采用滇池水华蓝藻中提取提纯的微囊藻毒素(microcystins,MCs)作为微生物生长的碳源和氮源,从长期暴露于蓝藻水华的滇池底泥中,通过从含低浓度到高浓度MCs的逐步培养驯化,获得了高效降解MCs的微生物混合菌群,在初始MC-RR和LR浓度大约分别为50 mg/L和30 mg/L下,3 d内可将MCs全部降解.进一步活性研究显示,不同含碳和含氮化合物虽然能够促进混合微生物菌群的生长,但对降解MCs却无明显的促进作用,说明MCs既可以作为微生物生长的碳源,又可以作为微生物生长的氮源,在富含有机物的天然水体中并不一定能够促进微生物对MCs的生物降解.  相似文献

为实现酱香型酒糟资源化综合利用和黄壤氮素有效性提升,采取田间培养试验,通过设置5个生物炭施用量0%(MB0)、0.5%(MB0.5)、1.0%(MB1.0)、2.0%(MB2.0)和4.0%(MB4.0),研究酒糟生物炭短期施用对贵州黄壤氮素有效性及细菌群落结构多样性的影响.结果表明,施用酒糟生物炭使土壤全氮(TN)和硝态氮(NN)含量分别提高35.79%~365.26%和122.96%~171.80%,微生物量氮(MBN)含量降低34.10%~59.95%,且随着生物炭施用量的增加,铵态氮/全氮(AN/TN)、硝态氮/全氮(NN/TN)和微生物量氮/全氮(MBN/TN)呈现出降低趋势.施用酒糟生物炭显著降低了土壤细菌OTU数量、群落丰富度和多样性,且随酒糟生物炭施用量的增加,该影响程度随之增加.与未施用酒糟生物炭MB0处理相比,酒糟生物炭的施用显著改变土壤细菌群落结构,随着生物炭施用量的增加,拟杆菌门(Bacteroidetes)相对丰度提高了1.76~2.11倍,而酸杆菌门(Acidobacteria)、绿弯菌门(Chloroflexi)、芽单胞菌门(Gemmatimonadetes)、浮霉菌门(Planctomycetes)、装甲菌门(Armatimonadetes)、奇古菌门(Thaumarchaeota)和硝化螺旋菌门(Nitrospirae)相对丰度均出现不同程度的降低,均以MB4.0处理降幅最为显著.同时,酒糟生物炭的施用还增加了一些土壤功能细菌的相对丰度,如链霉菌属(Streptomyces)、极小单胞菌属(Pusillimonas)等,降低了溶杆菌属(Lysobacter)和芽孢杆菌属(Gemmatimonas)等优势菌属的相对丰度.此外,冗余分析(RDA)结果显示,MBN/TN、NN和MBN是引起土壤细菌群落结构变化的主要氮素环境因子,而且MBN/TN、MBN与奇古菌门(Thaumarchaeota)和硝化螺旋菌属(Nitrospira)均呈显著正相关性,说明短期内酒糟生物炭的施用可以显著降低氨氧化古菌和硝化细菌的丰度,抑制土壤氨氧化作用和硝化速率,提高土壤氮素有效性.综上所述,短期内施用酒糟生物炭可以提高黄壤氮素养分,改变土壤细菌群落结构和多样性,并可以通过抑制土壤氨氧化作用和硝化作用有效阻控土壤氮素淋溶发生的风险,提高土壤氮素有效性.  相似文献

