近年来,荧光法成为微痕量硝基芳香爆炸物检测领域的热点技术之一.量子点作为一种新型荧光探针材料,具有高的量子产率和稳定性,能够灵敏响应硝基芳香爆炸物,在微痕量硝基芳香爆炸物的快速筛查中受到了重点关注.本文综述了量子点探针用于微痕量硝基芳香爆炸物检测中的实际应用和研究进展,强调了光诱导电子转移(PET)式和荧光共振能量转移(FRET)式量子点探针的响应机理,总结了PET式和FRET式量子点探针的特点,并对它们的发展趋势和降解爆炸物的应用进行了展望.  相似文献

Bacterial strains with poly-3-hydroxybutyrate (PHB) degradation potential were isolated from waste yard soil samples of selected industrial sites in Uttarakhand, India, and two microbial consortia were developed, i.e. Consortium I comprises Pseudomonas sp. strain Rb10, Pseudomonas sp. strain Rb11 and Bacillus sp. strain Rb18, and Consortium II is composed of Lysinibacillus sp. strain Rb1, Pseudomonas sp. strain Rb13 and Pseudomonas sp. strain Rb19. The current study involved enrichment selection via liquid and semi-solid media, followed by isolation and screening of bacterial strains using PHB pellets and films. Furthermore, the identification and characterisation was done by triphasic approach. The utilisation of PHB by the characterised strains was confirmed by Fourier transform infrared spectroscopy and scanning electron microscopy. Moreover, the minimum inhibitory concentration of solubilised PHB was found to be 2.5?mg/mL, which was detected through ‘clear zone assay’. Further, the selection and biocompatibility testing of potential isolates were performed for the formation of bacterial consortia. Thus, the present work would provide direct and standardised protocol for screening and selection of potential microbiomes for biodegradation of polymers by overcoming the negative effect of organic solvents. Moreover, indigenously developed consortia would be evaluated for their in situ biodegradation potential against various bioplastic films.  相似文献

• The autotrophic nitrogen removal combining Feammox and Anammox was achieved. • Activated carbon can be used as an electron shuttle to enhance Feammox activity. • Fe(III) was reduced to Fe(II) and the secondary Fe(II) mineral (FeOOH) was obtained. • The iron-reducing bacteria and Anammox consortium was enriched simultaneously. Ferric iron reduction coupled with anaerobic ammonium oxidation (Feammox) is a novel ferric-dependent autotrophic process for biological nitrogen removal (BNR) that has attracted increasing attention due to its low organic carbon requirement. However, extracellular electron transfer limits the nitrogen transformation rate. In this study, activated carbon (AC) was used as an electron shuttle and added into an integrated autotrophic BNR system consisting of Feammox and anammox processes. The nitrogen removal performance, nitrogen transformation pathways and microbial communities were investigated during 194 days of operation. During the stable operational period (days 126–194), the total nitrogen (TN) removal efficiency reached 82.9%±6.8% with a nitrogen removal rate of 0.46±0.04 kg-TN/m3/d. The contributions of the Feammox, anammox and heterotrophic denitrification pathways to TN loss accounted for 7.5%, 89.5% and 3.0%, respectively. Batch experiments showed that AC was more effective in accelerating the Feammox rate than the anammox rate. X-ray photoelectron spectroscopy (XPS) analyses showed the presence of ferric iron (Fe(III)) and ferrous iron (Fe(II)) in secondary minerals. X-ray diffraction (XRD) patterns indicated that secondary iron species were formed on the surface of iron-AC carrier (Fe/AC), and Fe(III) was primarily reduced by ammonium in the Feammox process. The phyla Anaerolineaceae (0.542%) and Candidatus Magasanikbacteria (0.147%) might contribute to the Feammox process, and Candidatus Jettenia (2.10%) and Candidatus Brocadia (1.18%) were the dominative anammox phyla in the bioreactor. Overall, the addition of AC provided an effective way to enhance the autotrophic BNR process by integrating Feammox and anammox.  相似文献

