石油污染地下水中细菌多样性研究   总被引:2,自引:4,他引:2  
采集了某废弃炼油厂的石油污染地下水样品,提取水中微生物总DNA,构建细菌16S rDNA克隆文库,并通过16S rDNA序列的系统发育分析,对样品中的细菌种群多样性以及群落结构进行了研究.结果表明,文库中阳性克隆的16S rDNA序列分属10个细菌类群,分别为γ-Proteobacteria(49.1%)、α-Proteobacteria(12.9%)、β-Proteobacteria(11.1%)、Bacteroidetes(9.2%)、Verrucomicrobia(6.7%)、Acidobacteria(2.5%)、δ-Proteobacteria(1.2%)、Actinobacteria(1.2%)、Planctomycetes(0.6%)、Unidentifiedbacteria(5.5%).在这一生态系统中,γ-Proteobacteria类细菌占据主导地位,接近50%,尤其是假单胞菌属(Pseudomonas)微生物在文库中的比例达35.6%.该石油污染地下水样品中细菌与许多其它已知的降解菌亲缘关系较近,如鞘胺醇单胞菌(Sphingomonas)、红球菌(Rhodococcus)和短波单胞菌(B...  相似文献

一株高效脱硫脱氨氮菌的分离、鉴定及系统发育分析   总被引:2,自引:1,他引:1  
从运行稳定的能同步脱臭的曝气生物滤池中采集样品,富集分离获得一株能高效脱硫脱氨氮的菌株TS-1。对分离菌株进行形态观察、生理生化试验及16S rRNA基因序列分析,结果表明,该菌株TS-1为革兰氏阳性菌,杆状;菌落在营养琼脂培养基上呈圆形,表面光滑,乳白色半透明;V-P试验阴性,能水解淀粉和明胶,利用柠檬酸盐生长;对菌株进行16S rRNA的PCR扩增,扩增产物测序结果表明分离菌株TS-1与巨大芽孢杆菌(Bacillus megaterium)同源性达到99%;以16S rRNA同源性为基础构建了包括24株相关种属细菌在内的系统发育树,在系统发育树中,分离菌株TS1与Bacillus megaterium在同一分支。结合形态观察、生理生化试验及16S rRNA基因序列分析结果,将其初步鉴定为巨大芽孢杆菌(Bacillus megaterium)。在常温(30±2)℃、转速为150 r/min的条件下,处理pH 7、S2-和NH+4-N分别为80 mg/L和88 mg/L的水样40 h,硫化物和氨氮的脱除率分别为91.8%和96.6%。  相似文献

活性污泥体系中,聚糖菌(GAOs)在厌氧环境下与聚磷菌(PAOs)形成对底物的竞争关系,对聚糖菌的研究对于优化生物除磷工艺有重要意义。以葡萄糖为惟一碳源,在磷限制条件下,利用特殊运行方式对活性污泥进行驯化培养出了稳定的聚糖菌颗粒污泥,厌氧阶段磷释放量与有机物吸收量浓度(mg/L)比从7.4%下降为0.25%。从培养好的活性污泥反应器中分离获得2株聚糖菌,经菌落PCR和16S rRNA序列分析确定了所得聚糖菌菌株G1和菌株G2分别是枯草芽孢杆菌(Bacillus subtilis)和解鸟氨酸克雷伯氏菌(Klebsiella ornithinolytica)。  相似文献

It is widely accepted that tobacco smoke is responsible for the vast majority of lung cancers worldwide. There are many known and suspected carcinogens present in cigarette smoke, including α-emitting radioisotopes. Epidemiologic studies have shown that increased lung cancer risk is associated with exposure to ionizing radiation, and it is estimated that the majority of smoking-induced lung cancers may be at least partly attributable to the inhaled and deposited radiation dose from radioisotopes in the cigarette smoke itself. Recent research shows that silencing of the tumor suppressor gene p16INK4a (p16) by promoter methylation plays a role in smoking-related lung cancer. Inactivation of p16 has also been associated with lung cancer incidence in radiation-exposed workers, suggesting that radionuclides in cigarette smoke may be acting with other compounds to cause smoking-induced lung cancer. We evaluated the mechanism of ionizing radiation as an accepted cause of lung cancer in terms of its dose from tobacco smoke and silencing of p16. Because both radiation and cigarette smoking are associated with inactivation of p16, and p16 inactivation has been shown to play a major role in carcinogenesis, ionizing radiation from cigarette smoke likely plays a role in lung cancer risk. How large a role it plays, relative to chemical carcinogens and other modes of action, remains to be elucidated.  相似文献

