In this study, a bacterial strain, CH-1, capable of degrading 3-bromocarbazole (3-BCZ) was isolated from a polluted soil. Based on its physio-biochemical characteristics and 16S rRNA genes, strain CH-1 was identified as a Stenotrophomonas sp. Strain CH-1 was able to degrade 70% of 50 mg/L 3-BCZ within 8 d at pH 7.0 and 30°C in mineral salt medium (MSM). During the process, the main intermediate metabolite was identified as (2E, 4Z)-6-(2-amino-5-bromophenyl)-2-hydroxy-6-oxhexa-2, 4-dienoic by gas (2E, 4Z)-6-(2-amino-5-bromophenyl)-2-hydroxy-6-oxhexa-2,4-dienoic via gas chromatograph-mass spectrometry (GC-MS) analysis. The metabolite disappeared after 14 d, suggesting that the metabolite can also be degraded by strain CH-1. 3-BCZ is a new persistent organic pollutant. This is the first report of the biodegradation of 3-BCZ. The results indicated that strain CH-1 may be a promising bacterial candidate for the bioremediation of environments polluted with polyhalogenated carbazoles (PHCs).  相似文献

Abstract

Bacillus spp. have long been used as biocontrol agents because of their efficient broad-spectrum antimicrobial activity. We identified a novel strain of Bacillus atrophaeus, named JZB120050, from soil. B. atrophaeus JZB120050 had a strong inhibitory effect against Botrytis cinerea and many other phytopathogens. Gas chromatography-mass spectrometry showed that B. atrophaeus JZB120050 produced many secondary metabolites, such as alkanes, alkenes and acids; some of which were related to pathogen inhibition. Enzyme activity analysis showed that B. atrophaeus JZB120050 secreted cell-wall-degrading enzymes, including chitinase, glucanase and protease, which degraded fungal cell walls. Both the novel glucanase gene bglu and chitinase gene chit1 were cloned and heterologously expressed in Escherichia coli and the products showed strong enzyme activity. In addition, B. atrophaeus JZB120050 secreted siderophores and formed a significant biofilm. Future studies should focus on these antimicrobial factors to facilitate widespread application in the field of agricultural biocontrol.  相似文献

研究了非离子型表面活性剂Triton X-100(TX-100)和Tween80(TW-80)对苯并[a]芘的增溶特性及对苯并[a]芘高效降解菌Bacillus pumilus strain Bap9生长的影响,结果表明,2种表面活性剂对苯并[a]芘均有良好的增溶效果,均能作为碳源和能源被菌株Bap9所利用,TX-100增溶能力和增殖能力相对更强;不同浓度的TX-100对菌株降解苯并[a]芘的影响不同,当浓度为1 000 mg/L时,对降解的促进作用最强,可将苯并[a]芘降解率提高20.8%;在苯并[a]芘降解过程中,TX-100亦能作为碳源被菌株Bap9利用,不产生二次污染,因此可用于苯并[a]芘污染环境的生物修复。  相似文献

根据ISO 11268-1:1993方法,完成了土壤中苯并[a]芘对赤子爱胜蚓(Eisenia fetida)的人工污染模拟实验.并采用Clontech PCR-SelectTM cDNA Subtration Kit构建了污染组和对照组蚯蚓之间的抑制消减文库.经测序和比对分析,在苯并[a]芘诱导上调文库中发现5个线粒体DNA(mtDNA)编码的基因,且在上调文库中具有较高的频率.它们分别为:细胞色素c氧化酶亚基Ⅰ、Ⅱ、Ⅲ,NADH氢脱氢酶亚基1,ATP合成酶亚基.研究表明,土壤中苯并[a]芘污染改变了蚯蚓线粒体基因组的表达水平,线粒体基因组表达异常是多环芳烃污染胁迫的重要分子生物标记物.  相似文献

Air pollution is emerging as one of the most important stresses, particularly in urban areas, of both developed and developing countries. It severely affects plant growth and development besides being a health hazard to humans and animals. Delhi has the dubious distinction of being one of the most polluted cities in the world. Incidentally, it is also one of the greenest cities with ongoing programs to increase the already considerable forest cover and biodiversity parks. Plants often act as a sink for airborne pollutants, including particulate matter. Hence it is important to evaluate their efficacy in absorbing pollutants and use the promising species for urban plantation. In the present study, the stress tolerance of four common trees of Delhi (Psidium guajava, Alstonia scholaris, Murraya koenigii, and Moringa oleifera), growing in three areas with varying pollution levels, was estimated by measuring Plant Secondary Metabolites (PSMs: total phenols, flavonoids, alkaloids, and tannins) along with the total chlorophyll content of the leaves. P. guajava showed significant increase in the contents of phenols, tannins, and alkaloids with higher Air Quality Index (AQI). Increased amounts of phenols and flavonoids were observed in A. scholaris, those of phenols and alkaloids in M. koenigii, and only of alkaloids in M. oleifera. There was a decrease in the total chlorophyll content of the leaves of all four species investigated. P. guajava and A. scholaris can be recommended for plantation along the sides of roads with heavy traffic movement. Since PSMs are seen to accumulate as a defense mechanism against air pollution too, as in various abiotic and biotic stresses, they can be used as biomarkers for stress tolerance in plants.  相似文献

