研究了蝮蛇清栓酶的有效成分精氨酸酯酶体外对中国仓鼠肺成纤维细胞 (CHL)增殖和染色体畸变的影响 .精氨酸酯酶的浓度以酶活单位U表示 .细胞增殖抑制试验和染色体畸变试验都用两种方法 :直接法和代谢活性法 .精氨酸酯酶设 3个浓度 :1.0× 10 -3 U /mL ,0 .5× 10 -3 U /mL和 0 .2 5× 10 -3U/mL .其中最高浓度比临床治疗浓度高约10 0倍 .细胞增殖抑制试验采用细胞贴壁培养 ,乙醇固定 ,结晶紫染色 ,单层细胞密度计测定细胞密度 .染色体畸变试验采用给药后 2 4h和 48h收集中期分裂细胞 ,低渗固定 ,制片 ,Giemsa染色 ,观察 10 0个中期分裂细胞的染色体结构异常及多倍体的出现率 .试验结果表明 ,精氨酸酯酶在上述浓度对CHL细胞的增殖没有抑制作用 ,对CHL细胞的染色体结构也没有影响 .表 2参 10 相似文献

Several theoretical, analytical, and institutional difficulties have impeded the development and application of the assessment of cumulative environmental impacts. Watershed development on coastal wetlands offers an ideal context for evaluating the land disturbance target approach to cumulative impact assessment. A model land use planning system involving a time series approach was developed for Elkhorn Slough in California. The approach included four major components: evaluation of erosion susceptibility, measurement of land disturbance, establishment of a land disturbance target, and a comparison of existing and target land disturbance values. Further research is needed to test the transferability of the approach in a wide range of coastal watersheds and to verify the applicability of the methods to other cumulative impact problems. 相似文献

采用蚕豆根尖微核技术研究了农药氰戊菊酯、三氟氯氰菊酯对蚕豆根尖细胞的遗传损伤.测定了不同浓度氰戊菊酯、三氟氯氰菊酯(5×10-10~5×10-2g·L-1)诱导下的蚕豆根尖细胞微核率、有丝分裂指数和染色体畸变率.结果表明,氰戊菊酯、三氟氯氰菊酯均能诱发较高的微核率,在一定浓度范围内,微核率随两种农药处理浓度的升高而增加,但随着处理浓度的进一步升高微核率反而呈下降趋势;两种农药均可使蚕豆根尖细胞有丝分裂指数增大;并能诱导蚕豆根尖细胞产生较高频率、多种类型的染色体畸变.氰戊菊酯和三氟氯氰菊酯对蚕豆根尖细胞具有明显的遗传毒性. 相似文献

The aim of this study was to determine the efficacy of combined maternal serum alpha-fetoprotein (MSAFP) and maternal serum human chorionic gonadotropin (MShCG) screening in detecting chromosome defects in the first trimester of pregnancy. Sera of 492 women (previously assayed for MSAFP) were analysed for MShCG under code without knowledge of cytogenetic results. Overall, 48 of 492 patients (9·8 per cent) had either an MSAFP multiple of the median ⩽0·5 or an MShCG β/a z ratio multiple of the median ⩽ 0·25, eight of whom had a fetus with a serious chromosome defect. A third of fetuses with Down' s syndrome and 83 per cent with trisomy 18 were detected at a potential‘cost’ of providing chorionic villus sampling or amniocentesis in 8·6 percent of women screened. 相似文献

Multipronuclear human eggs are frequent after in vitro fertilization. Their chromosome analysis can provide useful information. Before cleavage it can confirm the suspected poly-ploidy. Among the cleaved multipronuclear eggs it provides an estimation of the incidence of the possible return to diploidy. Ninety-four multipronuclear eggs were fixed at the first, second, or third cleavage according to the air-drying method of Tarkowski with or without colchicine exposure: 60 were successfully analysed. Twelve were stopped before cleavage (six without colchicine treatment and six with colchicine treatment). They were polyploid, confirming the cytological observation. Forty-eight eggs cleaved and were stopped by colchicine treatment and karyotyped. Seventeen eggs (35 per cent) had produced diploid embryos. Mosaicism was frequent (15 cases, 31 per cent). Triploidy was not frequent (8 eggs, 17 per cent). Haploidy constituted the remaining cases (8 eggs, 17 per cent). Our data indicate that the initial count of pronuclei is a reliable test. Multipronuclear one-cell oocytes were confirmed to be polyploid. Furthermore, the developmental capacity of the multipronuclear oocytes is variable. Most of them cleaved. 采用滤膜过滤微波干燥技术,建立了水中悬浮物的快速测定方法。以0.45μm硝酸乙酸纤维素(CN-CA)滤膜为过滤介质,采用变功率连续加热模式,空白湿滤膜在10 min内恒重(900 W/5 min+500 W/5 min),附有悬浮物的湿滤膜在13 min内恒重(900 W/8 min+500 W/5 min)。选择不同质量浓度的高岭土标准样品和不同类别的实际样品进行方法性能测试,结果表明,该方法检出限为5 mg/L,相对标准偏差(RSD)为2.7%~7.1%,相对误差为2.0%~5.0%,采用该方法与采用标准方法《水质悬浮物的测定重量法》(GB 11901—89)测定实际样品的结果吻合。该方法显著缩短了水中悬浮物测定的分析时间,精密度和准确度能够满足分析要求,可用于地表水及一般工业排放废水中悬浮物质量浓度的快速测定。 相似文献

From the public health point of view, several formal attempts have been made to measure the impact of prenatal diagnosis (PND) on the incidence of Down's Syndrome (DS), but the results have varied widely. The impact of PND (reduction in the birth rate of chromosomally abnormal neonates) is related to utilization rates but quantitative estimates of this have not been established. In a three-year (1981–1983) total population study from Queensland, Australia, we present results to measure the impact of a voluntary PND programme on the birth incidence of DS, and also other chromosomally abnormal births. Utilization rates for the PND service were 15·5 per cent in that population of mothers 35 years and over. Numbers and rates of all cases of chromosomal abnormalities are presented, subclassified by type of diagnosis–-either by PND or by clinical diagnosis after birth. For the total population, 7·3 per cent of cases of DS were detected prenatally, and 15·4 per cent of all chromosome abnormalities. (A method for measuring the impact of PND is described.) Using this in conjunction with our demographic data, we estimate that with a 15 per cent utilization rate of PND by older mothers, 14 per cent of DS births can be prevented in this age group, or a 5 per cent overall reduction can be achieved if mothers of all ages are considered. One index–-the ratio of the percentage of DS births which are preventable compared with the population utilization rates of PND–-has potential for widespread use. Queensland data for this ratio is 0·34, a figure consistent with that from other studies. Thus a 3·5 per cent drop in the overall DS birth rate may be expected for each 10 per cent increase in the utilization rates of PND for mothers of 35 years and over.