长三角典型城郊土壤抗生素空间分布的影响因素研究   总被引:4,自引:0,他引:4  
阐明土壤抗生素的空间分布特征对于认识抗生素的空间扩散和迁移过程、控制土壤抗生素污染、维护土壤安全具有重要意义.本研究通过空间采样,系统分析了长三角地区典型城郊流域土壤抗生素的空间分布特征,并探讨了人类活动、土壤性质和微生物群落对土壤抗生素分布的影响.结果表明,农田土壤中抗生素含量显著高于园地和林地,其中,检出浓度顺序为:四环素类(TCs)喹诺酮类(QNs)大环内酯类(MLs)磺胺类(SAs),平均含量分别为41.43、11.38、0.15、0.09μg·kg~(-1),农田土壤抗生素含量与有机肥施用密切相关.Spearman相关分析结果表明,TCs、QNs、SAs及抗生素总含量与距城镇距离、距道路距离、海拔及C/N具有显著的负相关性(p0.05),表明这3类抗生素的空间分布受人类活动的强烈影响,而MLs含量与人类活动的关系则较弱.土壤养分、颗粒组成、pH、容重等对抗生素的吸附、迁移、降解等环境行为具有强烈的影响,其空间异质性影响了土壤抗生素空间分布的差异性.土壤微生物群落与土壤抗生素之间具有较为复杂的联系,研究发现,部分变形菌门、酸杆菌门、拟杆菌门、放线菌门及浮霉菌门细菌与土壤抗生素含量呈现负相关.方差分解分析揭示出土壤性质、人类活动及其相互作用是土壤抗生素空间分布的主要影响因素,对土壤抗生素分布空间变化的总解释度接近于58%,达到了较高水平.  相似文献

为探究澜沧江流域的浮游细菌群落结构特征及驱动因子,应用16S rRNA高通量测序技术,分析了2017年2月澜沧江流域浮游细菌群落结构特征,并采用Pearson相关性分析(Pearson correlation analysis)和冗余分析(RDA)识别了澜沧江自然河道段和水库段浮游细菌群落结构变化的关键环境因子.结果表明,自然河道段ACE指数和Shannon指数均高于水库段,造成自然河道段和水库段浮游细菌多样性变化的主要环境因子为水温(WT)、溶解氧(DO)、浊度(Tur)、高锰酸盐指数(permanganate index)、p H和总氮(TN).对16S rRNA V3和V4测序,得到用于物种分类的OTU数共26772,涵盖了浮游细菌群落共45门,965属.菌群分类发现,变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)和放线菌门(Actinobacteria)为优势门,其中变形菌门(Proteobacteria)含量相对丰富,占细菌群落的36%~94%.澜沧江流域变形菌门(Proteobacteria)主要包括α-变形菌纲(α-Proteobacteria)、β-变形菌纲(β-Proteobacteria)和γ-变形菌纲(γ-Proteobacteria),分别占变形菌门(Proteobacteria)的比例为0.39%~21.56%、0.39%~55.80%和31.09%~99.18%.澜沧江水体浮游细菌群落空间差异明显,影响浮游细菌群落结构变化的环境因子主要为WT、高锰酸盐指数、Tur、DO和TN.自然河道段和水库段影响浮游细菌群落结构的环境因子不同,DO和Tur是影响自然河道段浮游细菌群落结构的关键环境因子,而水库段浮游细菌群落结构主要受WT、高锰酸盐指数和TN的影响.  相似文献

为了研究Pb污染对根际土壤微生物多样性的影响,揭示根际微生物对Pb胁迫的适应性机制,通过盆栽试验模拟轻度[w(Pb)为300 mg/kg]、中度[w(Pb)为600 mg/kg]和重度[w(Pb)为900 mg/kg]Pb污染土壤环境,利用高通量测序技术和生物信息学分析方法,测定了不同w(Pb)处理下三叶草根际土壤中细菌群落的丰度和组成.结果表明:①共检测到37个门,其中变形菌门为优势菌群,占比为50.7%~53.9%,其次为拟杆菌门、酸杆菌门和疣微菌门. ②共检测到623个属,其中鞘氨醇单胞菌属为优势菌属,占比为17.1%~19.4%. ③中度Pb污染土壤样本中细菌的多样性最高,重度Pb污染土壤样本中的细菌数量最少,并且其群落组成与其他处理差异最大.研究显示,重度Pb污染会显著抑制三叶草根际土壤样本中细菌的生长,降低土壤样本中细菌总量及其群落的多样性,但中度Pb污染会提高土壤样本中细菌群落的多样性. Pb污染会改变三叶草根际土壤样本中细菌群落组成和丰度,不同类型细菌对Pb污染土壤的适应性不同.根际土壤样本中Gp1菌群的丰度随着土壤w(Pb)的增加而增加,说明Gp1菌群可能是具有Pb污染抗性的优势菌群.   相似文献

