人工控制条件下 ,初步研究了长江华溪蟹 (Sinopotamonyangtsekiense)对Cu2 的毒性反应 ,并探讨其作为环境监测生物的可能性 .结果表明 ,随浓度的升高、浸药时间的延长 ,Cu2 对长江华溪蟹的毒性增大 .Cu2 的毒性与长江华溪蟹性别也有密切关系 ,雄性个体对Cu2 的忍耐度低于雌性个体 ;对达到性成熟的长江华溪蟹个体 ,Cu2 毒性与其大小无明显相关性 .温度影响Cu2 对长江华溪蟹的毒性 ,较高温度 (18℃ )下的毒性高于较低温度 (14℃ )下的毒性 .图 3表 1参 16  相似文献

The impacts of a functional and a demolished copper processing works on the aquatic and terrestrial environment in the vicinity of the works was investigated by determining the levels of selected trace metals in river water, river sediments, channel margin sediments and overbank soils. Samples were taken at five sites within an area of the Churnet Valley in Staffordshire, where the River Churnet flows through the two works. Analysis of river water samples by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) has shown that only copper is present above background levels considered to typify uncontaminated rivers. Analysis of river sediments, channel margin sediments and overbank soils by nitric-perchloric acid digestion and Inductively Coupled Plasma-Atomic Emission Spectroscopy (ICP-AES) analysis has indicated contamination by arsenic, cadmium and copper in the vicinity of both works. Arsenic and copper are deposited primarily within the aquatic environment, although some contamination of the terrestrial environment by copper is also observed. Cadmium is deposited primarily within the terrestrial environment. The deposition of arsenic and copper in river and channel margin sediments respectively is also related to current and historical contamination.  相似文献

选用101株分离自扁蓿豆、胡枝子、野豌豆、锦鸡儿属植物的根瘤菌菌株,进行了营养利用、抗生素抗性、耐逆性和酶活性测定等84个表型性状分析,发现分离自同种寄主植物的根瘤菌由于地理来源的不相同而存在着较大的多样性.通过数值分类,各已知种分别聚群,101株未知菌在84%的相似水平上分为4个各不同的群.根据1、2、3、4群菌的聚类情况,结合在不同水分梯度下采集的豆科牧草根瘤菌,结果发现除个别菌外,绝大部分菌株呈现随着水分梯度的不同,各菌以类群形式分布.证明土壤中水分含量对根瘤菌的分布有很大影响.  相似文献

Greenhouse experiments were conducted to determine the effects of sand burial on survival and growth of seedlings ofCirsium pitcheri. In 1992–1993, seedlings were buried to depths of 0, 25, 50, 75, and 100% of their height while in 1993–1994, the seedlings were buried to depths of 0, 4 cm (single burial), 4 cm (repeated burial of 1 cm every 8 days), 8 cm (single burial) and 8 cm (repeated burial of 2 cm every 8 days). Several physiological traits, net photosynthetic rate, chlorophyll a∶b ratio, leaf area, number and length of leaves, number of internodes, amount of tillering, and biomass, were measured. The results showed that all seedlings died in the complete (100%) burial, 20% died in the 75% burial and none died in the 0, 25 and 50% burial treatments. Burial of seedlings to a depth of 25% stimulated their growth but 75% burial significantly decreased the total dry weight. Repeated burial treatments exhibited significantly greater stimulation of growth than single burial. Surviving seedlings grew through the sand deposit by elongating the stem and leaf petioles, increasing the number of nodes and the length of internodes. This elongation occurred at the expense of development of the root system indicating that available energy was re-allocated to above-ground parts.  相似文献

Arsenic is present in airborne particulate material released by coal-fired power plants and non-ferrous metal smelters. We have assessed whether the physico-chemical properties of arsenic in such particles play a role in its lung retention and uptake by the body. Female hamsters were given a single intratracheal instillation of fly ash or copper smelter dust suspensions (at doses of 50 or 100 g As kg–1) or identical amounts of soluble tri- and pentavalent arsenic, in the presence or absence of an inert dust material (tungsten carbide). The concentration of the element was measured in a 24 hour urine sample collected on the 1st, 2nd and 6th day after treatment and arsenic remaining in lung tissue was determined at the end of the same time periods. Both lung retention and urinary As excretion indicate a prolonged contact of the lung tissue with particulate As in contrast to soluble As salts. In addition to the effect of solubility described here, more research is needed to determine the effect of particle size and lung loading on retention, as well as the potential differences in the lung inflammatory response using arsenic-rich particulates from various sources.  相似文献

