研究了滤纸对青蒿丛生芽诱导及遗传转化的影响.结果表明,培养基上加铺一层滤纸能够明显促进青蒿丛生芽的诱导,丛生芽诱导率可达到97%.在高效丛生芽诱导体系的基础上,探讨了滤纸在根癌农杆菌介导的青蒿遗传转化中的应用.优化后的青蒿转化体系,抗性丛生芽诱导率可达到59.7%,其中,12.5%的抗性丛生芽能发育成抗性生根植株,生根植株PCR检测均为阳性.在部分PCR检测阳性植株中检测到了GUS的稳定表达.该优化的转化体系对青蒿素生物合成分子调控的研究具有重要意义.图5表1参26  相似文献

选择采样期不同降雨天数的碱片,测定余边的硫酸盐化速率含量,其结果为:采样期全月无雨时,余边上的硫酸盐均未检出;半月以上至全月有雨时检出率为100%,检出量占样品量的2.70~15.95%。经扩散试验,查清了余边上的硫酸盐主要是有效面积上采集的样品的扩散,并提出了相应的修正方法。  相似文献

Abstract: Lakes are important water resources on the North Slope of Alaska. Freshwater is required for oilfield production as well as exploration, which occurs largely on ice roads and pads. Since most North Slope lakes are shallow, the quantity and quality of the water under ice at the end of winter are important environmental management issues. Currently, water‐use permits are a function of the presence of overwintering fish populations, and their sensitivity to low oxygen concentrations. Sampling of five North Slope lakes during the winter of 2004‐2005 shed some light on the winter chemistry of four lakes that were used as water supplies and one undisturbed lake. Field analysis was conducted for oxygen, conductivity, pH, and temperature throughout the lake depth, as well as ice thickness and water depth. Water samples were retrieved from the lakes and analyzed for Na, Ca, K, Mg, Fe, dissolved‐organic carbon, and alkalinity in the laboratory. Lake properties, rather than pumping, were the best predictors of oxygen depletion, with the highest dissolved‐oxygen levels maintained in the lake with the lowest concentration of constituents. Volume weighted mean dissolved‐oxygen concentrations ranged from 4 to 94% of saturation in March. Dissolved oxygen and specific conductance data suggested that the lakes began to refresh in May.  相似文献

The significance of technical data, as well as the significance of system boundary choices, when modelling the environmental impact from recycling and incineration of waste paper has been studied by a life cycle assessment focusing on global warming potentials. The consequence of choosing a specific set of data for the reprocessing technology, the virgin paper manufacturing technology and the incineration technology, as well as the importance of the recycling rate was studied. Furthermore, the system was expanded to include forestry and to include fossil fuel energy substitution from saved biomass, in order to study the importance of the system boundary choices. For recycling, the choice of virgin paper manufacturing data is most important, but the results show that also the impacts from the reprocessing technologies fluctuate greatly. For the overall results the choice of the technology data is of importance when comparing recycling including virgin paper substitution with incineration including energy substitution. Combining an environmentally high or low performing recycling technology with an environmentally high or low performing incineration technology can give quite different results. The modelling showed that recycling of paper, from a life cycle point of view, is environmentally equal or better than incineration with energy recovery only when the recycling technology is at a high environmental performance level. However, the modelling also showed that expanding the system to include substitution of fossil fuel energy by production of energy from the saved biomass associated with recycling will give a completely different result. In this case recycling is always more beneficial than incineration, thus increased recycling is desirable. Expanding the system to include forestry was shown to have a minor effect on the results. As assessments are often performed with a set choice of data and a set recycling rate, it is questionable how useful the results from this kind of LCA are for a policy maker. The high significance of the system boundary choices stresses the importance of scientific discussion on how to best address system analysis of recycling, for paper and other recyclable materials.  相似文献

Wastewater management in small and medium-sized enterprises representing the chemical and food industries was investigated. The results showed that wastewater discharged from an ink-production factory was highly contaminated with organic pollutants. Anaerobic biological treatment followed by chemical coagulation using ferric chloride aided with lime proved to be very effective and produced an effluent that complied with national regulatory standard for wastewater discharge into public sewage network. Also, management of wastewater from a confectionery factory representing the food sector was carried out. Aerobic biological treatment using plastic-packed trickling filter proved to be an effective treatment method. However, application of in-plant control measures alleviated the requirement for the construction of a wastewater treatment plant. The applied pollution prevention and cleaner production measures involved good housekeeping, recovery of spent chocolate, modification of floor cleaning and installation of suction devices for the removal of sugar and starch powders. All improvement measures were documented by cost/benefit analysis.  相似文献

