The microbial strain Azotobacter vinelandii UWD was grown under conditions of simulated microgravity in the National Aeronautics and Space Administration (NASA) Bioreactor. Bacterial growth in simulated microgravity differed significantly from that observed in conventional shake flask experiments: Cells tended to grow in a cluster-like pattern and polymer production started immediately after exposing them to conditions of simulated microgravity, and no lag time was observed. It was imperative to differentiate between the effects derived from microgravity and those imposed by the altered oxygen supply in the bioreactor. Aeration conditions were studied in both reactor types and a gas supply profile was developed for the bioreactor. This supply profile allowed for similar amounts of dissolved oxygen in the bioreactor and the shake flask in the initial stage of the fermentation and, therefore, for an evaluation of the effects of microgravity on biopolyester-producing bacteria. Since the optical density that is conventionally used as a measure for the cell growth could not be used due to the cluster-like growth pattern of the cells, it was determined that bacterial growth behavior in the bioreactor can be monitored through glucose or oxygen consumption.  相似文献

研究了氧化亚铁硫杆菌LX5(Thiobacillus ferrooxidans LX5)、氧化硫硫杆菌TS6(Thiobacillus thiooxidans TS6)和耐酸性酵母菌R30(Rhodotorula sp.R30)对重金属铬(Cr3+)的耐受性.结果表明,700mg/L的Cr3+对硫杆菌LX5、TS6的生长和氧化活性影响不大,但Cr3+浓度大于500mg/L时明显抑制酵母菌R30的生长.酵母菌R30与硫杆菌LX5和TS6复合能明显加速污泥淋滤的进程,最佳复合比为酵母菌R30接种量2.0%,硫杆菌LX5和TS6接种量10%.分别用含酵母菌R30数量为104个/mL和102个/mL的酸化污泥作接种物进行污泥淋滤,发现淋滤过程中pH值下降的速度没有明显差异,而在淋滤起始时添加2.0%的酵母菌R30,则淋滤反应提前36h结束.由此可见,在其它条件相同的污泥淋滤中,污泥中所含耐酸性酵母菌的数量是加快淋滤的关键.  相似文献

5-Hydroxymethylfurfural is found in several food commodities, including honey. Its concentration is used as one of honey's quality indicators since elevated 5-hydroxymethylfurfural levels indicate overheating and aging of the product. In this context, the European Commission and Codex Alimentarius established the maximum tolerable level for 5-hydroxymethylfurfural, for general type honeys intended for human consumption, at 40 mg kg?1. In this study, 5-hydroxymethylfurfural, 2-furfural, and the furfuralic derivative 5-methyl-2-furfural were investigated in Greek honey samples, through a high performance liquid chromatographic photo diode array mass spectrometric method developed and validated. Sample preparation was based on a modification of the ‘Quick, Easy, Cheap, Effective, Rugged, and Safe’ procedure. The method was sensitive, exhibiting limit of detection for 5-hydroxymethylfurfural at 0.1 mg kg?1, far below the permissible level of 40 mg kg?1. Levels of 5-hydroxymethylfurfural in 28 honey samples varied from 0.5 to 23.5 mg kg?1, thus, not worrying for consumers. Traces of 2-furfural were detected in limited samples.  相似文献

以Bi(NO33·5H2O、KI和g-C3N4为前驱体,采用常温沉淀法制备Bi5O7I/g-C3N4Z型异质结复合光催化剂,表征其光吸收性能、微观形貌、光生电子-空穴的分离效率等特性,研究新型光催化剂对RhB的可见光催化降解性能,探讨其可见光催化过程活性基团种类以及作用机理.结果表明:利用沉淀法合成Bi5O7I/g-C3N4的条件为:Bi(NO33·5H2O、KI和g-C3N4的投加量分别为4.85g、1.66g和1.61g,乙二醇的用量为50mL,反应液的pH值为12,反应搅拌速度为200r/min,反应温度为25℃.Bi5O7I/g-C3N4异质结无杂相生成且纯度高,异质结复合发生在g-C3N4的(002)晶面和Bi5O7I的(203)晶面,但g-C3N4和Bi5O7I的化学结构未受影响.Bi5O7I/g-C3N4呈三维纳米花瓣形貌结构,为光生电子-空穴的迁移提供了大量的接触位点.Bi5O7I的g-C3N4掺杂改性使其光催化活性显著增强,其光吸收边缘由425nm红移至462nm,Bi5O7I/g-C3N4的能带排列结构与Z型异质结匹配,促进了光生电子-空穴的分离.其光电流密度(11.5mA/cm)约为g-C3N4和Bi5O7I对应值的2.66倍和1.47倍.Bi5O7I/g-C3N4对罗丹明B的可见光催化降解率为93.9%,显著高于g-C3N4(58%)和Bi5O7I(49.7%)的降解效果,其光催化氧化活性主要来自羟基基团、超氧基团和光生空穴等中间态自由基.  相似文献

采用液质联用(HPLC-MS)的方法检测菌株Cupriavidus sp.DT-1降解2-羟基吡啶(2-HP)的代谢产物.并用三亲结合、荧光定量PCR (q-PCR)方法评价降解菌对3,5,6-三氯-2-吡啶酚(TCP)污染土壤的修复效果.结果表明,菌株可以进一步降解2-HP,依次生成尼古丁蓝、马来酰胺酸和反丁烯二酸,直至转化成菌株DT-1生长的碳源.接种菌株DT-1对污染土壤中TCP的降解起到较大的促进作用,2组试验土壤中TCP (50mg/kg)降解率分别为94.4%和86.7%,未接种菌株的土壤中TCP降解率仅为20.4%和28.4%.带有绿色荧光蛋白基因gfp标记的菌株DT-1-gfp可在土壤中存活35d以上,并对TCP污染土壤的细菌群落丰度有显著的恢复作用.  相似文献