牛粪发酵过程中抗生素耐药基因及相关菌群组成变化规律   总被引:2,自引:0,他引:2  
随着规模化奶牛养殖迅速发展,牛粪带来的环境污染问题日益严重。实验探究牛粪及其堆肥过程中养分变化、抗生素耐药基因消减情况以及细菌群落演替规律,探讨细菌菌群与养分、抗生素耐药基因之间的相关性。采集牛粪及其堆肥过程样品,测定样品中养分、检测抗生素耐药基因相对丰度,采用16S r RNA高通量测序技术研究牛粪堆肥过程细菌群落变化。结果表明,从新鲜牛粪(组FM)到发酵牛粪(组C),样品中有机质含量下降,全氮和全磷含量有不同程度增加,全钾(TK)显著升高(P<0.05)。堆肥发酵后,抗生素耐药基因erm B、tet M、bla CTX-M和aac(6')-Ib-cr的相对丰度呈现不同程度的下降,而基因sul1相对丰度增加。高通量测序结果表明,在门水平,各组均以Firmicutes、Proteobacteria和Bacteroidetes这3个菌门为主,在属水平,各组具有不同的优势物种,其中发现Bacteroides、Paeniclostridium和Pseudomonas等7个潜在病原菌属的相对丰度在堆肥后有效降低。通过相关性分析发现,潜在病原菌属与基因tet M和sul1呈显著正相关(P<0.05),此外,大部分优势菌属与养分TK有显著相关性(P<0.05)。研究结果表明,牛粪经堆肥发酵后,病原菌相对丰度有效降低,抗生素耐药基因相对丰度的下降与细菌菌群结构变化存在相关关系。  相似文献

A river embankment sediment beneath a road bridge in South Australia had been surveyed on a previous occasion to a depth of 10 cm and found to contain high levels of copper, lead and zinc deposited during previous lead and zinc-based primer paint stripping operations prior to repainting the bridge. Since the river embankment is geologically different from the surrounding non-embankment soils, and a sufficient distance from the bridge along the river embankment could not be traversed, the geological background Cu, Pb and Zn fraction was established initially using a solid phase sequential extraction (SPSE) procedure. Assessment of the degree of contamination and extent of vertical mobility was then established. Copper was a minor contaminant and present predominantly as the geological background species. Lead was partitioned evenly between the oxalate soluble fraction and residual fraction to a depth of 30 cm with a decrease in total Pb concentration from 497 to 141 mg Pb kg–1 while Zn was predominantly in the oxalate soluble fraction to a depth of 15cm with a decrease in concentration from 1013 to 150 mg Zn kg–1. The reduced rate of nitrification and the lower total concentration of NO3 -N together with the higher respiratory quotient, combined with the lower microbial quotient, are indicative of microbial stress due to heavy metal toxicity. In the case of Pb and Zn paint stripping residues, these changes in indicators of microbial health are possibly the result of the greater abundance and hence possible bioavailabilty of organo- and chloro-Pb compared to organo- and chloro-Zn complexes.  相似文献

通过基因全合成实现基因的分子改造和人工组建正成为一种实验室常规手段,因此,建立一种能够在相对低廉和短时间内准确和高效地设计和合成长片段基因的方法十分重要.本研究报道了一种重复性好、错误率低、低成本和简便的基因设计和全合成方法.此方法包含经DNA2.0软件的序列优化,Gene2Oligo软件的寡核苷酸设计,覆盖全长基因双链的寡核苷酸的组合和引物介导下的全基因的合成等步骤.运用此方法,对3个不同长度的基因(分别为653bp,1309bp和1498bp)成功地进行了密码子优化和一步全合成.其中的amyFF在大肠杆菌中表达量提高了12倍.图2参18  相似文献

Clones of Silene vulgaris ssp. maritima individuals from Dolfrwynog Bog, North Wales and from a coastal control site were propagated under standardised environmental conditions. After five months, cuttings were taken and tested for copper tolerance by means of root extension analysis in solutions of various different copper concentrations. Indices of tolerance suggested that individuals from the Dolfrwynog population were more resistant to elevated copper concentrations than were the control population individuals. Due to the initial five months under standardised conditions, it was assumed that the cloned plant material would have lost all plastic adaptive responses to its native environment. If this assumption was valid then the differences in copper tolerance observed between the two populations had a genetic basis and the Dolfrwynog Bog population is a copper tolerant ecotype.  相似文献
