在水稻幼苗25叶期,分别用浓度为C10.01%、C20.03%和C30.04%(W/V)的Ni(NO3)2溶液(含0.05%Tween20)喷雾处理,以05%(W/V)Tween20作对照.在4 h、24 h、48 h、72 h、96 h分别测定叶片中的过氧化氢酶(CAT)活性.结果表明,Ni(NO3)2对过氧化氢酶活性有抑制作用且随其浓度的升高,抑制作用越明显,在72 h抑制作用最强,过氧化氢酶活性比对照分别下降了21.7%、28.3%、55.8%.  相似文献

Effects of the simulated acid rain(AR) and ultraviolet-B(UV-B, 280-320 nm) radiation with a single or two ways simultaneously (AR + UV-B) on the antioxidant enzyme and photosynthesis of the rape seedlings were investigated by the hydroponic culture. The results of static experiment indicated that the tolerance of rape seedling to single stress(AR or UV-B) is stronger than that to dual stresses(AR + UV-B). Furthermore, the dual stresses had additive effect on catalase activity, and a synergistic effect on MDA content, net photosynthesis rate, water use efficiency as well as intercellular CO2 concentration. Meanwhile, it has an independent effect on chlorophyll content, stomatal conductance, and transpiration rate as well as membrane permeability. During 64 h restoration course, the dynamic change in the curves of physiological and biochemical indices were not identical, and none of them show a simple linear variation. According to the static and dynamic experiments, it was found that a responsive sequence of catalase activity, membrane permeability, MDA content and photosynthetic characteristics to the above-mentioned stresses was as follows: AR + UV-B 〉 UV-B 〉 AR.  相似文献

小鼠暴露于不同浓度乙酸铅1周后,对肝脏还原性谷胱甘肽(GSH)、谷胱甘肽还原酶(GR)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、活性氧物质(ROS)、总氧自由基清除能力(TOSC)进行测定.结果显示:小鼠铅暴露后,肝脏GSH水平和TOSC值略有下降;GR、CAT、SOD活性被抑制,在高剂量铅暴露组尤为明显;ROS升高1 5倍.这些结果表明,铅能对小鼠肝脏的抗氧化系统造成损伤.  相似文献

近年来,海洋石油开采与运输泄漏、石油及产品离岸排放等事故逐年增多,对近海海域生态环境产生了巨大破坏。为探究重质原油和轻质原油对紫贻贝(Mytilus edulis)毒理效应,测定了不同浓度重质原油和轻质原油暴露下紫贻贝鳃和外套膜中过氧化氢酶(CAT)和超氧化物歧化酶(SOD)的活性及体内石油烃的含量变化。结果显示,石油污染暴露下,紫贻贝腮和外套膜中CAT和SOD活性变化明显,与暴露浓度和暴露时间有关。CAT活性在轻质原油组随着浓度的增大,呈现下降趋势,且随着暴露时间的延长呈现先下降后上升趋势;在重质原油暴露组,随着浓度的增大,呈现先上升后下降趋势,且随着暴露时间的延长呈现先下降后上升趋势。SOD活性在轻质原油与重质原油暴露组,随着浓度的增大,呈现下降趋势,且随着暴露时间的延长呈现上升趋势并存在一定的剂量-效应关系。重质原油和轻质原油暴露168 h后紫贻贝体内总石油烃含量呈线性递增,生物富集系数(BCF)随着暴露浓度的增加不断减小并最终趋于平稳。结果表明,以紫贻贝腮和外套膜中SOD和CAT活性作为石油烃污染的生物标志物具有一定应用前景,紫贻贝对原油溶液中石油烃的生物富集作用可用来判断污染原油的来源和性质。  相似文献

