The influence of culture conditions on the ultrastructure and enzyme activities of amniotic fluid cells are reported. Morphological changes were determined as a function of the number of lysosomal-like inclusion bodies per cell, and these results correlated to the activity of Thiexosaminidase, a-mannosidase, β-glucuronidase, arylsulphatase C and 5′ nucleotidase. The parameters examined were pH of the culture media, type of media, increasing cell passage and day of harvest. Our results indicate that enzyme activities are less sensitive to changes in culture conditions as compared to ultrastructural changes. We therefore recommend that in order to obtain reliable ultrastructural results for the diagnosis of storage disorders, cultures should be grown in MEM as the culture medium, the pH of the medium carefully monitored to remain below pH 7·4, examining the cultures no later than the eighth cell passage and no later than the 10th day after subculture.  相似文献

Amniotic fluid cells obtained by transabdominal amniocentesis at 15–17 weeks of gestation and cultured for 15 and 21 days were separated into three fractions by density gradient centrifugation in Percoll. Each fraction (or peak) corresponded to the following densities: peak A, 1·02–1·03 g/ml; peak B, 1·04–1·05; peak C, 1·05–1·06. Peak A was composed of both non-viable and viable cells; the latter adopted the morphology of epithelial cells in culture. Peak B contained a mixture of fibroblasts and epithelioid cells and peak C had only epithelioid cells. The variability of N-acetyl-β-hexosaminidase and alkaline phosphatase activities was reduced in peaks B and C as compared to that of peak A and of unseparated cells suggesting that more defined and homogeneous cell types for enzymatic determinations can be obtained by centrifugation in Percoll density gradient. In prenatal diagnosis of biochemical defects, the separation of cells would permit a more precise diagnosis by eliminating enzyme variability due to the presence of different cell types or non-viable cells.  相似文献

We studied a family at risk for atypical TSD in which the index case showed, clinically, a late onset and a gradual psychomotor deterioration and biochemically, a residual hex. A activity in leucocytes. Two prenatal diagnoses of affected fetuses were made in this family, The first one on amniotic cells, the second one on trophoblast biopsy samples. Both of them were confirmed after abortion on cultured cells. Prenatal diagnosis of TSD, even of some atypical forms is possible using trophoblast biopsy, but formal confirmation should be obtained on cultured trophoblasts.  相似文献

Microbial fuel cells (MFCs) have become a promising technology for wastewater treatment accompanying electricity generation. Carbon and nitrogen removal can be achieved by utilizing the electron transfer between the anode and cathode in an MFC. However, large-scale power production and high removal efficiency must be achieved at a low cost to make MFCs practical and economically competitive in the future. This article reviews the principles, feasibility and bottlenecks of MFCs for simultaneous carbon and nitrogen removal, the recent advances and prospective strategies for performance improvement, as well as the involved microbes and electron transfer mechanisms.  相似文献

Epidemiological studies have demonstrated the exacerbation of respiratory diseases following sandstorm-derived particulate matter(PM) exposure.The presence of anthropogenic and biological agents on the sandstorm PM and the escalation of PM 2.5 μm(PM2.5)pollution in China have led to serious concerns regarding the health effects of PM2.5during Asian sandstorms.We investigated how changes in PM2.5composition,as the weather transitioned towards a sandstorm,affected human airway epithelial cells.Six PM2.5samples covering two sandstorm events and their respective background and transition periods were collected in Baotou,an industrial city near the Gobi Desert in China.PM samples from all three periods had mild cytotoxicity in human bronchial epithelial cell line BEAS-2B,which was positively correlated with the contents of polycyclic aromatic hydrocarbons and several metals.All PM samples potently increased the release of interleukin-6(IL-6) and interleukin-8(IL-8).Endotoxin in all samples contributed significantly to the IL-6 response,with only a minor effect on IL-8.Cr was positively correlated with both IL-6 and IL-8 release,while Si was only associated with the increase of IL-6.Our study suggests that local agricultural and industrial surroundings in addition to the sandstorm play important roles in the respiratory effects of sandstorm-derived PM.  相似文献

This paper examines the realism of the resilience ambition and process of the U.S. housing system, shedding light on its heterogeneity as well as the financialization currently acting as the driving force in real estate production. The resilience ambition leading to enhanced justice and egalitarianism is understood as the provision and maintenance of post-disaster housing for all within an institutionally diverse landscape of housing policy makers and implementers. Particular emphasis is given to the post-Katrina institutional transformations resulting from multifarious interactions between multilevel institutional structures and a diverse landscape of low-income housing policy implementers – referred as social resilience cells (SRCs) in this paper. The nature and level of these transformations determine the degree to which resilience in its heterogeneous form has been incubated in New Orleans. The paper concludes with a discussion on the macro conditions and bottom-linked governance structures under which all SRCs could be better bolstered in a post-financialization, radicalised neowelfare U.S., and which in turn create possibilities for materialising the resilience ambition.  相似文献

NOx emission is produced during combustion of fuels at high temperature. Excessive release of NOx causes several effects on living organisms and environment. In this work, the efforts to reduce NOx emission by developing electrochemically activated cells (EACs) for a diesel engine fuelled with diesel and biodiesel fuel are discussed. EAC technique is vital after treatment technology attempted in this work to simultaneous control of NOx, HC, and PM emissions. In this method, two types of EACs were developed. The CuO–YSZ electrolyte and CuO–YSZ electrolyte with BaO coating were developed and tested with diesel and biodiesel exhaust. Compared with diesel fuel, use of biodiesel fuel increased NOx emission by 11% and PM emission was slightly reduced with biodiesel, which was due to the presence of fuel bond oxygen content in biodiesel. The investigation has demonstrated low-temperature activation of the EACs at 250–350°C which was due to the addition of CuO to YSZ. In this work, maximum NOx reduction was achieved for CuO–YSZ cells with BaO NOx storage and the simultaneous control of HC and PM emission also was observed in this technique. NOx reduction by EAC is a vital technique and can be retrofitted with any diesel engine for emission reduction.  相似文献

探讨了2,3,7,8-四氯二苯并-ρ-二噁英(TCDD)在体外对雌激素依赖性乳腺癌细胞MCF-7端粒酶活性的影响.用0.5μmol·L-1的2,3,7,8-四氯二苯并-ρ二噁英(TCDD)作用于MCF-7细胞,裂解后提取MCF.7细胞端粒酶.采用BCA方法定量蛋白、重复序列扩增-银染法检测端粒酶活性.结果表明.TCDD作用MCF-7细胞后,细胞端粒酶活性增强,且随作用时间端粒酶活性减弱,但明显高于对照组(P<0.05).结果表明:TCDD有显著增强MCF-7细胞端粒酶活性的作用.  相似文献