Summary. The plant apparency hypothesis predicts that apparent plants invest in broadly effective defences such as tannins while unapparent plants invest in specific toxins such as alkaloids. The stress hypothesis states that plants invest in cheaper defences if they have evolved in habitats that impose abiotic limitations to plant fitness. We tested these hypotheses by determining the concentrations of alkaloids and tannins in a representative sample of the vascular plants of continental Chile (with exclusion of Pteridophyta, Cactaceae, and Poaceae) consisting of 396 species. In a subsample of 166 species which contained both alkaloids and tannins, we constructed the A/T index (A/T = [alkaloids]/ [tannins]). We discarded the presumed effect of phylogeny (as estimated by taxonomy) on the variation observed in the data because no correlation of A/T with taxonomic relationships among species either at family or genus levels was found in a nested ANOVA with genera nested in families. Concentration of alkaloids was negatively correlated with that of tannins. We compared the value of A/T among species differing in life form (herbs, shrubs or trees), herb longevity (annual or perennial), leaf-shedding manner of woody plants (deciduous or evergreen), latitudinal range, and level of water stress typical in their natural habitat. Unapparent plants (herbs, annual) exhibited higher mean A/T index than apparent plants (shrubs and trees, perennial). A/T did not correlate with latitudinal range. Mean A/T values decreased from deserts to deciduous forests. The comparisons were not always significant due to the inevitable unbalance of the data set which lowers the power of the statistical tests employed. The results suggest that chemical defences are indeed distributed in a non-random manner among plants, and that to a large extent the predictions derived from the apparency and stress hypotheses are sustained.  相似文献

Three methods (membrane filtration, multiple tube fermentation, and chromogenic substrate technology kits manufactured by IDEXX Laboratories, Inc.) are routinely used to measure indicator bacteria for beach water quality. To assess comparability of these methods, quantify within-laboratory variability for each method, and place that variability into context of variability among laboratories using the same method, 22 southern California laboratories participated in a series of intercalibration exercises. Each laboratory processed three to five replicates from thirteen samples, with total coliforms, fecal coliforms or enterococci measured depending on the sample. Results were generally comparable among methods, though membrane filtration appeared to underestimate the other two methods for fecal coliforms, possibly due to clumping. Variability was greatest for the multiple tube fermentation method. For all three methods, within laboratory variability was greater than among laboratories variability.  相似文献

Environmental pollution is one of the major problems being facedtoday due to the advent of extensive industrialization. Heavy metals are the most important contaminants in wastewater that are present in abundance and are toxic. Heavy metal contentsof sewage in the industrial estate of Aligarh (U.P.) have beendetermined by atomic absorption spectrophotometry. The analysisof samples collected from six different locations revealedsignificantly high levels of Fe, Zn, Cu, Cr and Ni. Certainbacteriological (Total bacterial count, Total coliform, Fecal coliform, and Fecal Streptococci) parameters of domestic and industrial sewage as well as soils were monitored from March1990 to January 1993. Total bacterial count, total coliform,fecal coliform and fecal streptococci were found to be lowestin all the samples of industrial wastewater compared to thosein domestic sewage and soil systems. The soil however, contained highest total culturable bacterial population. In view of the common practice of the application of sewage to agricultural land in the neighbouring area, the discharge ofindustrial wastewater without proper treatment into public sewers should be strictly prohibited.  相似文献

Cryptosporidium oocysts and Giardia cysts has been isolated from waters worldwide. In Italy, studies on these parasites in the environment are still limited due to absence of epidemiological evidence and difficulty of adequate methodologies of sampling and analysis. The new Drinking Water Directive 98/83/CE states that Cryptosporidium has to be determined in water intended for human consumption if Clostridium perfringens is detected. This investigation contributes to the knowledge of both Cryptosporidium and Giardia occurrences in Italian aquatic environments through a two-year monitoring program. In addition, indicator bacteria and Clostridium perfringens were monitored in sewage, surface waters, drinking water, and swimming pools and possible correlations were calculated among all the selected parameters. Cysts and oocysts were detected in sewage and surface water and Giardia numbers always prevailed over Cryptosporidium. The parasites were not detected in drinking water, while a positive sample was obtained from the analysis of swimming pools. Pearsons correlation coefficients evidenced a reciprocal correlation between both the protozoa and the Enterococci counts.  相似文献
