The activity of four enzymes, including GOT-1, has been investigated in the erythrocytes of a 10q→24qter trisomic fetus. Analyses have been performed on a feto-maternal blood mixture sampled by fetoscopy and on red cells obtained by cardiac puncture, following therapeutic abortion. The demonstration of a 40 percent increase of GOT-1 activity, as compared to normal fetuses of similar gestational age, suggests that gene dosage studies may be a useful confirmatory technique in prenatal diagnosis of unbalanced chromosomal aberrations. Practical application of a similar diagnostic approach is conditioned by (1) precise characterization of fetal chromosome imbalance; (2) confirmed assignment of the gene locus coding for the gene product under investigation; (3) evidence of a linear proportionality between gene dose and concentration of the gene product in patients with the same chromosome imbalance detected in the fetus; (4) knowledge of the range of normal variation at different weeks of gestation of the enzyme activity to be tested in the fetus; (5) safety of fetal sampling procedure.  相似文献

The biodegradation of electrospun nano-fibers of poly(-caprolactone) (PCL) was initially investigated with respect to the environmental application of PCL non-woven fabrics, using pure-cultured soil filamentous fungi, Aspergillus oryzae, Penicillium caseicolum, P. citrinum, Mucor sp., Rhizopus sp., Curvularia sp., and Cladosporium sp. Three kinds of non-woven PCL fabrics with different mean fiber diameters (330, 360, and 510 nm) were prepared by changing the viscosities of the pre-spun PCL solutions (150, 210, and 310 cPs, respectively). All of the pure-line soil filamentous fungi tested grew on the two fiber materials. Electron microscopy was used to observe the biodegradation processes revealing remarkable growth of two fungi, Rhizopus sp. and Mucor sp., along with the accompanying collapse of the nano-fiber matrices. In the biochemical oxygen demand (BOD) test, the biodegradation of the 330 nm PCL nano-fibers by Rhizopus sp. and Mucor sp. exceeded 20 and 30% carbon dioxide generation, respectively. The biodegradability of the PCL non-woven fabrics decreased with the mean fiber diameter and the 330 nm PCL nano-fiber that was made from 150 cPs solution (concentration, 7 wt%) exhibited the highest biodegradability. These results might offer some clues for the applications of the PCL non-woven fabrics having the controlled biodegradability in the environmental uses.  相似文献

A simple and rapid in vitro test was designed for the assessment of the biodegradation of polyester-based plastics by selected biodegrading bacterial strains. Variovorax paradoxus LMG 16137 was used for the degradation of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) and Acidovorax avenae subsp. avenae LMG 17238 fo the synthetic-based polyesters poly(-caprolactone) (PCL), poly(butylene succinate-co-butylene adipate), and a starch-PCL blend. Degradation by the bacteria was studied in liquid medium with the plastics (films, granules, and injection-molded test bars) as sole sources of carbon. Degradation was followed through gravimetry, growth of the culture, and tensile testing. The effects of incubation time, inoculum density, aeration, incubation temperature, and pH of the medium on the mass loss were investigated and conditions optimized. The test allowed to obtain reproducible results on the mass loss of plastic samples in less than 3 weeks and yielded excellent partially degraded samples for further analysis.  相似文献

Biodegradable hydrogels prepared by -irradiation from microbial poly(amino acid)s are reviewed. pH-sensitive hydrogels were prepared by means of -irradiation of poly(-glutamic acid) (PGA) produced byBacillus subtilis IFO3335 and poly(-lysine) (PL) produced byStreptomyces albulus in aqueous solutions. The preparation conditions, swelling equilibria, hydrolytic degradation, and enzymatic degradation of these hydrogels were studied. 以相同Al13含量、不同浓度的PAC为原料,利用乙醇-丙酮混合溶剂法、SO4^2-/Ba^2 沉淀置换法以及柱层析法分离提纯纳米Al13形态,同时利用Al-Ferron逐时络合比色法、^27Al-NMR等多种现代分析手段进行分析鉴定,比较其分离效果,并采用烧杯实验法和透光率脉动检测技术对Al13形态以及PAC、AlCl3的絮凝效果及絮凝过程中絮集物形成和增长的变化差异作了对比性研究.实验结果表明,乙醇.丙酮混合溶剂法对Al13分离纯化效果最好,分离所得Al13平均粒径大,聚合程度高.柱层析法受柱体积的影响只对低浓度的PAC有较好的分离效果;SO4^2-/Ba^2 沉淀置换法虽具有较好的分离效果,但分离过程有其他杂质离子的加入,影响到产品品质.混凝效果和动态实验结果表明,Al13形态是在絮凝过程中起电中和作用的主要形态,具有较强的除浊及脱色能力,在混凝过程中当絮体受到剪切力的破坏时Al13形态比PAC和AlCl3具有更强重新絮凝的能力.  相似文献

采用电解法合成了聚合氯化铝溶液 ,并采用2 7AlNMR法、透射电镜等方法对其形态特征进行了研究 .2 7AlNMR法表明在电化学制备PAC中主要含有铝单体 ,铝的二聚体及Al13形态 .Al13含量与其溶液的碱化度 (B)有关 .在总铝浓度 (AlT)为 2 .0mol L ,B为 2 .2时Al13含量达到最高 ,占AlT 的 70 .2 % ,并在溶液中较稳定存在 .透射电镜实验结果表明 ,电解制备的PAC的粒度分布均匀 ,无团聚现象产生  相似文献

We report an anencephalic fetus with acrania, cervicodorsal rachischisis, and a 46,X,del(X)(p22·1) karyotype. Necropsy revealed a left diaphragmatic hernia, ipsilateral lung hypoplasia, and intestinal malrotation. In order to investigate the characteristics of pure Nano-Al13, Nano-Al13 was separated and purified from a series of poly-aluminum chloride (PAC) solutions which had the same Al13 percentage but different total Al concentrations, by using column chromatography, ethanol-acetone resolving and SO2− 4/Ba2+ displacement. The Al13 species yield was characterized by Al-ferron timed complexation spectrophotometry and 27Al-NMR (nuclear magnetic resonance). The coagulation efficiency of Nano-Al13, PAC and AlCl3 in synthetic water was also investigated by Jar tests. The dynamic process and aggregation state of kaolin suspensions coagulating with Nano-Al13, PAC and AlCl3 were similarly investigated using a photometric dispersion analyzer 2000 (PDA2000). The experimental results indicated that the ethanol-acetone resolving method was simple and could separate the PAC solution at different concentrations, while column chromatography could separate PAC solutions at low concentrations. The SO4 2−/Ba2+ displacement method could separate PAC solutions at high concentrations. However, extra inorganic cation and anion could be added in the solution during separation. The coagulation efficiency and dynamic experimental results showed that Nano-Al13 with high positive-charged species was effective in removing turbidity and color. The dynamic process results showed that Nano-Al13 also had the best recovery capability after shearing compared with PAC and AlCl3 because the Nano-Al13 conformation is more effective in charge neutralization.