Micro-mesoporous ZSM-5 zeolites were obtained by the post-treatment of tetrahydroxy ammonium hydroxide (TPAOH) solution with different concentration. The hierarchical pore structure formed during the desilication process facilitates the dispersion of Pt nanoparticles and Pt/ZSM-5 catalysts exhibit rather high catalytic activity for the deep oxidation of various VOCs at low temperature. The catalyst treated with TPAOH of 0.1 mol/L (Pt/ZSM-5(0.1)) shows the lowest degradation temperature (T90%) of 128 and 142°C, respectively for benzene and n-hexane. Compared with the untreated Pt/ZSM-5 catalyst, the abundant mesopores, small Pt particle size and finely dispersed Pt contribute to the superior catalytic activity and stability of the Pt/ZSM-5 catalysts for VOCs removal. More importantly, the existence of H2O in the feed gases hardly affected the activity of Pt/ZSM-5(0.1) catalyst at the low reaction temperature of 128°C, which is very important for VOCs low-temperature removal in the future practical applications.  相似文献

Volatile fatty acids (VFAs), which are largely generated during the anaerobic acidification process, are considered to be reliable indicators of the stable process operation. However, the common methods for monitoring VFAs are offline, and they are typically manual requiring time-consuming, costly and complex instruments. This study aims to develop a novel online analyzer for automatic measuring VFAs, which was based on the 5-pH point titration, embedded with a proportional-integral-derivative (PID) feedback control system. The results show that it can achieve accurate and rapid monitoring of VFAs ranging between 0-400 mg/L (<9 min/sample) but simultaneously faces the problems of overtitration and interference of complex characteristics of wastewater. In order to improve its accuracy and stability, the effects of three general coefficients (KI, KP, and KD) of PID on the titration were investigated, and the optimal values of KI, KP, and KD were found to be 1.5, 1.0, and -1.0~0.5, respectively. Besides, the initial titration speed was set at 0.06 mL/min, equal to the minimum speed of the peristaltic pump, and the dichotomy approach was integrated into the PID feedback controller. Owing to the above improvements, the relative mean deviation and standard deviation of measuring VFAs in both synthetic and real wastewaters were mostly lower than 5.0% and 5.0 mg/L, proving the online analyzer is rapid, accurate and reliable.  相似文献

采用溶胶-凝胶法(sol-gel method)制备铁钴双金属氧化物(Fe-CoO_x)催化剂,研究其协同超声(超声)催化过硫酸氢钾(PMS,KHSO5)降解酸性橙7(AO7)的效果及机理。结果显示:Fe-CoO_x/US/PMS体系的催化效果相较同条件下Fe Ox/US/PMS、CoO_x/US/PMS体系更好;随着初始PMS浓度的增加,AO7的降解率先增加后降低;增加US功率后AO7的降解率先增加后降低;Fe-CoO_x初始投加量从0.035 g/L上升至0.05 g/L后,30 min时AO7的降解率由30.58%上升至77.50%,继续增加投量,提升效果不明显;AO7的降解率与初始AO7呈负相关;投加叔丁醇(TBA)、甲醇(MA),50 min时AO7的降解率分别为60.64%、26.58%,表明体系中自由基主要为·OH和SO-4·。XRD和XPS结果显示:Fe-CoO_x催化剂粒径为8.8~10.7 nm,催化剂中的Fe、Co主要以Fe2+、Fe3+、Co2+、Co3+存在,并且表面羟基氧的含量为24.07%。研究显示:Fe-CoO_x催化剂具有较好的催化性能,能够有效地协同超声催化PMS生成自由基,对水中污染物的降解具有良好的效果。  相似文献

为了更好解读IEC 60068-2-5:2010中的辐照度和温度设定,及如何执行该标准,结合标准中对辐照度和温度设定的要求,提供具体的解决方案。建议在执行标准时,应使用日光过滤片来过滤氙灯光源,实现该标准对光源的要求。同时建议使用340 nm点控制,辐照度设定为0.60 W/m2@340 nm。对于温度的设定,建议按照标准中图2的温度上升、下降与时间的关系,对其进行设定。另外,标准中程序A的循环接近最严酷的自然条件,而程序B和程序C的循环比程序A的严酷很多。因此,一般情况下推荐程序A,对于耐候性好的材料,需要加速测试的,推荐程序B。  相似文献

为研究温州降雨径流污染情况,监测了温州2个采样区4场较为典型的降雨过程,测定了不同下垫面径流中BOD5和COD的浓度,并且计算出污染物的EMC值、M(V)曲线和BOD5/COD值等.结果表明,温州城市降雨径流中BOD5和COD的含量分别介于ND~69.21 mg.L-1和ND~636 mg.L-1之间,不同城市下垫面径流中BOD5和COD的含量均随着降雨历时的延长而呈下降趋势,处理初期径流水样对减少耗氧性有机污染具有重要意义.从COD和BOD5的EMC值来判断,部分城市下垫面降雨径流中污染物浓度超出地表五类水标准,甚至高于污水排放二级标准,此类降雨径流若直接入河将对下级受纳水体造成耗氧性有机污染的压力.根据M(V)累积曲线判断,COD初期冲刷效应在城市降雨径流中是较为普遍的,城市降雨径流中的BOD5表现出与COD类似的初始冲刷情况,该结果亦表明径流初期耗氧性有机污染严重.城市降雨径流中COD和BOD5的浓度受下垫面类型和降雨条件的影响,而BOD5/COD指标的变化特征则指出生物降解是削减有机污染负荷的有效途径之一,但不同下垫面径流有机污染的治理工作应采用不同的最佳管理方案(BMPs).  相似文献
