Penicillium oxalicum SL2 (SL2) is a previously screened Pb-tolerant fungus that can promote crops growth. The relationship between SL2 colonization and Pb immobilization was studied to provide a theoretical basis for microbial remediation of Pb-contaminated paddy soil. In this study, green fluorescent protein (GFP) labeled SL2 was inoculated into different Pb-contaminated paddy soils (S1-S6). The Pb extracted from the soil by HNO3, EDTA and CaCl2 were used to characterize the available Pb. The results showed that the colonization of SL2 was divided into lag phase (0-7 days), growth phase (7-30 days), and mortality phase (30-90 days). SL2 colonized well in sandy soils rich in clay and total phosphorus with initial pH of 4.5-7.0. In addition, SL2 increased soil pH and decreased soil Eh, which was beneficial to immobilize Pb. In different soils, the highest percentages of CaCl2-Pb, EDTA-Pb, and HNO3-Pb immobilized by SL2 were 34.34%-40.53%, 17.05%-20.11%, and 7.39%-15.62%, respectively. Pearson correlation analysis showed that the percentages of CaCl2-Pb and EDTA-Pb immobilized by SL2 were significantly positively correlated with the number of SL2 during the growth phase. SL2 mainly immobilized Pb in the growth phase and a higher peak number of SL2 was beneficial to the immobilization of Pb.  相似文献

A pilot-scale Orbed oxidation ditch was operated for 17 months to optimize nitrogen removal from domestic wastewater of average COD to total nitrogen ratio of 2.7, with particular concern about the roles of dissolved oxygen (DO), mixed liquor suspended solids (MLSS) and return activated sludge (RAS) recycle ratio. Remarkable simultaneous nitrification and denitrification (SND) was observed and mean total nitrogen (TN) removal efficiency up to 72.1% was steadily achieved, at DO concentration in the out, middle and inner channel of 0.1, 0.4 and 0.7 mg/L, respectively, with an average M LSS of 5.5 g/L and RAS recycle ratio of 150%. Although the out channel took the major role in TN removal, the role of middle channel should never be ignored. The denitrification potential could be fully developed under low DO, high MLSS with adequate RAS ratio. The sludge settleability was amazingly improved under low DO operation mode, and some explanations were tried. In addition, a scries of simplified batch tests were done to determine whether novel microorganisms could make substantial contribution to the performance of nitrogen removal. The results indicated that the SND observed in this Orbal oxidation ditch was more likely a physical phenomenon.  相似文献

Abstract: The fitness of species with little genetic diversity is expected to be affected by inbreeding and an inability to respond to environmental change. Conservation theory suggests that endangered species will generally demonstrate lower genetic diversity than taxa that are not threatened. This hypothesis has been challenged because the time frame of anthropogenic extinction may be too fast to expect genetic factors to significantly contribute. I conducted a meta‐analysis to examine how genetic diversity in 894 tetrapods correlates with extinction threat level. Because species are not evolutionarily independent, I used a phylogenetic regression framework to address this issue. Mean genetic diversity of tetrapods, as assessed by protein heterozygosity, was 29.7–31.5% lower on average in threatened species than in their nonthreatened relatives, a highly significant reduction. Within amphibians as diversity decreased extinction risk increased in phylogenetic models, but not in nonphylogenetic regressions. The effects of threatened status on diversity also remained significant after accounting for body size in mammals. These results support the hypothesis that genetic effects on population fitness are important in the extinction process.  相似文献

Pollen pollution and allergy are becoming prominent issues in China. However, few studies on pollinosis have been reported. As an allergen in the atmosphere, allergenic Humulus scandens pollen was collected from four districts of Shanghai, including Wusong (WS), Jiading (JD), Xujiahui (XJH) and Songjiang (SJ). The mass concentrations of SO2, NO2, O3, PM10, and PM2.5 (particulate matter with air dynamic diameter less than 10 and 2.5 µm, respectively) near the four sampling sites were also recorded during Humulus scandens pollen season. The allergenicity of the Humulus scandens pollen was assessed by using of a rat model and enzyme linked immunosorbent assay (ELISA). Relationships between the allergenicity and air pollutants were correlated. Our results demonstrated that the biological viability of the pollens collected from the four districts exhibited no significant differences. ELISA and dot blotting results further demonstrated that the serum of sensitized rats exhibited much higher immune-reactive response than that of control groups. Western blotting showed that the 15 KD (1KD = 1000 dalton) proteins of Humulus pollen led to the allergic response. The allergenic intensity of Humulus pollen protein from different samples followed the pattern: WS > JD > XJ > SJ. There was a negative relationship between the allergenicity of Humulus pollens and PM10 (R = -0.99) / PM2.5 (R = -0.73), and a positive relationship with O3 (R = 0.92). These data clearly showed that PM10 and PM2.5 could enhance Humulus pollen protein release, and O3 could aggravate the allergenicity of the Humulus pollen.  相似文献

Metabolic reactions of and exposed to a halon concentration of 10 ppb over a period of 26 and 45 days, respectively were investigated. The response of both plant species to the exposure was a slight increase of the protein contents and of major pigments. A large increase of up to 200% was observed in the activity of the detoxifying enzyme glutathione S-transferase in fumigated plants. These physiological changes, namely the increase of the glutathione S-transferase activity, are interpreted as detoxification reactions.  相似文献

研究了15℃、20℃、25℃、30℃和35℃五个不同温度条件下盾叶薯蓣一些生理参数的变化.结果显示,盾叶薯蓣叶片的POD活性和MDA含量的变化呈V字形,其中在25℃时处于最低点;可溶性蛋白含量的变化呈M字形,可溶糖含量的变化呈W字形.在15℃、20℃、30℃和35℃下, 5d内,随着时间的延长,MDA含量增加,POD活性、可溶性蛋白和可溶性糖的含量下降;在25℃下,盾叶薯蓣的POD活性以及MDA、可溶性蛋白和可溶性糖的含量变化较小,相对稳定,所以25℃是最为适宜的生长温度. 图1表1参20  相似文献