The present investigation determined the effects of epibrassinolide (EBL) on the levels of indole-3-acetic acid (IAA), abscisic acid (ABA), and polyamine (PA) and antioxidant potential of 7-d old Raphanus sativus L. cv. ‘Pusa chetki’ seedlings grown under Cr (VI) metal stress. Reduced titers of free (0.767 μg g−1 FW) and bound (0.545 μg g−1 FW) IAA in Cr (VI) stressed seedlings were observed over untreated control. Supplementations of EBL to Cr (VI) stressed seedlings were able to enhance both free (2.14-5.68 μg g−1 FW) and bound IAA (2.45-7.78 μg g−1 FW) concentrations in comparison to Cr (VI) metal treatment alone. Significant rise in free (13.49 μg g−1 FW) and bound (12.17 μg g−1 FW) ABA contents were noticed for Cr (VI) stressed seedlings when compared to untreated control. No significant increase in ABA contents were recorded for Cr (VI) stressed seedlings upon supplementation with EBL over Cr (VI) treatment alone. A significant increase in Put (18.40 μg g−1 FW) and Cad (9.08 μg g−1 FW) contents were found for 10−9 M EBL plus Cr (VI) metal treatments when compared to Cr (VI) treatment alone. Spermidine (Spd) contents were found to decline significantly for EBL treatment alone or when supplemented with Cr (VI) treatments over untreated controls and Cr (VI) treatment alone. Antioxidant levels were found to enhance, with glutathione (57.98 mg g−1 FW), proline (4.97 mg g−1 FW), glycinebetaine (39.01 μmol mL−1), ascorbic acid (3.17 mg g−1 FW) and phytochelatins (65.69 μmol g−1 FW) contents noted for EBL supplemented to Cr (VI) metal solution over Cr (VI) treatment alone. Reduced activities of guaiacol peroxidase (0.391 U mg−1 protein) and catalase (0.221 U mg−1 protein) and enhanced activities of glutathione reductase (7.14 U mg−1 protein), superoxide dismutase (15.20 U mg−1 protein) and ascorbate peroxidase (4.31 U mg−1 protein) were observed in seedlings treated with EBL plus Cr (VI) over Cr metal treatment alone. Reduced MDA (2.55 μmol g−1 FW) and H2O2 (33.24 μmol g−1 FW) contents were recorded for 10−9 M EBL supplemented to Cr (VI) stress over Cr (VI) treatment alone. Enhancement in free radical scavenging potential as indicated by higher values of 1,1-diphenylpicrylhydrazyl, deoxyribose and reducing power activity assays, and increased levels of phenols and soluble sugars also showed significant influence of EBL in alleviating Cr (VI) stress in radish seedlings.  相似文献

采用对苯二甲酸(terephthalic acid,TA)作为羟基自由基(hydroxyl radical,·OH)捕捉剂,利用荧光分光光度法测定了二氧化氯(CIO2)接触后颤蚓体内-OH浓度变化,结合颤蚓灭活率、过氧化氢酶(catalase,CAT)和丙二醛(malondi.alderhyde,MDA)等指标的测试结果,研究了CIO2对颤蚓成虫的灭活效果及灭活机理。结果表明,ClO2对颤蚓成虫灭活效果显著。当C1O2投加浓度超过0.4mg/L,可对颤蚓体内·OH产生明显的诱导效应,并且·OH诱导效应与颤蚓灭活率直接相关。接触条件下CAT变化情况表明,颤蚓受到了明显的氧化胁迫,且对颤蚓体内抗氧化系统有破坏作用,MAD变化结果则表明ClO2对颤蚓的脂质过氧化作用不明显,可以对颤蚓表皮层有效渗透。上述结果证实了CIO2对颤蚓成虫的灭活作用机制之一是诱导其体内生成·OH,产生氧化胁迫,而CIO2对颤蚓抗氧化系统的破坏作用和对颤蚓表皮层的有效渗透进一步增强灭活效果。  相似文献