Hypnum cupressiforme epiphytic moss and tree bark (elm and holm oak) samples have been collected in three sites of the city of Florence in the period 1995-1998. Lead, zinc, copper, and cadmium were determined by differential pulse anodic stripping voltammetry (DPASV) in about 200 samples collected at different heights above ground. A new clean-up procedure by nitrogen jet has been followed and its efficiency has been also verified by electron microscopy (SEM and ESEM techniques). Lead median contents in moss and bark samples fall within the ranges of 0.052-0.86 and 0.20-1.30 μmoles g-1 (dry weight), resulting values for moss and bark are proportional to the vehicular traffic density. the increasing use of lead-free gasoline has not been followed by a decrease of lead moss concentration.  相似文献

通过沙土柱实验考察了硫包衣尿素和甲醛尿素2种缓释肥料在不同pH值和盐度的海水介质中养分的释放特性,并利用摇瓶培养实验考察了这两种肥料对SZ36-1原油生物降解作用的影响.结果表明,在实验设置的pH(7.00—8.00)和盐度(24.00—32.00)范围内,pH值对硫包衣尿素和甲醛尿素肥料氮素释放性能有较大影响,其中,近中性的海水介质有利于硫包衣尿素中氮素的释放,弱碱性环境更有利于甲醛尿素的释放;而盐度对两种肥料的氮素释放均无明显影响.两种缓释肥料都能够持续释放氮素而维持石油烃降解菌QPH-2的生长,并促进原油中各组分包括烷烃和芳烃明显降解.其中,添加硫包衣尿素体系中各烷烃组分的降解率为13.94%—59.02%,芳烃类组分降解率为9.60%—32.33%;添加甲醛尿素体系中各烷烃组分的降解率为11.56%—49.65%,芳烃类组分降解率为13.49%—41.67%.  相似文献

环境内分泌干扰物(endocrine disrupting chemicals,EDCs)可模仿或拮抗天然激素与核受体结合,干扰核受体的同源或异源二聚,进而通过共调节因子的招募调控转录活性,最终引起内分泌干扰效应。目前研究主要针对EDCs与核受体的结合过程,忽视了其对核受体二聚化过程的影响,而该过程的阻断可直接导致转录失活。EDCs对于不同核受体二聚化的影响不同,只有激动剂EDCs能够促进雄激素受体(androgen receptor,AR)的同源二聚化,而雌激素受体(estrogen receptor,ER)在与具有激动或拮抗活性的EDCs结合后都可诱导ER二聚体的形成,但二聚化类型不同。通过检索ToxCast和Tox21数据库发现多达227种EDCs可以诱导ER二聚化,相比于ERα-ERα同源二聚体(6.09%~7.38%的活性率),EDCs更易诱导ERα-ERβ异源二聚体(11.25%~12.22%的活性率)和ERβ-ERβ同源二聚体(10.02%~11.69%的活性率)。EDCs也能够差异性诱导其他核受体如维生素D受体(vitamin D receptor,VDR)与维甲酸X受体(retinoid X receptor,RXR)形成的异源二聚体,不同类型的二聚体对于研究EDCs转录活性的生理学相关性具有重要意义。基于经济合作与发展组织(Organization for Economic Co-operation and Development,OECD)报告的参考化学品研究发现,相比于配受体结合活性,二聚活性与转录活性之间有着更好的相关关系。本文从EDCs介导的核受体二聚化转录机制、二聚化与转录活性间的关系以及二聚化研究方法三方面,总结EDCs对核受体二聚化的影响,以期为深入理解EDCs的分子作用机制,推进化合物的内分泌干扰风险评估提供参考。  相似文献

建立了一种能够同时测定饮用水中9种卤乙酸的气相色谱方法.研究了前处理过程中不同实验条件对卤乙酸回收率的影响,通过简化前处理方法,优化色谱操作条件,提高了卤乙酸的回收率,降低了方法检测限,9种目标化合物的线性相关度均在0.99以上,回收率保持在88%—109%之间,能够满足美国环保局和中国生活饮用水卫生标准水质监测分析的要求.  相似文献

