旱柳(Salix matsudana Koidz)对Pb的耐性、富集、转运与胁迫响应研究 总被引:1,自引:0,他引:1
耐性机理的系统解析对于旱柳在土壤Pb污染控制上的应用具有重要意义.为了明确旱柳对Pb的耐性机理,通过盆栽试验研究了旱柳对Pb的耐性、富集、转运特征,采用STEM、EDAX、FTIR分析了旱柳对Pb的胁迫响应机制.结果表明:低含量(≤600 mg·kg-1)Pb对旱柳生长有促进作用,高含量(≥1000 mg·kg-1)Pb明显抑制旱柳的生长;旱柳对Pb具有很高的耐性,耐性指数TI(%)可达108.32,属Pb高耐性植物;旱柳对Pb的富集能力较强,富集系数均值可达7.5773,而对Pb的转运能力较弱,转运系数均小于1;低含量Pb胁迫不影响叶绿素的合成,也不会促生丙二醛(MDA),而高含量Pb胁迫明显抑制叶绿素的合成,促使MDA大量生成,导致严重的细胞膜脂过氧化和膜损伤;抗氧化酶(POD、CAT、SOD)及可溶性蛋白能够对低含量Pb胁迫做出积极有效的响应,但无法清除高含量Pb胁迫所产生的毒害;扫描透射电镜(STEM)分析表明,Pb胁迫使根系发黑、萎蔫,茎部破败、堵塞,叶片褶皱、卷曲,破坏气孔结构;傅里叶红外光谱(FTIR)分析表明,Pb胁迫使导致膜脂过氧化和膜损伤的自由羟基增多,而使对Pb具有络合、螯合作用的连生羟基、巯基、羰基数量减少;能谱(EDAX)分析表明,Pb胁迫抑制了Ca、K的吸收,同时干扰其他矿质元素的吸收利用. 相似文献

Many studies have evaluated effectiveness of corridors by measuring species presence in and movement through small structural corridors. However, few studies have assessed whether these response variables are adequate for assessing whether the conservation goals of the corridors have been achieved or considered the costs or lag times involved in measuring the response variables. We examined 4 response variables—presence of the focal species in the corridor, interpatch movement via the corridor, gene flow, and patch occupancy—with respect to 3 criteria—relevance to conservation goals, lag time (fewest generations at which a positive response to the corridor might be evident with a particular variable), and the cost of a study when applying a particular variable. The presence variable had the least relevance to conservation goals, no lag time advantage compared with interpatch movement, and only a moderate cost advantage over interpatch movement or gene flow. Movement of individual animals between patches was the most appropriate response variable for a corridor intended to provide seasonal migration, but it was not an appropriate response variable for corridor dwellers, and for passage species it was only moderately relevant to the goals of gene flow, demographic rescue, and recolonization. Response variables related to gene flow provided a good trade‐off among cost, relevance to conservation goals, and lag time. Nonetheless, the lag time of 10–20 generations means that evaluation of conservation corridors cannot occur until a few decades after a corridor has been established. Response variables related to occupancy were most relevant to conservation goals, but the lag time and costs to detect corridor effects on occupancy were much greater than the lag time and costs to detect corridor effects on gene flow. 臭椿(Ailanthus altissima)具有很强的抗逆性,能适应多种不良的环境,在中国广泛分布,是一个很有潜力的修复铜污染土壤的物种。通过种植实验检验臭椿修复铜污染土壤的能力。臭椿实生苗移植于4种不同铜的质量分数(0、29、57和86 mg·kg-1)、3种不同的施肥处理(对照、有机肥、无机肥)的土壤,经过5周种植,测量臭椿根、茎、叶的铜含量及各部分生物量。试验结果表明:1)土壤铜浓度的差异对臭椿生长没有显著影响;2)土壤施肥处理显著影响臭椿生长,施加无机肥生长最佳,对照次之,施有机肥最低;3)随土壤铜浓度增加,臭椿根、茎、叶的铜含量增加,然后趋缓,但似乎根、茎、叶的铜含量变化不同步;4)土壤肥力对臭椿茎、叶的铜含量没有显著影响,但对根的铜含量影响显著,施加无机肥的根中铜含量显著增加。臭椿经过短短5周处理,其体内铜的质量分数就到达120μg·g-1,且铜在各组织中的分布比为根:茎:叶=2?1?2。相比一些超富集铜的草本植物,臭椿的铜吸收能力尚有距离,但其高生长、高抗逆性、深根系的特点,加之其对其他众多重金属元素的吸收能力使得这个物种在生物修复污染土壤方面的作用不容小觑。本研究丰富了治理土壤重金属污染的物种库,为进一步寻找重金属“超富集植物”,更有效的治理土壤污染提供一定的参考价值。 相似文献