Hundreds of millions of people around the world are exposed to elevated concentrations of inorganic and organic arsenic compounds, increasing the risk of a wide range of health effects. Studies of the environmental fate and human health effects of arsenic require authentic arsenic compounds. We summarize here the synthesis and characterization of more than a dozen methylated and thiolated arsenic compounds that are not commercially available. We discuss the methods of synthesis for the following14 trivalent(Ⅲ) and pentavalent() arsenic compounds: monomethylarsonous acid(MMA~Ⅲ), dicysteinylmethyldithioarsenite(MMA~Ⅲ(Cys)_2), monomethylarsonic acid(MMA~Ⅴ),monomethylmonothioarsonic acid(MMMTAⅤ) or monothio-MMA~Ⅴ, monomethyldithioarsonic acid(MMDTA~Ⅴ) or dithio-MMA~Ⅴ, monomethyltrithioarsonate(MMTTA~Ⅴ) or trithio-MMA~Ⅴ,dimethylarsinous acid(DMA~Ⅲ), dimethylarsino-glutathione(DMA~Ⅲ(SG)), dimethylarsinic acid(DMA~Ⅴ), dimethylmonothioarsinic acid(DMMTA~Ⅴ) or monothio-DMAⅤ, dimethyldithioarsinic acid(DMDTA~Ⅴ) or dithio-DMA~Ⅴ, trimethylarsine oxide(TMAO~Ⅴ), arsenobetaine(AsB), and an arsenicin-A model compound. We have reviewed and compared the available methods,synthesized the arsenic compounds in our laboratories, and provided characterization information. On the basis of reaction yield, ease of synthesis and purification of product, safety considerations, and our experience, we recommend a method for the synthesis of each of these arsenic compounds.  相似文献

本文以一株筛自胶州湾沉积物中的好氧反硝化细菌Zobellellasp.B307为研究对象,在短期暴露条件下,通过细菌的生长、脱氮能力,相关酶活以及代谢途径等指标的变化,研究纳米氧化锌(ZnO-NPs)对该菌株的毒性效应;结合锌离子溶出试验、CAT和ROS等氧化应激水平测定,探讨ZnO-NPs对该菌株的致毒机制.结果表明,200mg/L的ZnO-NPs会使菌株硝酸盐氮去除率降至57.53%,LDH升高至对照组的378%,ROS水平高达对照组的5.34倍,SOD活性比对照组升高了60.32%,NIR活性仅为对照组的14.46%;ZnO-NPs主要通过诱导菌株活性氧的生成使其膜通透性改变、相关酶活性下降,并使相关蛋白质、氨基酸的合成及基因表达等代谢通路受到影响,进而抑制该菌株的反硝化能力;游离锌离子的产生可能不是ZnO-NPs对菌株的主要致毒途径.  相似文献

为探究官能团修饰对微塑料生物效应的影响规律和机制,分析了 3种聚苯乙烯微塑料(PS、PS-NH2和PS-COOH)在斑马鱼(Danio rerto)胚胎内的累积特征,并利用微生物组学和代谢组学相结合的分析技术,解析了斑马鱼幼鱼体内代谢功能及微生物群落的响应规律.结果显示,3种微塑料都可以在斑马鱼体内产生累积,其含量范围为143~175 μg·g-1,且不存在明显差异性.斑马鱼胚胎绒毛膜是微塑料早期暴露的主要累积场所,但不能有效阻止小粒径微塑料的穿透.未修饰的PS明显影响了斑马鱼幼鱼体内氨基糖苷类的代谢能力,PS-NH2主要对氨基酸的代谢过程产生了影响,而PS-COOH则主要作用于三羧酸循环、氨基酸及糖酵解等代谢通路.3种微塑料均导致了斑马鱼幼鱼体内代谢功能的变化,从而对斑马鱼产生了毒性效应,而微塑料的官能团修饰则加强了这一影响程度.与对照组相比,PS-NH2明显降低了斑马鱼幼鱼体内的微生物群落的多样性,显著增加了变形菌门(Proteobacteria)的占比,导致斑马鱼幼鱼体内的菌群失衡,从而引起鱼体代谢功能的紊乱.微塑料的官能团修饰可能会明显改变其对生物体的胁迫效应,导致难以预测的生态风险.  相似文献

This study was designed to investigate the in vivo effects of benzo[a]pyrene (BaP) on hepatic ethoxyresorufin-O-deethylase (EROD) activity and its correlation with cytochrome P450 1A (CYP1A) protein levels in Sebastiscus marmoratus, which were exposed through a water column to BaP (10, 100, 1000 ng/L, respectively) or were treated with intraperitoneal injections of BaP (0.5, 1, 5, 10 mg/kg, respectively) every 7 d. The results showed that after 25 d of waterborne exposure to 1000 ng/L BaP, fish hepatic CYP1A levels and EROD activity were significantly induced. In contrast, EROD activity was not altered 7 d after second ip injections, whereas, CYP1A protein levels were increased. Dose-dependent increase of biliary BaP metabolites demonstrated that the catalytic activity of CYP1A was induced by treatment with BaP. The lowest observable effect concentration with regard to biliary BaP metabolites (100 ng/L) was much lower than that with reference to EROD activity (1000 ng/L). The results suggest that biliary polycyclic aromatic hydrocarbon (PAH) metabolites were shown to better reflect the contamination gradients of PAHs than EROD activity. It appeared to be necessary to measure CYP1A protein levels to complement the EROD activity in relevant toxicological assessments.  相似文献
