研究了不同浓度异噻唑啉酮类化合物BIT(1,2-苯并异噻唑啉-3-酮)及其类似物X(N-丙酰基-1,2-苯并异噻唑啉酮-3-酮)对三角褐指藻生长的抑制效应.根据藻细胞生长、比生长速率、藻细胞密度比、藻细胞内色素含量变化结果表明,在实验所设定的浓度(0~3mg/L)范围内,BIT和X在高浓度下对三角褐指藻生长均具有一定的抑制效应,增加细胞生长的延滞期,但在低浓度下不明显,BIT的抑制效应优于X,但BIT和X的这种抑制作用均会随着处理时间的延长而减弱,藻细胞逐渐恢复快速增殖.根据Logistic曲线方程拟合获得的BIT和X对藻的96h半效应质量浓度(EC50)分别为1.95mg/L(R2=0.988, P=0.0013)和3.26mg/L(R2=0.908, P=0.0279),EC50值的大小进一步说明BIT比X对三角褐指藻的抑制作用强. 相似文献

3种有机磷农药-久效磷、对硫磷和辛硫磷对三角褐指藻的生长都有抑制效应,它们的72h半抑制剂量(72h·EC50)分别为9.74mg/L、8.20mg/L和1.52mg/L。结果表明,3均能引起藻细胞活性氧含量的增加,从而导致生物膜系统的膜脂过氧化和脱酯化伤害。这种由膜脂过氧化和脱酯化作用造成的膜损伤在3种农药中显示出较大的差异性。 相似文献

重金属胁迫对三角褐指藻生长及叶绿素荧光特性的影响 总被引:4,自引:0,他引:4
利用调制式叶绿素荧光仪研究了不同浓度的重金属Cu2+、Zn2+、Cd2+胁迫不同时间(24、48、72和96 h)后,三角褐指藻叶绿素荧光特性的变化情况.测定的主要参数有:光系统II(PSII)的最大光能转化效率(Fv/Fm)、PSII的潜在活性(Fv/Fo)、PSII的实际光能转化效率(Yield=ΦPSII)、相对光合电子传递效率(rETR)、光化学淬灭(qP)和非光化学淬灭(NPQ).研究结果表明,重金属(Cu2+、Zn2+、Cd2+)胁迫下三角褐指藻的叶绿素荧光参数Fv/Fm、Fv/Fo、Yield、rETR和qP均明显降低,并且随着重金属离子浓度的增加,胁迫时间的延长,下降幅度逐步增大;NPQ先上升后下降或者逐渐上升;细胞密度和叶绿素含量也显著降低.对3种重金属离子的半抑制浓度(EC50)的计算结果表明,3种重金属毒性大小顺序为Cd2+> Cu2+> Zn2+. 相似文献

This study aimed to investigate a suitable method to stain the nuclei of Phaeodactylum tricornutum with a flow cytometer equipped with a 488-nm laser to detect the DNA. Exponentially growing cells were fixed using a 70% ethanol, a 1% glutaraldehyde and a 1% formaldehyde solution, respectively. The samples were treated with RNase A and were then stained with propidium iodide. Samples not treated with RNase A were stained with SYBR-green I. All these samples were observed under a fluorescence microscope and the content of the nuclear DNA was quantitated by flow cytometry. Specific fluoresce spots indicate that fluorescence was detected only in the nuclei of cells stained with SYBR-green I and non-RNase A treated, using fluorescence microscopy. The result of flow cytometry showed that the DNA histogram of cells stained with SYBR-green I had distinct G1 and G2/M peaks, with a fit degree (RCS) of about 3, and a coefficient of variation (CV) of around 8.6%. However, the DNA histograms of cells stained with propidium iodide had a higher CV value, a poor fit degree, and no obvious G1 and G2/M peaks. The results indicate that SYBR Green I is an adequate detection method for nuclear DNA quantitation in cells of P. tricornutum using a flow cytometer equipped with a 488-nm laser, and without RNase A treatment. © 2018 Science Press.