Saxitoxin (STX) is a cyanotoxin, which can cause neurotoxic effects and induce ecological changes in aquatic environments, a potential risk to public and environmental health. Many studies of cytotoxicity on animal cells and algae have been performed, although few compare the toxic effects between the two models. In this sense, we investigated the oxidative stress induced by STX (0.4-3.0 nM) in two different cellular models: Neuro-2A (N2A) cells and Chlamydomonas reinhardtii alga by quantification of malondialdehyde (MDA) levels as indicative of lipid peroxidation (LPO). Also was evaluated the antioxidant defense of these cells systems after exposure to STX by the addition of antioxidants in N2A cells culture, and by the measure of antioxidants enzymes activity in C. reinhardtii cells. The MDA levels of N2A cells increased from 15% to 113% for 0.4 and 3.0 nM of STX, respectively, as compared to control. Superoxide-dismutase and catalase did not appear to protect the cell from STX effect while, in cells treated with vitamin E, the rates of MDA production decreased significantly, except for higher concentrations of STX. No MDA productions were observed in algal cells however some effects on antioxidant enzymes activity were observed when algae were exposed to 3.0 nM STX. Our results indicate that the concentrations of STX that may induce oxidative stress through LPO are different in animal and phytoplankton communities. A combination of algal and animal bioassays should be conducted for reliable assessment of oxidative stress induced by STX.  相似文献

以疣荔枝螺(Thais clavigera)为研究材料,取不同程度性畸变雌性个体,以生存于同一生境中的雄性个体为对照,检测了性畸变个体与正常雌性和雄性超氧化物歧化酶(SOD)、脂质过氧化物(LPO)及过氧化氢酶(CAT)活性的差异,探讨TBT对性畸变个体SOD、LPO及CAT活性的影响。结果表明,疣荔枝螺性畸变程度愈严重,以上生化指标的差异愈明显,并呈现一定的规律性。  相似文献

文章对经过矿物微尘处理后大肠杆菌的总蛋白质浓度、SOD(超氧化物歧化酶)同工酶活性和CAT(过氧化氢酶)同工酶活性进行了测定和比较分析。结果表明:矿物微尘对大肠杆菌体内的蛋白质合成有很强的抑制作用,总蛋白质浓度在加入了微尘后表现出明显的下降。但SOD和CAT酶活性随着微尘浓度的增加而提高,表明大肠杆菌对微尘的作用作出了抗逆性反应。  相似文献

Polygodial is a drimane sesquiterpene dialdehyde derived from certain terrestrial plant species that potently inhibits ascidian metamorphosis, and thus has potential for controlling fouling ascidians in bivalve aquaculture. The current study examined the effects of polygodial on a range of biochemical biomarkers of oxidative stress and detoxification effort in the gills of adult Perna canaliculus Gmelin. Despite high statistical power and the success of positive controls, the antioxidant enzymes glutathione reductase (GR), glutathione peroxidase (GPOX), catalase (CAT), and superoxide dismutase (SOD); thiol status, as measured by total glutathione (GSH-t), glutathione disulphide (GSSG), and GSH-t/GSSG ratio; end products of oxidative damage, lipid hydroperoxides (LHPO) and protein carbonyls; and detoxification pathways, represented by GSH-t and glutathione S-transferase (GST), were unaffected in the gills of adult P. canaliculus exposed to polygodial at 0.1 or 1 × the 99% effective dose in fouling ascidians (IC99). Similarly, GR levels, thiol status, and detoxification activities were unaffected in mussels exposed to polygodial at 10 × the IC99, although GPOX, CAT, and SOD activities increased. However, the increases were small relative to positive controls, no corresponding oxidative damage was detected, and this concentration greatly exceeds effective doses required to inhibit fouling ascidians in aquaculture. These findings compliment a previous study that established the insensitivity to polygodial of P. canaliculus growth, condition, and mitochondrial functioning, providing additional support for the suitability of polygodial for use as an antifouling agent in bivalve aquaculture.  相似文献