Environmental changes strongly impact the distribution of species and subsequently the composition of species assemblages. Although most community ecology studies represent temporal snap shots, long‐term observations are rather rare. However, only such time series allow the identification of species composition shifts over several decades or even centuries. We analyzed changes in the species composition of a southeastern German butterfly and burnet moth community over nearly 2 centuries (1840–2013). We classified all species observed over this period according to their ecological tolerance, thereby assessing their degree of habitat specialisation. This classification was based on traits of the butterfly and burnet moth species and on their larval host plants. We collected data on temperature and precipitation for our study area over the same period. The number of species declined substantially from 1840 (117 species) to 2013 (71 species). The proportion of habitat specialists decreased, and most of these are currently endangered. In contrast, the proportion of habitat generalists increased. Species with restricted dispersal behavior and species in need of areas poor in soil nutrients had severe losses. Furthermore, our data indicated a decrease in species composition similarity between different decades over time. These data on species composition changes and the general trends of modifications may reflect effects from climate change and atmospheric nitrogen loads, as indicated by the ecological characteristics of host plant species and local changes in habitat configuration with increasing fragmentation. Many populations of threatened mammals persist outside formally protected areas, and their survival depends on the willingness of communities to coexist with them. An understanding of the attitudes, and specifically the tolerance, of individuals and communities and the factors that determine these is therefore fundamental to designing strategies to alleviate human‐wildlife conflict. We conducted a meta‐analysis to identify factors that affected attitudes toward 4 groups of terrestrial mammals. Elephants (65%) elicited the most positive attitudes, followed by primates (55%), ungulates (53%), and carnivores (44%). Urban residents presented the most positive attitudes (80%), followed by commercial farmers (51%) and communal farmers (26%). A tolerance to damage index showed that human tolerance of ungulates and primates was proportional to the probability of experiencing damage while elephants elicited tolerance levels higher than anticipated and carnivores elicited tolerance levels lower than anticipated. Contrary to conventional wisdom, experiencing damage was not always the dominant factor determining attitudes. Communal farmers had a lower probability of being positive toward carnivores irrespective of probability of experiencing damage, while commercial farmers and urban residents were more likely to be positive toward carnivores irrespective of damage. Urban residents were more likely to be positive toward ungulates, elephants, and primates when probability of damage was low, but not when it was high. Commercial and communal farmers had a higher probability of being positive toward ungulates, primates, and elephants irrespective of probability of experiencing damage. Taxonomic bias may therefore be important. Identifying the distinct factors explaining these attitudes and the specific contexts in which they operate, inclusive of the species causing damage, will be essential for prioritizing conservation investments. Abstract: Widespread poaching prior to the 1989 ivory ban greatly altered the demographic structure of matrilineal African elephant (Loxodonta africana) family groups in many populations by decreasing the number of old, adult females. We assessed the long‐term impacts of poaching by investigating genetic, physiological, and reproductive correlates of a disturbed social structure resulting from heavy poaching of an African elephant population in Mikumi National Park, Tanzania, prior to 1989. We examined fecal glucocorticoid levels and reproductive output among 218 adult female elephants from 109 groups differing in size, age structure, and average genetic relatedness over 25 months from 2003 to 2005. The distribution in group size has changed little since 1989, but the number of families with tusked old matriarchs has increased by 14.2%. Females from groups that lacked an old matriarch, first‐order adult relatives, and strong social bonds had significantly higher fecal glucocorticoid values than those from groups with these features (all females R2= 0.31; females in multiadult groups R2= 0.46). Females that frequented isolated areas with historically high poaching risk had higher fecal glucocorticoid values than those in low poaching risk areas. Females with weak bonds and low group relatedness had significantly lower reproductive output (R2[U]=0.21). Females from disrupted groups, defined as having observed average group relatedness 1 SD below the expected mean for a simulated unpoached family, had significantly lower reproductive output than females from intact groups, despite many being in their reproductive prime. These results suggest that long‐term negative impacts from poaching of old, related matriarchs have persisted among adult female elephants 1.5 decades after the 1989 ivory ban was implemented. 相似文献

Phosphogypsum (PG), a solid by-product of phosphoric acid production, contains radionuclides and trace metals in concentrations which may pose a potential hazard to human health and the environment. to investigate the possibility of bioaccumulation of radium and six heavy metals over time when aquatic organisms experience both trophic and environmental exposure to PG, we designed a laboratory experiment representing three levels of an aquatic food chain. During the 135 day experiment, a meiobenthic copepod species (Amphiascoides atopus) was cultured in the presence of PG. the copepods were subsequently fed to grass shrimp (Palaemonetes vulgaris and P. pugio) which were in turn fed to gulf killifish (Fundulus grandis); both the grass shrimp and the killifish also experienced an environmental PG exposure. Other than elevated radium levels in the experimental grass shrimp, the experiment demonstrated little effect of environmental or trophic exposure to PG on microinvertebrates, macroinvertebrates, or fishes that could be attributed to PG. in all cases where increased concentrations were indicated within the experimental group, roughly equivalent increases in metal concentrations also occurred in the control group. 相似文献

The presence of Maillard reaction products (MRP) in foods and food components is due to the non-enzymatic reaction between protein and carbohydrate residues triggered by thermal steps during food processing. The objective of this study was to assess the effect of MRPs and increasing lysine concentrations on S. Typhimurium growth and the expression of cadA which may be an indirect determinant of Salmonella virulence response. Variations in lysine concentrations (from 0 to 0.5 mM) did not exert any effect either on the final optical density after 6-hour incubation or the growth rates of S. Typhimurium in media containing MRPs. In contrast to the reduced final absorbancy of the bacterial cultures grown with histidine and arginine MRPs supplementations (0.1%), growth rates, in general, remained unaltered by all MRPs at each lysine concentration when compared to the control (M9 pH 5.8, no MRPs added). The induction levels of cadA in media containing 0.1% MRPs were close to cadA induction in the reference media (M9, pH 5.8 and no MRPs) and did not exceed the corresponding values by more than approximately 30%. Although the observed negligible induction of cadA under these conditions complies with the concept of its potential “anti-virulence” function, additional studies involving various concentrations and more refined MRPs are needed. 相似文献