Aerobic granule is a special microbial aggregate associated with biofilm structure. The formation of aerobic granular sludge is primarily depending on its bacterial community and relevant microbiological properties. In this experiment, a strain with high microbial attachment was isolated from aerobic granular sludge, and the detailed characteristics were examined. Its high attachment ability could reach 2.34 (OD600nm), while other low attachment values were only around 0.06-0.32, which indicated a big variation among the different bacteria. The strain exhibited a very special morphology with many fibric fingers under SEM observation. A distinctive behaviour was to form a spherical particle by themselves, which would be very beneficial for the formation and development of granular sludge. The EPS measurement showed that its PN content was higher than low attachment bacteria, and 3D-EEM confirmed that there were some different components. Based on the 16S rRNA analysis, it was identified to mostly belong to Stenotrophomonas. Its augmentation to particle sludge cultivation demonstrated that the strain could significantly promote the formation of aerobic granule. Conclusively, it was strongly suggested that it might be used as a good and potential model strain or chassis organism for the aerobic granular sludge formation and development.  相似文献

杂环化合物喹啉是焦化等工业废水中的一种难降解化合物.有关喹啉微生物降解的研究还很有限,分离筛选多样的喹啉降解细菌,对认识喹啉的降解机制和强化废水中喹啉的降解具有重要意义.本研究通过富集和多种分离条件的培养,从焦化废水活性污泥及唪啉驯化的生物膜样品中分离获得56株与喹啉降解相关的菌株,以16S rDNA双酶切方法进行ARDRA分型,将这些菌株分为12个OTU.选取部分代表菌株进行16S rDNA测序分析,表明分离所获得的菌株主要是Ochrobactrum、Bacillus、Pseudomonas和Rhodococcus属的微生物.通过对部分菌株测定其喹啉降解能力.发现大部分菌株都能以喹啉为唯一碳源进行生长并高效降解喹啉,极少数菌株不能单独降解喹啉.降解喹啉的Ochrobactrum属菌株还未见报道.  相似文献

亚硝酸盐型甲烷厌氧氧化(nitrite-dependent anaerobic methane oxidation,N-DAMO)是新近发现的生物反应,是偶联碳氮循环的关键环节,是环境领域和微生物领域的重大发现.N-DAMO的发现对于完善碳氮生物地球化学循环、丰富微生物学内容和研发新型生物脱氮除碳工艺均具有巨大的推动作用.催化N-DAMO反应的微生物为Candidatus Methylomirabilis oxyfera(M.oxyfera),其隶属于一新发现的细菌门——NC10门.近年来,M.oxyfera的生物学研究取得了许多突破性进展,如初步探明了其个体形态特征、细胞化学组分特征、富集培养特征、生理生化特征及生态学特征,最突出的例子包括发现了M.oxyfera独特的细胞(星状)形态及特殊的脂肪酸(10Me C16∶1Δ7)组分等.最近,N-DAMO的机制研究方面也有了突破性进展:发现了地球上第4种生物产氧途径.目前认为,M.oxyfera具有内产氧功能,其首先将NO-2还原为NO,然后将2分子NO进行歧化反应生成N2和O2,最后利用生成的O2对甲烷进行氧化.本文系统地介绍了M.oxyfera各方面的微生物特性.  相似文献

Wildlife management is limited by uncontrolled and often unrecognized environmental variation, by limited capabilities to observe and control animal populations, and by a lack of understanding about the biological processes driving population dynamics. In this paper I describe a comprehensive framework for management that includes multiple models and likelihood values to account for structural uncertainty, along with stochastic factors to account for environmental variation, random sampling, and partial controllability. Adaptive optimization is developed in terms of the optimal control of incompletely understood populations, with the expected value of perfect information measuring the potential for improving control through learning. The framework for optimal adaptive control is generalized by including partial observability and non-adaptive, sample-based updating of model likelihoods. Passive adaptive management is derived as a special case of constrained adaptive optimization, representing a potentially efficient suboptimal alternative that nonetheless accounts for structural uncertainty.  相似文献

According to a rights view it is acceptable to kill animals if they are innocent threats or shields or are in a lifeboat situation. However, according to advocates of such a view, our practices of killing animals for food or scientific research may be morally unacceptable. In this paper we argue that, even if we grant the basic assumptions of a rights view, a good deal of killing of animals for food and scientific research continues to be morally acceptable.  相似文献

用采自山东胜利油田的石油污染水体,以原油为唯一碳源,经人工海水培养基富集培养,得到lO株细菌.通过降解率的测定,发现其中一株细菌HB-1具有较强的石油降解能力,200 r/min振荡培养6 d,其原油的降解率达54.74%.根据菌落特征、菌体形态、生理生化特性和16S rDNA序列分析,确定HB-1为不动杆菌属(Acinetobacter sp.).进一步实验表明,该菌株最适宜牛长和降解石油的条件为:温度25~32℃,初始pH值6.5~7.5,盐度3%,此条件符合实际海洋环境的要求.图6表2参22  相似文献
