC=0.15:0.57:0.28.与目前最好的污泥处理乳剂FLOPAMR EMZ35及STCPA EM6040相比,LDHEPCL-PAM-PDMDAAC共聚物乳剂具有絮团致密、絮层高度低、与聚合铝(PAC)及羧甲基壳聚糖(CMC)配伍较好等优点.  相似文献

Microbial fuel cells (MFC) utilize microbes as catalysts to convert chemical energy to electricity. Inocula used for MFC operation must therefore contain active microbial population. The dye reduction-based electron-transfer activity monitoring (DREAM) assay was employed to evaluate different inocula used in MFCs for their microbial bioelectrical activity. The assay utilizes the redox property of Methylene Blue to undergo color change from blue to colorless state upon microbial reduction. The extent of Methylene Blue reduction was denoted as the DREAM assay coefficient. DREAM assay was initially performed on a microbial culture along with the growth curve and estimation of colony forming units (CFUs). DREAM coefficient correlated to the CFU/mL obtained over time as growth progressed. The assay was then extended to water samples (domestic sewage, lake and a man-made pond) serving as inocula in MFCs. Domestic wastewater gave the highest DREAM coefficient (0.300 ± 0.05), followed by pond (0.224 ± 0.07) and lake (0.157 ± 0.04) water samples. Power density obtained conformed to the DREAM coefficient values, with the three samples generating power densities of 46.45 ± 5.1, 36.12 ± 3.2 and 25.08 ± 4.3 mW/m2 respectively. We have also studied the role of addition of various carbon sources and their concentrations towards improving the sensitivity of the assay. The DREAM assay is a rapid, easy-to-perform and cost-effective method to assess inocula for their suitability as anolytes in terms of electron transfer potential in MFCs.  相似文献

Background, aim, and scope  It has been known that the pollutants of electronic wastes (E-wastes) can lead to severe pollution to the environment. It has been reported that about 50% to 80% of E-wastes from developed countries are exported to Asia and Africa. It has become a major global environmental problem to deal with ‘E-wastes’. E-waste recycling has remained primitive in Jinghai, China. This not only produces enormous environmental pollution but also can bring about toxic or genotoxic effects on the human body, threatening the health of both current residents and future generations living in the local environment. The concentration of lead in the blood of children in the E-waste polluted area in China is higher than that of the control area. But little is known about the cytogenetic effect to human beings caused by the pollution of E-wastes. In the present study, experiments have been performed to investigate the genetics of permanent residents of three villages with numerous E-waste disposal sites and to analyze the harmful effects of exposure to E-wastes. Materials and methods  In total, 171 villagers (exposed group) were randomly selected from permanent residents of three villages located in Jinghai County of Tianjin, China, where there has been massive disposal of E-wastes. Thirty villagers were selected from the neighboring towns without E-waste disposal sites to serve as controls. Chromosomal aberrations and cytokinesis blocking micronucleus were performed to detect the cytogenetic effect, dic + r (dicentric and ring chromosome), monomer, fragments (acentric fragments, minute chromosomes, and acentric rings), translocation, satellite, quadriradial, total aberrations, and micronuclear rate were scored for each subject. DNA damage was detected using comet assay; the DNA percentage in the comet tail (TDNA%), tail moment (TM), and Olive tail moment (OTM) were recorded to describe DNA damage to lymphocytes. Results  The total chromosome aberration rates (5.50%) and micronuclear rates (16.99%) of the exposure group were significantly higher than in the control group (P = 0.000). The percentage of DNA in the comet tail, tail moment, and Olive tail moment detected by comet assay showed that there was a significant difference in DNA damage in the exposure group (P = 0.000). The chromosome aberration, micronucleus rate, and DNA damage observed in women were significantly higher than those in men. Chromosome aberration and micronuclear rates of both smokers and non-smokers in the exposure group are obviously higher than that in the control group (P = 0.000). Discussion  The use of outdated (and unsafe) ways to deal with E-wastes can lead to exposure to a variety of substances harmful to human health. The components of pollution may enter the human body through the air, drinking water, and food chain to damage human genetic material, resulting in genomic instability. The rates of chromosomal aberration, micronucleus formation, and the degree of DNA damage in women in the group exposed to electronic waste were significantly higher than in men. The reason for this may be concerned with the traditional lifestyle of the local residents or the difference of sensitivity to the exposure to E-wastes or any others. Further investigations are needed to provide evidence to demonstrate this. Conclusions  Here, we report the obviously cytogenetic toxicity to the exposure population by the E-waste pollution for the first time. E-waste pollution may be a potential agent of genetic mutation, and may induce cytogenetic damage within the general population exposed to the pollution. These findings need to be considered, and steps should be taken to protect the current population and future generations from the effects of pollution with E-wastes. Recommendations and perspectives  The above results remind us that the impact of E-waste recycling on environmental quality of Jinghai should be evaluated soon. Moreover, it is urgent for the government to prohibit E-waste import and its processing by outdated ways. The future studies such as pollutant details of drinking water, air, and soil in the area as well as epidemiological investigations on the harmful effect to children must be performed eagerly. All the data available do provide a compelling case for immediate action in both countries to address workplace health and safety and waste management. 模拟了车用工况下燃料电池发动机脉冲式排氢的特点,设计开发了一套质子交换膜燃料电池的尾气净化系统,并以5 kW燃料电池的尾气排放为例,研究其净化效果。研究了电池阳极排放氢气缓冲前后尾气中氢气浓度的变化,以及不同空速条件下氢气的去除效果。结果表明:电堆阳极排放氢气经过缓冲处理后,尾气中氢气浓度趋于平稳,大部分工况下处在2%以下,达到了安全处理的要求;尾气中氢气的去除效果与空速密切相关。对自制的整体式催化剂而言,当空速低于20 000 h-1时,氢气的去除率能达到约95%,而当空速达到39 270 h-1时,氢气的去除率仅为10.9%。鉴于这一问题,提出通过采用尾排空气的分流手段调整催化燃烧反应器对空速的要求,以提高尾气中氢气的去除率。  相似文献

Oral ingestion of contaminated soil is an important pathway of human exposure to hydrophobic organic contaminants (HOCs), particularly for children in developing countries. The mobilization potential of various contaminants from ingested soil is often characterized using an in vitro gastrointestinal model, based on the quantities of contaminants remaining in digestive fluid after digestion and separation. Recently, it was experimentally demonstrated that a large fraction of mobilized contaminants sorbed on the digested residue could be released if the dissolved fraction was removed by intestinal absorption. This hypothesis was further tested in this study. Soil spiked with dichlorodiphenyltrichloroethane and its metabolites (DDXs) and polycyclic aromatic hydrocarbons (PAHs) was digested using an in vitro gastrointestinal model. A human colon carcinoma cell line (Caco-2) was cultured in digestive fluid with or without soil residue (pre-equilibrated with the soil) for 2 h. A large proportion of the contaminants (37-68%) was sorbed on the digested residue. Without this residue, 66 ± 13% of DDXs and 73 ± 14% of PAHs dissolved in the fluid, as means and standard deviations, were absorbed by the cell monolayer after exposure. With both digestive fluid and residue, the sorbed fraction of PAHs and DDXs decreased by 38-92%, while the ratios of the cellular to the dissolved concentrations were 2.7-2.8 times higher than those without the residue. This supported the hypothesis that the cell absorption of dissolved HOCs induces desorption of the sorbed fraction from digestive residue, and the desorbed HOCs can be absorbed as well.  相似文献