Effluents from wastewater treatment plant (WWTP) have been reported to have a broad spectrum of endocrine disrupting compounds (EDCs). The majority of studies have focused on the occurrence of estrogenic activity, while ignoring nuclear hormone receptors (NRs) pathways. In the present study, a battery of in vitro yeast bioassays and a cell bioassay, including antagonistic and agonistic effects on estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PR), estrogen- related receptor (ERR) and aryl hydrocarbon receptor (AHR), were conducted to evaluate the removal efficien- cies of EDCs by different treatment processes of a WWTP located in Beijing. Estrogenic, anti-estrogenic, anti- androgenic, anti-progesteronic, anti-ERR and the activa- tion of AHR activities were detected in samples from all treatment processes and the receiving water. The concen- tration of estrogenic contaminants with estradiol (E2) equivalent concentrations ranged from 0.82 x 10-9 to 3.54 x 10 9g Ee_EQ.L-1. The concentration of anti-estrogenic contaminants with 4-hydroxytamoxifen (4-OHT) equiva- lent concentrations ranged from 1.24 × 10-6 to 2.36 x 10-6 g 4-OHT-EQ.L-1. The concentration of anti-androgenic contaminants ranged from 2.21 x 10-s to 3.52 × 10-6g flutamide-EQ. L-1. The concentration of anti-progesteronic contaminants ranged from 3.15 x 10^-5 to 2.71 x 10^-4g RU486-EQ. L-1. The concentration of anti-ERR contami- nants ranged from 7.09 x 10-5 to 6.50 x 104 g 4-OHT-EQ × L^-10. The concentration of AHR activators ranged from 1.7 × 10-10 to 3.4 × 10^-10g TCDD-EQ-L-1. These processes including secondary clarifier, coagulation, as well as coal and sand filtration could eliminated 67.2% of estrogenic contaminants, 47.0% of anti-estrogenic contaminants, 98.3% of anti-androgenic contaminants, 88.4% of anti- progesteronic contaminants, 65.4% of anti-ERR contami- nants and 46.9% of AHR activators. WWTP effluents contain multiple receptor disruptors may have very complex adverse effects on exposed organisms.  相似文献

黄颡鱼黑色素细胞原代培养及迁移相关基因克隆分析   总被引:1,自引:0,他引:1  
黑素皮质素受体-1(MC1R)在鱼体中对黑色素细胞的分化及迁移起主要的调控作用,而且在神经系统和免疫系统中均表现出重要的生理功能。本研究对黄颡鱼表皮黑色素细胞的培养条件进行探索,并在此基础上对MC1R基因进行克隆,为后续研究黄颡鱼黑色素异常的细胞及分子机理研究提供基础。结果表明,黄颡鱼表皮黑色素细胞在27℃培养条件下,72 h开始贴壁生长,培养基中添加20%小牛血清培养效果比添加10%小牛血清好。此外,我们对黄颡鱼MC1R基因进行克隆。MC1R基因克隆方面,黄颡鱼MC1R基因全长为936 bp,编码312个氨基酸,与剑尾鱼(Xiphophorus maculates)、孔雀鱼(Poecilia reticulata)、条斑星鲽(Verasper moseri)、海鲈(Dicentrarchus labrax)、大菱鲆(Psetta maxima)等5种鱼类MC1R基因的同源性分别为97%、96%、90%、90%、90%,系统分析结果显示,黄颡鱼MC1R基因在进化树上的位置与黄颡鱼的分类所处位置基本吻合。黄颡鱼表皮黑色素细胞原代培养的条件为:27℃,添加20%小牛血清,所克隆基因为黄颡鱼MC1R基因。本研究为后续研究黄颡鱼黑色素异常的细胞及分子机理研究提供基础。  相似文献