Background, aim, and scope  The paper describes the spatial contamination of the River Kymijoki, South-Eastern Finland, and the coastal region of the Gulf of Finland with PCDD/Fs and mercury. The findings of ecotoxicologial and human health studies are also reported, including environmental and human risk assessments. Sediments from the River Kymijoki, draining into the Gulf of Finland, have been heavily polluted by the pulp and paper industry and by chemical industries. A wood preservative, known as Ky-5, was manufactured in the upper reaches of the river between 1940 and 1984 causing severe pollution of river sediments with polychlorinated dibenzo-p-dioxins (PCDD) and dibenzofurans (PCDF). Moreover, the sediments have been polluted with mercury (Hg) from chlor-alkali production and the use of Hg as a slimicide in pulp and paper manufacturing. Materials and methods  An extensive sediment survey was conducted as well as sediment transport modeling, toxicity screening of sediment invertebrates, and a survey of contaminant bioaccumulation in invertebrates and fish. Studies on human exposure to PCDD/Fs and the possible effects on hypermineralization of teeth as well as an epidemiological study to reveal increased cancer risk were also conducted. An assessment of the ecological and human health risks with a null hypothesis (no remediation) was undertaken. Results  The sediment survey revealed severe contamination of river and coastal sediments with PCDD/Fs and Hg. The total volume of contaminated sediments was estimated to reach 5 × 106 m3 and hot spots with extremely high concentrations (max 292,000 ng g−1 or 1,060 ng I-TEQ g−1 d.w.) were located immediately downstream from the pollution source (approximately 90,000 m3). Sediment contamination was accompanied by changes in benthic assemblages, but direct effects were masked by many factors. The fish showed only slightly elevated PCDD/F levels in muscle, but orders of magnitude higher in the liver compared with reference freshwater sites and the Baltic Sea. The concentrations in human fat did not reveal high human exposure in the Kymijoki area in general and was lower than in sea fishermen. The relative risk for total cancer among farmers was marginally higher (RR = 1.13) among those living close to the river, compared with farmers living further away, and the possibility of increased cancer risk cannot be ruled out. A conservative risk assessment revealed that the present probability of exceeding the WHO upper exposure limit of 4 pg WHO-TEQ kg−1 d−1 for PCDD/Fs and DL-PCBs was 6%. The probability of exceeding the WHO limit value of 0.23 μg kg−1 d−1 for methyl mercury was estimated to be notably higher at 62%. Based on these studies and the estimated risks connected with different remediation techniques a general remediation plan with cost benefit analysis was generated for several sub-regions in the river. Dredging, on-site treatment, and a close disposal of the most contaminated sediments (90,000 m3) was suggested as the first phase of the remediation. The decision regarding the start of remediation will be made during autumn 2008. Conclusions  The sediments in the River Kymijoki are heavily polluted with PCDD/Fs and mercury from earlier chlorophenol, chlor-alkali, and pulp and paper manufacturing. A continuous transport of contaminants is taking place to the Gulf of Finland in the Baltic Sea. The highly increased PCDD/F and Hg levels in river sediments pose an ecotoxicological risk to benthic fauna, to fish-eating predators and probably to human health. The risks posed by mercury exceed those from PCDD/Fs and need to be evaluated for (former) chlor-alkali sites and other mercury releasing industries as one basis for remediation decision making. Recommendations and perspectives  The studies form the basis of a risk management strategy and a plan for possible remediation of contaminated sediments currently under consideration in the Southeast Finland Regional Environment Centre. It is recommended that a detailed restoration plan for the most seriously contaminated areas should be undertaken. Based on current knowledge, the restoration of the whole river is not feasible, considering the current risk caused by the contaminated sediment in the river and the costs of an extensive restoration project. The experiences gained in the present case should be utilized in the evaluation of PCDD/F- and mercury-contaminated sites in other countries. The case demonstrates that the historic reservoirs are of contemporary relevance and should be addressed, e.g., in the national implementation plans of the Stockholm Convention.
